Prenatal Androgenization Modifies H3K9me3 Binding in the Promoter of the Androgen Receptor Gene in the Arcuate Nucleus of the Adult Female Mouse.

Animal models have shown that prenatal exposure to excess androgens is associated with the development of polycystic ovary syndrome (PCOS) features. We have identified that prenatally androgenized (PNA) mice modelling PCOS show suppressed androgen receptor mRNA (Ar) expression in the arcuate nucleus (ARC) across development. This could contribute to PCOS-related impaired gonadal steroid hormone feedback to GnRH neurons. However, the mechanism of Ar mRNA suppression following PNA is not determined. We performed a chromatin immunoprecipitation (ChIP) assay coupled with quantitative PCR (qPCR) to

Incidental diagnosis of an accessory ovary during cesarean section: a case report and literature review.

INTRODUCTION AND IMPORTANCE: An accessory ovary refers to the presence of more than two ovaries, with a third ovary located near a eutopic ovary and directly connected to the normal ovary. Due to its rarity, limited evidence exists regarding optimal management, particularly when it is diagnosed during pregnancy. CASE PRESENTATION: A 31-year-old multigravida woman underwent a cesarean section at term for obstetric indications. The pregnancy had been uneventful, and no adnexal abnormality had been identified during the antenatal period. During intraoperative exploration, two adjacent ovaries wi

Regulation of Lipid Biology-Associated Gene Expression in Granulosa Cells in Hyperandrogenic PCOS: A Possible Link Between Dyslipidemia and Hyperandrogenism.

Polycystic ovary syndrome (PCOS) is a heterogeneous reproductive endocrine condition in women, with implications in fertility and long-term metabolic health. PCOS with hyperandrogen (HA-PCOS; hyperandrogenic PCOS) has been recently identified as one of the four subtypes of PCOS. Dyslipidemia is known to be associated with clinical hyperandrogenism in PCOS. Indeed, patients with HA-PCOS were found to have the highest incidence of dyslipidemia among patients with the other three subtypes of PCOS. In the present study, we identified genes involved in lipid-associated processes (namely, lipid bios

Fertile Ground for Collaboration: A survey of pediatric surgeons on pediatric and adolescent gynecologic care.

BACKGROUND: Pediatric and Adolescent Gynecology (PAG) emerged to bridge gaps between pediatrics and adult gynecology, while pediatric surgeons currently provide most operative gynecologic care for young patients. The goal of this survey was to understand pediatric surgical subspecialty perspectives on access to PAG services, perceived management patterns for common gynecologic conditions, and to assess opportunities for collaborative clinical and surgical care with PAG providers METHODS: A voluntary, anonymous, eight-question descriptive survey was distributed to members of the American Academ

A new constitutive Foxl2-Cre mouse model reveals Foxl2-derived cell fate in reproductive and non-reproductive tissues†.

FOXL2 is a highly conserved transcription factor that is essential for ovarian development and female fertility. In humans, FOXL2 mutations or deletions cause blepharophimosis, ptosis, and epicanthus inversus syndrome (BPES) leading to eyelid defects and, in BPES type I, primary ovarian insufficiency. BPES is also associated with a wide range of less common phenotypes such as craniofacial features, cleft palate, or heart defects. In this study, we generated a new constitutive Foxl2-Cre mouse model using CRISPR/Cas9 editing. Crossing Foxl2-Cre mice with Rosa-tdTomato reporter confirmed robust a

Ectopic adrenal tissue in the mesosalpinx: a rare incidental finding in an adult woman.

Ectopic adrenal tissue is most often identified incidentally on histopathological examination. It is reported mainly in male children and is usually located within the abdominopelvic cavity. Although adrenal cortical rests have been described in the female genital tract, involvement of the mesosalpinx (paratubal region) is exceptionally rare. We report a microscopic adrenal cortical rest in the left mesosalpinx of a 47-year-old woman who underwent total abdominal hysterectomy with bilateral salpingo-oophorectomy for abnormal uterine bleeding and evaluation of bilateral complex ovarian cysts. Hypoadiponectinemia does not enhance anxiety-like behaviour in a lean PCOS-like mouse model.

