INTRODUCTION: The SOX9 gene encodes a transcription factor that acts downstream of the Y-linked SRY gene and plays a pivotal role in fetal testis development. Duplication of SOX9 or its regulatory sequences is a known cause of testicular or ovotesticular disorder of sex development (DSD) in chromosomal females (XX DSD). Numerous reports have described canine XX DSD, characterized by virilization (e.g., enlarged clitoris) and the presence of testes or ovotestes. This study aimed to identify SOX9 variants in a cohort of French Bulldogs with XX (SRY-negative) DSD.
METHODS: In total, 27 DSD dogs

Background & objectives Polycystic Ovary Syndrome (PCOS) is an endocrine disorder affecting reproductive-age women worldwide. Lipid abnormalities, such as elevated low-density lipoprotein (LDL) and triglyceride (TG) levels and reduced high-density lipoprotein (HDL) levels, are commonly observed in women with PCOS, increasing their risk of cardiovascular disease (CVD). Therefore, this study aims to quantify the magnitude and pattern of lipid levels (total cholesterol, HDL, LDL, and TG) in women with different phenotypes of PCOS versus control women. Methods Worldwide published observational (cr

BACKGROUND: Adrenocortical hyperplasia is an autosomal recessive disorder characterized by congenital metabolic dysfunction, resulting in endocrine disturbances and abnormal sexual development (such as female masculinity), requiring lifelong hormonal therapy. Notably, non-classical congenital adrenal hyperplasia (NCCAH) exhibits clinical manifestations resembling polycystic ovary syndrome (PCOS), yet their therapeutic approaches in fertility treatment differ substantially. Importantly, NCCAH carries the risk of delivering offspring with severe adrenal hyperplasia, which presents far graver con

BACKGROUND: Non-classical congenital adrenal hyperplasia (NCCAH) is a milder variant of 21-hydroxylase deficiency, an autosomal recessive disorder leading to impaired cortisol biosynthesis and compensatory adrenal androgen excess. Unlike the classical form, NCCAH typically presents later in life, often mimicking polycystic ovary syndrome (PCOS) in women and remaining largely asymptomatic in men.
METHODS: A literature search was conducted in MEDLINE (PubMed) in October 2024 using the term "Non-Classical Congenital Adrenal Hyperplasia" in combination with keywords related to sex differences, di

IN BRIEF: A 'two hit' developmental origin involving testosterone and anti-Müllerian hormone is proposed to initiate PCOS pathogenesis during gestation. Epigenetic mechanisms amplify genetically heritable traits, while accompanying metabolic perturbations, including gestational hyperglycemia, hypertension and maternal obesity, exaggerate PCOS expression.
ABSTRACT: Pre- or perinatal excess of anti-Müllerian hormone (AMH) or testosterone faithfully reproduce many polycystic ovary syndrome (PCOS)-like reproductive and metabolic traits in animal models. Epigenetic transgenerational transmission o

Endometriomas can be managed through various approaches, including careful observation, medical suppressive therapy, or surgical intervention. Malignancy is rarely a concern in women of reproductive age, making pain relief and the management of infertility the primary foci of treatment. Pain is seldom due to endometriomas per se, and is most often associated with deep endometriotic lesions, usually involving the parametria and the bowel. Patients with infertility are better served with assisted reproductive technology (ART), as endometriomas do not appear to affect oocyte developmental potenti

INTRODUCTION: Clinical characteristics of polycystic ovary syndrome (PCOS) (menstrual irregularities, obesity, anovulation, etc.), occur together with a series of conditions at skin level like seborrhea, acne, hirsutism, and androgenetic alopecia. Estroprogestins (EPs) are recognized as the most powerful therapy to treat hyperandrogenism and/or hyperandrogenemia and regularize menstrual cycle.
EVIDENCE ACQUISITION: This review includes the most relevant publications about the effects of EPs in PCOS women published between 1995 and 2025.
EVIDENCE SYNTHESIS: According to the available data, EP

Polycystic ovary syndrome (PCOS) is a common and heterogeneous endocrine disorder that affects 11%-13% of women worldwide, with profound implications for fertility and long-term metabolic health. Here we identify four reproducible subtypes-PCOS with hyperandrogen, with obesity, with high-sex hormone-binding globulin and with high-luteinizing hormone-anti-Müllerian hormone-through unsupervised clustering of 9 clinical variables in 11,908 affected women, validated across 5 international cohorts. Prospective 6.5-year follow-up and in vitro fertilization treatment data revealed distinct reproducti

11-beta hydroxylase deficiency (11βOHD) is a rare variant of congenital adrenal hyperplasia (CAH) with autosomal recessive inheritance, resulting in androgen excess and mineralocorticoid precursor accumulation. Fertility is often impaired due to hyperandrogenism and anatomical abnormalities, and spontaneous successful pregnancy in classic form is rare. We describe two cases. Case 1 is a 27-year-old woman with genetically confirmed classic 11βOHD (homozygous CYP11B1 splice-site variant c.240-2A>G), initially misdiagnosed as 21-hydroxylase deficiency, who achieved spontaneous conception despite

