Antiprotozoal Aminosteroids from Pachysandra terminalis.

Trypanosoma brucei rhodesiense (Tbr) and Plasmodium falciparum (Pf) are protozoan parasites that cause severe diseases, namely, Human African Trypanosomiasis (HAT) and Malaria. Due to limited treatment options, there is an urgent need for new antiprotozoal drugs. Pachysandra terminalis (P. terminalis), a plant belonging to the family Buxaceae, is known as a rich source of aminosteroid alkaloids, and a previous study of our working group already showed that the alkaloid-enriched fraction of P. terminalis aerial parts showed promising activity against protozoan parasites. In the present study, t

Human African trypanosomiasis.

Human African trypanosomiasis or sleeping sickness is caused by infection with Trypanosoma brucei gambiense or Trypanosoma brucei rhodesiense parasites, which are transmitted by tsetse flies in sub-Saharan Africa. Control of human African trypanosomiasis is based on case detection, treatment, and vector control. In the past decade, simple rapid diagnostic tests were introduced for gambiense human African trypanosomiasis, facilitating screening in primary health-care facilities. A new oral drug, fexinidazole, became the first-line treatment for gambiense human African trypanosomiasis without se

Evaluation of two candidate molecules-TCTP and cecropin-on the establishment of Trypanosoma brucei gambiense into the gut of Glossina palpalis gambiensis.

Trypanosomiasis, transmitted by tsetse flies (Glossina spp.), poses a significant health threat in 36 sub-Saharan African countries. Current control methods targeting tsetse flies, while effective, allow reinfestation. This study investigates paratransgenesis, a novel strategy to engineer symbiotic bacteria in tsetse flies, Sodalis glossinidius, to deliver anti-trypanosome compounds. Disrupting the trypanosome life cycle within the fly and reducing parasite transmission could offer a sustainable solution for trypanosomiasis control. In this context, we tested the effect of cecropin, reported t

Cas10 based 7SL-sRNA diagnostic for the detection of active trypanosomosis.

Animal Trypanosomosis (AT) is a significant disease affecting cattle across sub-Saharan Africa, Latin America, and Asia, posing a major threat to economic productivity and animal welfare. The absence of reliable diagnostic tests has led to an over-reliance on widespread pre-emptive drug treatments, which not only compromise animal health but also heighten the risk of drug resistance. The chronic nature of AT, characterized by cyclical low or undetectable parasite levels, and the necessity for field-applicable tests that can distinguish between active infection and prior exposure, present consi

Anti-Trypanosomatid and Antiplasmodial Activities of Alkaloids From Hippeastrum Species.

Diseases caused by trypanosomatid parasites like human African trypanosomiasis (HAT), Chagas disease (CD), leishmaniasis, and malaria are persistent health problems in developing countries that still demand new drug development. The species of the Amaryllidoideae subfamily (Amaryllidaceae) represent a vast source of alkaloids with a wide range of bioactive properties, including antiparasitic effects. The aim of this study was to evaluate the antiparasitic activity of the alkaloids hamayne, 7-hydroxyclivonine, 4-O-methylnangustine, and candimine against Trypanosoma brucei rhodesiense, Trypanoso

Production of Protease Inhibitor With Penicillium sp. - Optimization of the Medium for Growth in Pellet Form and Cytotoxicity Testing.

Penicillium sp. (IBWF 040-09) produces a protease inhibitor that can potentially be used against the main protease of human African trypanosomiasis. Since the target substance is formed intracellularly (under nutrient limitation), the fungal pellet is preferred compared to the free mycelia in bioreactor cultivation. The optimization of the production of protease inhibitor became the main focus of this study. The effects of the concentrations of spores, calcium chloride, and Pluronic F68 were investigated with regard to fungal growth, pellet morphology, and the production of protease inhibitor.

The impact of canine African trypanosomiasis in anti-poaching dogs in the Republic of Zambia.

