RNA Polymerase 1 inhibitors against African trypanosomes in vitro and in mice.

Therapy of human African trypanosomiasis, fatal if not treated, has been greatly improved by orally available and safer fexinidazole, but reliable cure of late-stage central nervous system (CNS) infection remains a challenge. African trypanosomes are unique among eukaryotes in having an RNA Polymerase 1 that transcribes not only rRNA but also mRNA that encodes the abundant variable surface glycoprotein (VSG) that coats the cell membrane of bloodstream-form parasites and allows them to evade host immune defenses. RNA Pol 1 inhibitor BMH-21, an intercalator not associated with DNA damage and a l

Germline-targeted baboon apolipoprotein L-1 protects mice against African trypanosomes.

Some primates are immune to infection by most African trypanosome parasites due to apolipoprotein L-1 (APOL1), a primate-specific ion channel-forming protein. Our long-term objective has been to reduce African trypanosomiasis in livestock by genetic bioengineering of cattle with primate APOL1. To select which primate APOL1, we analyzed Papio ssp. APOL1 proteins and found that Papio hamadryas APOL1 was a strong candidate for transgenic animal production based on its trypanosome-killing capacity, ion channel properties, and stability. We generated seven transgenic murine lines based on the P. ha

Trypanosoma vivax in Water Buffaloes (Bubalus bubalis): A Host-Centered Synthesis of Pathogenesis, Epidemiology, Diagnosis, and Integrated Control with Implications for Tropical Production Systems.

Trypanosoma vivax is a hemoprotozoan parasite of major veterinary importance affecting domestic ungulates in Africa and the Americas. While traditionally addressed within cattle-centered paradigms, accumulating evidence indicates that water buffaloes (Bubalus bubalis) are both clinically susceptible and epidemiologically significant hosts. This structured narrative review provides a host-centered synthesis of global evidence on T. vivax infection in buffaloes, integrating pathogenesis, transmission biology, epidemiology, diagnostics, chemotherapy, and integrated control. The analysis encompass

A strategic discovery roadmap towards high-quality leads and drug development candidates for kinetoplastid diseases. Part 2: from molecule to confirmed hit.

Given the medical importance and challenges related to kinetoplastid diseases, a strategic roadmap is needed for the identification of high-quality leads and drug development candidates. Within the aim to deliver more compelling proof-of-concept read-outs, this part proposes a systematic flow-chart of laboratory experiments and decision criteria, focusing on African trypanosomiasis, Chagas disease and visceral and cutaneous leishmaniasis. Next to precision experimental design and reporting, an overview is provided of various complementary laboratory models reproducing kinetoplastid infection a

Targeting Kinetoplastid Parasites with ProTide Prodrugs: A Proof-of-Concept Study.

Neglected tropical diseases (NTDs) remain a major global health challenge, particularly in low- and middle-income countries. Kinetoplastid parasites causing Chagas disease, leishmaniasis, and African trypanosomiasis rely on host purine salvage pathways, making nucleoside analogues attractive therapeutic candidates. However, their clinical utility is limited by poor cellular uptake and rapid metabolism. Herein, we report the application of the ProTide prodrug technology, a clinically validated approach that enhances the intracellular delivery of nucleoside monophosphates for the treatment of ki

Barriers to integration of passive screening for sleeping sickness in Bibanga Health District, Democratic Republic of the Congo.

INTRODUCTION: The control of human African trypanosomiasis (HAT) relies primarily on the early detection and appropriate treatment of cases, which remain central components of current elimination strategies, while no vaccine or chemoprophylaxis is available. However, under-screening by mobile units is observed. Low HAT prevalence may lower motivation to participate in active screening programs that are important for early detection of cases, because people do not perceive the risk of infection. In this context, the integration of passive HAT screening into primary health services is encouraged

A national atlas of the distribution of tsetse flies in Côte d'Ivoire.