BACKGROUND: While oocyte IVM is practiced sporadically it has not achieved widespread clinical practice globally. However, recently there have been some seminal advances in our understanding of basic aspects of oocyte biology and ovulation from animal studies that have led to novel approaches to IVM. A significant recent advance in IVM technology is the use of biphasic IVM approaches. These involve the collection of immature oocytes from small antral follicles from minimally stimulated patients/animals (without hCG-priming) and an ∼24 h pre-culture of oocytes in an advanced culture system ('pr

CONTEXT: Polycystic ovary syndrome (PCOS) has historically been conceptualized as a disorder of the reproductive system in women. However, offspring of women with PCOS begin to show metabolic features of PCOS in childhood, suggestive of childhood manifestations.
OBJECTIVE: To identify childhood manifestations of genetic risk for PCOS.
METHODS: We calculated a PCOS polygenic risk score (PRS) for 12 350 girls and boys in 4 pediatric cohorts-ALSPAC (UK), COPSAC (Denmark), Project Viva (USA), and The HOLBÆK Study (Denmark). We tested for association of the PRS with PCOS-related phenotypes throug

Human granulosa cells in different stages are essential for maintaining normal ovarian function, and granulosa cell defect is the main cause of ovarian dysfunction. To address this problem, it is necessary to induce functional granulosa cells at different stages in vitro. In this study, we established a reprogramming method to induce early- and late-stage granulosa cells with different steroidogenic abilities. We used an AMH-fluorescence-reporter system to screen candidate factors for cellular reprogramming and generated human induced granulosa-like cells (hiGC) by overexpressing FOXL2 and NR5

Polycystic Ovary Syndrome (PCOS) is a prevalent endocrine disorder in women of reproductive age, affecting 5-15% globally with a large proportion undiagnosed. This review explores the multifaceted nature of PCOS and its impact on pregnancy, including challenges in fertility due to hormonal imbalances and insulin resistance. Despite restoring ovulation pharmacologically, women with PCOS face lower pregnancy rates and higher risks of implantation failure and miscarriage. Our review focuses on the complexities of hormonal and metabolic imbalances that impair endometrial receptivity and decidualiz

This study was conducted in two groups of girls with PCOS (polycystic ovary syndrome) categorized as slim (group N) and overweight-to-obese (group Ov/Ob). The study's primary outcome was to assess the impact of a 12-week anti-inflammatory diet (AIDiet) intervention, without energy deficit, on daily diet quality improvement, evaluated according to the KIDMED index. The secondary outcome was improving inflammatory, redox, hormonal, and metabolic statuses. In the study, which was completed by 13 girls from the Ov/Ob group and 19 girls from the N group, a significant improvement in the mean KIDMED

Polycystic ovary syndrome is a common endocrine disorder in reproductive-age women. Accordingly, abnormal microenvironment may negatively influence oocyte developmental competence as a result of the altered expression profile of cumulus cells (CCs), mainly the key players of oocyte maturation, such as epidermal growth factor receptor (EGFR) and prostaglandin E receptor-2 (PTGER2). This study aimed to examine the expression levels of miR-514, miR-642b, and their candidate target genes (EGFR and PTGER2, respectively) in CCs of immature and mature oocytes in patients with PCOS. A total of 40 oocy

Polycystic ovary syndrome (PCOS) is a common endocrine disorder associated with infertility and poor reproductive outcomes. The follicular fluid (FF) microenvironment plays a crucial role in oocyte development. This review summarizes evidence elucidating the alterations in FF composition in PCOS. Various studies demonstrated a pronounced proinflammatory milieu in PCOS FF, characterized by increased levels of cytokines, including but not limited to interleukin-6 (IL-6), tumor necrosis factor α, C-reactive protein, and IL-1β, concomitant with a reduction in anti-inflammatory IL-10. T lymphocytes

Abnormal autophagy is one of the vital features in polycystic ovary syndrome (PCOS). However, the underlying molecular mechanisms remain unelucidated. In this study, we aimed to investigate whether Block of Proliferation 1 (BOP1) is involved in the onset of autophagy activation of granulosa cells in PCOS. Firstly, we found that BOP1 expression was significantly down-regulated in the ovaries of PCOS mice, which was associated with the development of PCOS. Next, local injection of lentiviral vectors in the ovary for the overexpression of BOP1 significantly alleviated the phenotypes of elevated a

BACKGROUND: Genetic predisposition and environmental factors are considered risk factors for polycystic ovary syndrome (PCOS). Genome-wide association studies (GWAS) have been reported from various subpopulations to evaluate SNPs associated with PCOS risk. No PCOS-associated GWAS study has been reported from India so far.
PURPOSE: The current study was conducted to identify the PCOS-susceptible loci among the North Indian population and to validate the significant loci reported by previous GWAS studies.
METHODS: A total of 272 participants with 134 PCOS patients and 138 age-matched healthy c
