Clinical and molecular features of ovarian stimulation in peripubertal girls with mosaic Turner's syndrome.

STUDY QUESTION: Do peripubertal girls with mosaic Turner's syndrome (TS) respond to ovarian stimulation (OS) for oocyte freezing as adult women with normal ovarian reserve? SUMMARY ANSWER: Clinical and molecular reproductive/endocrine features of OS in these patients are similar to those of adult females. WHAT IS KNOWN ALREADY: OS for oocyte freezing is quite a new concept in peripubertal and young adolescent girls with TS because ovarian tissue cryopreservation (OTC) does not have proven efficacy, likely due to already diminished ovarian reserve and accelerated follicle atresia. No data are

Systems genetics approaches model the heritable architecture of polyendocrine metabolic ovarian syndrome.

Polyendocrine metabolic ovarian syndrome (PMOS), formerly known as polycystic ovary syndrome (PCOS), is the most common endocrine disorder in women and is closely associated with complex diseases such as cardiovascular disease and type 2 diabetes. However, the mechanistic links between PMOS and its comorbidities remain poorly understood. Here, we present an integrative systems genetics platform that leverages genetic diversity in both mice and humans to dissect the drivers of PMOS and its associated complications. This framework uncovered conserved genetic and environmental factors underlying

Karyotype-based evaluation of ovarian reserve before ovarian tissue cryopreservation in Turner syndrome patients.

BACKGROUND: Turner syndrome (TS) girls experience accelerated ovarian reserve depletion, exhibiting a strong demand for ovarian tissue cryopreservation (OTC) to preserve fertility and ovarian function. However, the current knowledge of OTC in TS is limited, and predicting follicular presence is insufficient. This study aims to investigate ovarian follicular reserve based on karyotypes, thereby contributing empirical data to informed decision-making in TS populations. METHODS: The retrospective case-control study comprised 26 TS patients and 52 age-matched non-TS (NTS) controls who underwent OT

Dipeptidyl aminopeptidase-like protein 6 regulates the INa-Ito balance influencing cardiac electrophysiology and arrhythmogenesis.

Dipeptidyl aminopeptidase-like protein 6 (DPP6) is a subunit of the Kv4 channels that carry the transient-outward current (Ito) in cardiac Purkinje cells (PCs) and ventricular myocytes (VMs). DPP6 genetic variants have been linked to severe arrhythmia syndromes. Given the influence of other Ito subunits on the Nav1.5-mediated cardiac sodium current (INa), we examined whether DPP6 regulates both Ito and INa. We explored the impact of the DPP6 missense variants c.821G>A and c.637C>T, segregating in families with long-QT syndrome (LQTS), and c.2252C>T and c.1578G>C, associated with J-wave syndrom

Preservation vs. Resection? Pediatric and Non-Pediatric Management Patterns in Ovarian Torsion.

BACKGROUND: Ovarian torsion (OT) is a rare but urgent surgical condition in children and adolescents. Evidence on how management differs between pediatric (PD) and non-pediatric (Non-PD) departments in Germany remains limited. METHODS: We conducted a retrospective cohort study using anonymized claims data from two major German statutory health insurance funds (2010-2019), covering 6.3 million insured individuals (≈1 million children). Patients ≤18 years with an inpatient diagnosis of OT (ICD-10-GM N83.5) were analyzed with respect to demographics, department type (PD vs. Non-PD), hospital typ

A Narrative Review of Recent Insights on Nerve Growth Factor Signaling in Physiological and Pathological Ovarian Processes in Mammals.

Nerve Growth Factor (NGF), a member of the neurotrophin family, is currently regarded as a key regulator of ovarian physiology beyond its well-known neurotrophic functions. The mammalian ovary is one of the most highly innervated peripheral organs. Increasing evidence indicates that NGF and its receptors, TrkA and p75NTR, are widely expressed in ovarian tissues. Through the activation of the PI3K/AKT, MAPK/ERK, and PLCγ signaling pathways, NGF influences granulosa cell proliferation, steroidogenesis, and ovulation. Physiological levels of NGF are essential for primordial follicle activation, F

Early-life exposure to ambient temperature and polycystic ovary syndrome: a nationwide cohort study.

BACKGROUND: Early-life temperature exposure, especially during oogenesis, may impact ovarian function later in life. However, the effect of ambient temperature during sensitive developmental windows on ovulation disorders remains unknown. METHODS: We investigate the associations of early-life ambient temperature exposure with incident polycystic ovary syndrome (PCOS) in the Growing Up Today Study II, a United States (US) nationwide prospective cohort. We included females born after 1989 with available outcome and exposure data during the preconception (N = 3265), gestation (N = 3265), and chi

Circadian Biology and Gynecological Diseases: Genetic Mechanisms and Potential Therapeutic Targets Underlying the Causal Link Between Insomnia and Ovarian Cysts.

BACKGROUND: Although circadian disruption has been implicated in gynecologic malignancies, genetic evidence for a causal relationship between insomnia and ovarian cysts remains lacking. The biological pathways linking sleep disturbance to ovarian pathology are therefore poorly understood, limiting mechanistic insights and therapeutic development. METHODS: We employed a two-sample Mendelian Randomization (MR) framework using large-scale Genome-Wide Association Study (GWAS) datasets of European ancestry to examine the effects of seven sleep-related traits on five gynecologic outcome types. Epigenetic safety of in vitro maturation in PCOS: genome-wide DNA methylation profiling of cord blood from a randomized controlled trial.