African animal trypanosomosis (AAT) is one of the major diseases of livestock in Côte d'Ivoire. However, the lack of comprehensive, nationally aggregated data hinders the rational planning of control measures and the estimation of their impacts on disease transmission. To address this shortcoming, the Institut Pierre Richet (IPR, Ministry of Health) and the Directorate of Veterinary Services (Ministry of Livestock) embarked on the development of a national tsetse flies and AAT in Côte d'Ivoire. The atlas is a dynamic, georeferenced database on the distribution of disease and its vectors, and t

Risk of transmission of sleeping sickness in the four historical epicentre villages of the Bipindi focus, Cameroon.

BACKGROUND: Bipindi is a historic focus of human African trypanosomiasis (HAT). With the number of cases declining to near-elimination levels, understanding the current entomological risk is crucial to guiding elimination strategies. METHODS: Two entomological surveys in four historical sentinel villages used pyramidal traps to capture tsetse flies. Dissected midguts were analyzed by polymerase chain reaction (PCR) for trypanosome infection. Blood meal origins were identified using cytochrome B gene heteroduplex analysis. RESULTS: Glossina palpalis palpalis was accounted for 99.0% of 708 cap

IL-4Rα signalling and Iloprost treatment modulate neuroimmune and neurocognitive responses in Trypanosoma brucei-Infected mice.

Despite the long-standing recognition of the impact of trypanosomiasis on brain functions, efforts to link the associated disease pathology to neuroinflammation and cognitive deficits have been largely unsuccessful, with trypanocidal drugs often proven ineffective. In this study, using novel object recognition task for recognition memory, we compared interleukin-4 receptor alpha-inhibited mice with trypanosome infection. Furthermore, we treated with Iloprost, a prostacyclin analogue, and the standard drug Diminazene aceturate subsequently for changes in transcriptional expressions of IL-4 and

Suramin Interactions Across Biological Systems: From Molecular Targets to Therapeutic Implications.

Suramin is a century-old polysulfonated naphthylurea that remains a first-line treatment for early-stage human African trypanosomiasis (HAT). Remarkably, despite its age, suramin continues to draw attention because of its unusually broad spectrum of biological activities. Historically known as an antagonist of purinergic (P2) receptors and an inhibitor of extracellular enzymes, suramin has more recently been shown to interact with a range of intracellular and mitochondrial proteins. These include succinate dehydrogenase, the ADP/ATP carrier (AAC), the aspartate/glutamate carriers AGC1 and AGC2

Global, regional, and national disease burden of arthropod-borne diseases: Projections to 2030 based on the global burden of disease study 2021.

BACKGROUND: Arthropod-borne diseases (ABDs) represent an ongoing threat to global public health, affecting millions annually with viral, bacterial and parasitic infections worldwide. METHODS: Data on prevalence and disability-adjusted life years (DALYs) for ABDs were obtained from the Global Burden of Disease (GBD) 2021 study. Based on DALYs, the nine ABDs were categorized into protozoan, helminthic, and viral diseases. Temporal trends were quantified using the average annual percentage change (AAPC) in age-standardized prevalence rates (ASPRs) and age-standardized DALY rates (ASDRs). Frontie

A strategic discovery roadmap towards high-quality leads and drug development candidates for kinetoplastid diseases. Part 1: setting the stage.

Given the impact of kinetoplastid diseases, the limited therapeutic options and risk of treatment failure, continued research efforts to discover novel drug entities are required. The ambition to deliver drug development candidates has mainly been taken on board by academia and public private partnerships, but remains highly challenging because of the lack of adequate funding and standardized laboratory procedures. Establishing a systematic roadmap of experiments and decision criteria to attain high-quality leads and drug candidates with lower risk profiles remains the logical path to deliver

Trypanosoma brucei TEL2 inhibits VSG switching and protects PIKKs from the 26S proteasome-mediated degradation.