Polycystic ovary syndrome (PCOS) is characterized by reproductive and metabolic disturbances and associated with higher prevalence of anxiety and depression. Circulating adiponectin, an insulin-sensitizing adipokine, is reduced in women with PCOS, and low adiponectin has been linked to impaired mental health, particularly in females. We investigated whether low serum adiponectin is associated with impaired mental health in women with PCOS and whether adiponectin deficiency exacerbates anxiety-like behaviour in a PCOS-like mouse model. Serum adiponectin was measured in women with (n = 179) and

Maternal polycystic ovary syndrome and the risk of neurodevelopmental disorders in preterm neonates.

INTRODUCTION: Polycystic ovary syndrome (PCOS) is a common endocrine disorder associated with increased obstetric complications. However, its impact on the neurodevelopment of preterm neonates remains underexplored, particularly in low-resource settings. The study aimed to assess the association between maternal PCOS and the risk of neurodevelopmental disorders in preterm neonates. MATERIAL AND METHODS: A prospective case-control study was conducted at Azadi Teaching Hospital, Kirkuk, Iraq, from June 2023 to June 2025. A total of 200 women with preterm labor were enrolled and divided into two

A novel Kazal-type protease inhibitor from Aedes aegypti effective for NS2B-NS3 protease from the Zika virus and subtilases.

In recent years, Zika virus infections have spread geographically across various territories, including French Polynesia, the South Pacific, and South America, with a significant impact on Brazil. Severe Zika virus infections can lead to neurological complications, such as Guillain-Barré syndrome in adults. Furthermore, infection during pregnancy can result in microcephaly and other congenital brain malformations in the fetus. Aedes aegypti cohabits with humans and is the principal vector responsible for transmitting Zika virus to humans. Recent advances have elucidated the molecules involved

Dihydroartemisinin for the treatment of polycystic ovary syndrome: study protocol for a multi-centre placebo-controlled randomised clinical trial.

INTRODUCTION: Polycystic ovary syndrome (PCOS) is the most common endocrine disorder that affects reproductive-aged individuals. Our previous single-arm pilot trial showed that dihydroartemisinin (DHA) normalised menstrual cycles, ameliorated hyperandrogenaemia and reduced antral follicles in individuals with PCOS. However, no comparative evidence confirms the efficacy of DHA in individuals with PCOS when compared with placebo. METHODS AND ANALYSIS: This is a multi-centre randomised, double-blinded, placebo-controlled trial of individuals with PCOS who have irregular menstrual cycles and hype

Hereditary PCOS and the environmental toxicant F-53B converge on lipid metabolic reprogramming to impair fetal brain development.

Polycystic ovary syndrome (PCOS) is associated with an increased risk of neurodevelopmental disorders in offspring, yet how maternal PCOS interacts with environmental toxicants to influence fetal brain development remains unclear. We hypothesized that an AMH-programmed PCOS-like background increases neurodevelopmental vulnerability, which is worsened by gestational F-53B exposure through lipid metabolic reprogramming. Our study reveals how endocrine-metabolic dysfunction and environmental toxicants interact to impact fetal brain development. F0 dams were exposed to anti Müllerian hormone in lat

Functional identification of miR-21-5p from fallopian tubal extracellular vesicles as a promoter of preimplantation embryo viability.

In brief: Human fallopian tube extracellular vesicles (EVs) enhance preimplantation embryo development by delivering miR-21-5p, which suppresses PTEN/p53-dependent apoptosis and promotes blastocyst formation. Restoring this physiological miRNA signal may offer a biomimetic strategy to optimize embryo culture in in-vitro fertilization. Abstracts: Extracellular vesicles (EVs) released by the maternal reproductive tract provide essential biochemical support for early embryo development, yet the identity of their functional molecular cargo remains largely unclear. In this study, we isolated and ch

Peripheral precocious puberty with unilateral ovarian agenesis in early childhood: a case report and review of literature.