BACKGROUND: In vitro maturation (IVM) provides a safer alternative to conventional in vitro fertilization (IVF) for women with polycystic ovary syndrome (PCOS) by mitigating the risk of ovarian hyperstimulation. However, concerns persist regarding whether IVM perturbs epigenetic reprogramming in the offspring. Current evidence is constrained by candidate-gene approaches or a lack of parental controls. This study aimed to evaluate the genome-wide DNA methylation safety of IVM compared with conventional IVF using a rigorous trio-based design. METHODS: This secondary epigenetic analysis was nest

[Magnetic resonance imaging in the differential diagnosis of fetal abdominopelvic cysts: a retrospective case series].

INTRODUCTION: Abdominopelvic cystic lesions are relatively common findings during pregnancy. Ultrasound examinations performed during pregnancy can easily identify these cysts; however, accurate characterization is not always possible using ultrasound alone. The introduction of rapid magnetic resonance (MR) sequences allows more precise characterization of these lesions and also assists in therapeutic decision-making. OBJECTIVE: To identify fetal abdominopelvic cysts and assess their characteristics, ranging from the most common presentations to less frequently occurring cases. METHOD: In th

Genomic analyses implicate hormonal and metabolic dysregulation in polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) and its underlying features remain poorly understood. In this genetic study (n = 544,513), we expand the number of genetic loci from 16 to 29, and additionally identify 31 associated plasma proteins. Many risk-increasing loci were associated with later age at menopause, underscoring the reproductive longevity related to an increased oocyte number and/or availability across the lifespan. Hormonal regulation in the etiology of this condition, through metabolic and reproductive features, was emphasized. The proteomic analysis highlighted metabolic biology known to

Ecdysone receptor autonomously controls germ cell differentiation in the Drosophila ovary.

BACKGROUND: Brucellosis is a zoonotic disease that causes substantial public health problems and endangers the development of animal husbandry in endemic areas. Early diagnosis of infected animals and humans is a crucial step in reducing the incidence of brucellosis. In this study, we designed different combinations of Brucella major outer membrane proteins (omps) including omp10, omp16, omp19, omp25, omp31 and BP26 as antigens and evaluated their efficiency in serodiagnosis for brucellosis. The efficiency assay was conducted using the method of indirect enzyme-linked immunosorbent assay (iELI

Single-cell transcriptomic atlas of ovarian endometriosis patient's follicular fluid: unraveling novel insights into abnormal follicular development.

In brief: We present the first single-cell transcriptomic atlas of follicular fluid from an ovarian endometrioma (OMA) patient, revealing a novel TCR+ macrophage subtype, aberrant granulosa cell developmental trajectories, immune dysregulation, and enhanced MIF/MK signaling, providing new insights into mechanisms underlying impaired follicular development in OMA. Abstract: Ovarian endometrioma (OMA), an endometriosis subtype, involves hemorrhagic cysts formed by ectopic endometrial-like tissue, causing pelvic pain, inflammation, and infertility by disrupting ovarian function. Despite its impac

Gonadotropin-Releasing Hormone Analog Followed by Sex Hormone Therapy as a Preventive Strategy against Ovarian Complications in Congenital Lipoid Adrenal Hyperplasia: A Case Report.

BACKGROUND: Congenital lipoid adrenal hyperplasia (lipoid CAH) is caused by pathogenic variants in STAR. In 46,XX patients, gonadotropin-driven cholesterol ester accumulation leads to premature ovarian insufficiency, with approximately one-third developing large ovarian cysts or torsion often requiring surgical intervention. No preventive strategy is established. We report a 46,XX patient treated with a gonadotropin-releasing hormone (GnRH) analog followed by estrogen and progestin, with longitudinal ovarian monitoring. Divergent functions of the RNA-binding protein, Mei-P26 in germline and somatic lineages of Drosophila testis.

The Drosophila TRIM-NHL family RNA-binding protein Mei-P26 has been implicated in suppressing tumorigenesis in the female germline. By restricting growth and proliferation while promoting differentiation within the ovarian stem cell lineage, Mei-P26 maintains germline homeostasis. Whether it plays comparable or distinct roles in the testis during spermatogenesis, however, remains unclear. Here we show that overexpression of mei-P26 in germline cells leads to truncated, agametic testes. These testes display germline cell cycle arrest and defective somatic cyst cell proliferation and differentia

Ectopic Kidney With Vesicoureteral Reflux and Associated Gonadal and Müllerian Anomalies in a Prepubertal Child: A Case Report.

The association between congenital renal abnormalities and Müllerian duct anomalies (MDAs) has long been recognized. We present an unusual case, which is of a female child with left grade 4 vesicoureteric reflux (VUR) with ectopic pelvic kidney, who underwent laparoscopic extravesical ureteric reimplantation. Incidental findings on laparoscopy were a left-sided unicornuate uterus placed below the iliac vessels and an ectopic right ovary placed near the deep inguinal ring, which was missed on imaging. Parents of the patient were counselled regarding the complications associated with Müllerian a