Trypanosoma brucei causes debilitating human African trypanosomiasis. It undergoes antigenic variation and sequentially expresses distinct VSGs, its major surface antigen, thereby effectively evading the host's immune response. We have shown that many telomere proteins suppress VSG switching, as VSGs are expressed exclusively from subtelomeric loci. We have now identified the T. brucei TEL2 homolog based on its sequence and predicted structure similarities to those of known TEL2 homologs. TEL2 homologs have a conserved function of maintaining the protein stability of PI3K-related protein kinas

Chalcones and flavonoids: New perspectives in the modulation of Trypanosoma brucei metacaspase-2.

African Human Trypanosomiasis (AHT), caused by Trypanosoma protozoa, faces therapeutic challenges due to limited efficacy and adverse effects. Natural compounds, such as chalcones and flavonoids, represent promising alternatives. This study evaluated nine compounds (six chalcone derivatives and three flavonoids) as modulators of Trypanosoma brucei metacaspase-2. Screening at 1 and 6 μM revealed inhibitory activity, with compounds EM-1A, EM-1B, EM-1C, and EM-F12C showing notable IC50 values. Mechanistic studies identified competitive inhibition for EM-1A, EM-1C, and EM-F12C, while EM-1B exhibit

Identification of ZINC database-derived antitrypanosomal candidates targeting Trypanosoma congolense lysosomal-α-mannosidase using computational, biochemical and in vivo methods.

Animal African Trypanosomiasis (AAT), primarily caused by Trypanosoma congolense, continues to threaten livestock productivity across sub-Saharan Africa. The persistence of the disease is largely driven by antigenic variation of Variant Surface Glycoproteins (VSGs), which enable immune evasion. Lysosomal α-D-mannosidase (LDM) suggestively plays a key role in VSG catabolism, and its inhibition may impair parasite survival. This study employed computational and biochemical approaches to characterize T. congolense lysosomal α-D-mannosidase (TcLDM) and identify potential inhibitory compounds. A hi

Xeno-monitoring the impact of Vector Control on trypanosome transmission in the Forecariah sleeping sickness focus (Guinea).

BACKGROUND: Forecariah is one of the most active foci of gambiense human African trypanosomiasis (gHAT) in Guinea. The study aimed to evaluate the impact of a vector control intervention carried out in this focus from 2018 to 2021. To achieve this, thousands of deltamethrin-impregnated blue tiny targets were deployed annually. METHODOLOGY AND PRINCIPAL FINDINGS: Thirty-one sentinel traps were used to assess the following entomological and parasitological parameters: (i) the apparent density (AD) of tsetse flies, monitored twice per year; (ii) the tsetse fly infection rates, evaluated once per

Differential Virulence and Pathogenicity of Canine Drug-Resistant Trypanosoma brucei brucei Field Isolates in Experimental Murine Infection.

PURPOSE: African animal trypanosomosis remains a significant veterinary and economic threat in sub-Saharan Africa, worsened by rising multidrug resistance. In dogs, Trypanosoma brucei brucei infections are often acute and fatal, yet the relationship between drug resistance and pathogenicity remains poorly defined. This study aimed to compare the virulence and pathogenicity of three canine multidrug-resistant T. b. brucei field isolates from Southeast Nigeria using an experimental murine model. METHODS: Twenty Sprague Dawley rats were randomly assigned to four groups (n = 5). Groups I-III rece

Transmission and cost-effectiveness modelling to estimate the progress towards elimination and future strategy optimisation for gambiense human African trypanosomiasis in Uganda.

BACKGROUND: Gambiense human African trypanosomiasis (gHAT) is a vector-borne disease with hundreds of thousands of people living at risk across Sub-Saharan Africa. Uganda, having reduced gHAT cases reported annually from 948 in 2000 to zero local cases since 2020, is a frontrunner to be verified by the World Health Organization (WHO) as having achieved elimination of transmission (EoT) of gHAT. It is now crucial to quantify the impact of the interventions deployed, quantify whether the last transmission event (LTE) and last case have already occurred, and determine the resources required to su