Zambia introduced canine units in 2014 as an anti-poaching tool. Working in challenging environments, these dogs face several health threats, one being the protozoal, vector-borne canine African trypanosomiasis (CAT). This report aims to investigate the incidence, management and impact of CAT on Zambia's canine units. Case data during 2019-2023 were collated to investigate incidence and clinical trends. A questionnaire examining mitigation methods and perceived impact of the disease was distributed. Participants, conservation organisations with canine units, were identified using non-probabili

Overproduction and Characterization of Recombinant Soluble Trypanosoma brucei Phospholipase A2.

Trypanosoma brucei phospholipase A2 (TbPLA2) is a validated drug target but the difficulty in expressing its soluble recombinant protein has limited its exploitation for drug and vaccine development for African and American trypanosomiases. We utilized recombinant deoxyribonucleic acid (DNA) technology approaches to express soluble TbPLA2 in Escherichia coli and Pichia pastoris and biochemically characterize the purified enzyme. Full-length TbPLA2 was insoluble and deposited as inclusion bodies when expressed in E. coli. However, soluble and active forms were obtained when both the full-length

The extracellular region of Trypanosoma congolense Membrane Bound Acid Phosphatase induces partial protection in immunized BALB/c mice.

African trypanosomiasis is a parasitic disease caused by African trypanosomes and there is no vaccine because the trypanosomes possess an intricate machinery for antigenic variation. Owing to the nonsusceptibility of trypanosomes' Membrane-Bound Acid Phosphatase (MBAP; EC 3.1.3.2) to antigenic variation and its central role in molecular trafficking, we hypothesized that MBAP is a promising target for vaccine development. Herein, a DNA vaccine was constructed using recombinant DNA technology to insert the gene of the extracellular portion (EP) of Trypanosoma congolense MBAP without its signal p

The STROGHAT study protocol: An intervention study to evaluate safety, effectiveness and feasibility of treating gambiense HAT seropositive subjects with acoziborole.

BACKGROUND: Coordinated efforts in the control of gambiense human African trypanosomiasis (gHAT) have significantly reduced its endemicity. WHO targets interruption of transmission by 2030. However, challenges remain, including low sensitivity of the current parasitological confirmation tests, leaving a potential human reservoir untreated. Acoziborole, a single-dose oral treatment, effective in both disease stages with a good safety profile, offers the potential of treatment of parasitologically negative gHAT seropositive subjects, which could improve diagnostic sensitivity. The STROGHAT study

Molecular and morphological characterization of one known and three new species of fish parasitic Trypanosoma Gruby, 1972 from the south coast of South Africa.

The genus Trypanosoma Gruby, 1843 encompasses unique, flagellate haemoparasites infecting all vertebrate classes globally (excluding Antarctica). While trypanosomes in terrestrial mammals are well-studied due to their medical and veterinary significance, those in fishes remain largely unexplored, with limited data on their life cycles and ecological roles. Furthermore, the phylogenetic relationships of numerous aquatic species are unresolved. This gap is notable in South Africa, a region with high marine fish biodiversity, yet only 2 documented marine trypanosome species, 1 in teleosts and 1 i

High-confidence glycosomal membrane protein inventory unveils trypanosomal peroxin PEX15.

Trypanosomatid parasite infections cause Chagas disease, human African trypanosomiasis, and leishmaniasis, affecting over 12 million people worldwide. Glycosomes, the peroxisome-related organelles of trypanosomes, are essential for survival, making their metabolic functions and biogenesis mediated by peroxins (PEXs) suitable drug targets. We report a comprehensive protein inventory of glycosomal membranes, defined through advanced subcellular membrane protein profiling combined with quantitative mass spectrometry and including 28 high-confidence glycosomal membrane proteins. We validate four p

Potential of entomopathogenic fungi for Glossina austeni control: insights into microbiome alterations and implications on sustainable management of the pest.

Glossina austeni Newstead (Diptera: Glossinidae) is a competent vector of the trypanosomes causing human African trypanosomiasis and the African animal trypanosomosis. Management of this pest has primarily involved trapping methods, Sterile Insect Technique, and research into vector competence-symbiotic interactions. Nevertheless, the use of entomopathogenic fungi (EPF) in integrated pest management programs for G. austeni control remains limited. Moreover, different tsetse fly species exhibit varying susceptibility to different EPF strains, indicating that no single strain is universally effe