Peripheral precocious puberty (PPP) is a rare condition in early childhood, particularly among girls under 3 years of age. One cause of PPP is functional ovarian cysts that secrete estrogen at levels sufficient to trigger the development of premature secondary sexual characteristics. Unilateral ovarian agenesis is an uncommon congenital anomaly. The coexistence of both conditions in a single patient is extremely rare and poses unique diagnostic and management challenges. We report the case of a 2-year and 10-month-old girl presenting with concurrent breast development, pubarche, and recurrent

Malignant ovarian teratoma in a Lusitano filly: Diagnostic challenges and clinical significance.

An 8-month-old Lusitano filly (a young female horse) was referred with a five-day history of abdominal distension, respiratory distress, and hyporexia. Clinical and ultrasonographic examinations revealed ventral edema, anemia, leukocytosis, hypoalbuminemia, and bilateral pleural effusion. The filly subsequently developed progressive clinical deterioration, including persistent hyporexia, scant fecal output, and acute colic with nasogastric reflux, followed by sudden respiratory decompensation, collapse, and death. Post-mortem examination revealed a large, multilobulated abdominal mass involvin

A rare case of canal of Nuck cyst in a nulliparous woman of reproductive age: A case report and literature review.

The cyst of the canal of Nuck is a rare developmental disorder of the female inguinal region. It arises from the failure of the obliteration of the peritoneal fold accompanying the round ligament through the inguinal canal. Although predominantly encountered in the pediatric population, its occurrence in women of reproductive age presents a significant diagnostic challenge, given the overlap with more common inguinal pathologies. This report describes the case of a 33-year-old nulliparous woman who presented with a six-month history of progressive left inguinal swelling. Pelvic magnetic resona

Approach to the diagnosis and treatment of polycystic ovarian syndrome in adolescent patients.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders affecting women of reproductive age. Its diagnosis during adolescence is particularly challenging, as physiological pubertal changes overlap with key features of PCOS; nevertheless, this developmental stage often represents the earliest window for clinical manifestation. Accordingly, a definitive diagnosis should be established only in the presence of clear and persistent evidence of both hyperandrogenism and menstrual irregularity. Adolescents presenting with a single feature should be considered at increased risk

The Ontogeny of Distinct Reproductive Phenotypes in Girls at Risk for PCOS During the Postmenarchal Transition.

CONTEXT: Daughters of women with polycystic ovary syndrome (PCOS-d) and girls with overweight/obesity (OW-g) have hyperandrogenemia (HA) beginning in childhood. However, other features of their early reproductive phenotypes differ, suggesting that there are distinct mechanisms conferring increased risk for PCOS. OBJECTIVE: We performed a cross-sectional study of adolescent girls during the early postmenarchal transition. DESIGN, SETTING, AND PARTICIPANTS: PCOS-d (n = 15), OW-g (n = 12), and lean control girls (LC; n = 17) were studied within .2 to 1.2  years of menarche. Metabolic and reprod

Hypertension and dyslipidemia in women with PCOS: a population-based multiregister study in Sweden.

STUDY QUESTION: What is the impact of PCOS and the hyperandrogenic (HA) PCOS phenotype on the risk of developing hypertension and dyslipidemia? SUMMARY ANSWER: PCOS is an independent risk factor for the development of hypertension and dyslipidemia, with the risk being higher among women with the HA PCOS phenotype. WHAT IS KNOWN ALREADY: PCOS is an established risk factor for insulin resistance and the metabolic syndrome. However, prospective data regarding the risk of hypertension and dyslipidemia in population-based cohorts of women with PCOS are limited. STUDY DESIGN, SIZE, DURATION: This

Effects of peripubertal onset of hyperandrogenism on sociosexual behavior of mice modeling polyendocrine metabolic ovarian syndrome.

Sex differences in circulating androgens are well-known to organize and activate neural and behavioral sex differences of traditional animal models, but much less is known about the consequences of intrasexual differences in androgens. One approach to this question is studying the effects of hyperandrogenism in females. Hyperandrogenism is associated with several endocrine conditions affecting women, with polyendocrine metabolic ovarian syndrome (PMOS) being the most common during reproductive years. PMOS symptoms often emerge around puberty, raising the possibility of organizational effects o
