Circadian Biology and Gynecological Diseases: Genetic Mechanisms and Potential Therapeutic Targets Underlying the Causal Link Between Insomnia and Ovarian Cysts.

BACKGROUND: Although circadian disruption has been implicated in gynecologic malignancies, genetic evidence for a causal relationship between insomnia and ovarian cysts remains lacking. The biological pathways linking sleep disturbance to ovarian pathology are therefore poorly understood, limiting mechanistic insights and therapeutic development. METHODS: We employed a two-sample Mendelian Randomization (MR) framework using large-scale Genome-Wide Association Study (GWAS) datasets of European ancestry to examine the effects of seven sleep-related traits on five gynecologic outcome types. We f

Neuropsychiatric disorders in children of mothers with polycystic ovary syndrome: a systematic review and meta-analysis.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a common endocrine disorder among women of reproductive age, yet its association with neuropsychiatric disorders (NPDs) in offspring remains inconsistent and requires updated synthesis. METHODS: We systematically searched PubMed, Embase, Web of Science, and Cochrane Library up to March 2025. A total of 21 observational studies involving 6.8 million mothers and 7.4 million offspring were included. Quality assessment was performed using the Newcastle-Ottawa Scale. Random-effects meta-analyses were conducted to estimate pooled odds ratios (ORs) with

Epigenetic safety of in vitro maturation in PCOS: genome-wide DNA methylation profiling of cord blood from a randomized controlled trial.

BACKGROUND: In vitro maturation (IVM) provides a safer alternative to conventional in vitro fertilization (IVF) for women with polycystic ovary syndrome (PCOS) by mitigating the risk of ovarian hyperstimulation. However, concerns persist regarding whether IVM perturbs epigenetic reprogramming in the offspring. Current evidence is constrained by candidate-gene approaches or a lack of parental controls. This study aimed to evaluate the genome-wide DNA methylation safety of IVM compared with conventional IVF using a rigorous trio-based design. METHODS: This secondary epigenetic analysis was nest

[Magnetic resonance imaging in the differential diagnosis of fetal abdominopelvic cysts: a retrospective case series].

INTRODUCTION: Abdominopelvic cystic lesions are relatively common findings during pregnancy. Ultrasound examinations performed during pregnancy can easily identify these cysts; however, accurate characterization is not always possible using ultrasound alone. The introduction of rapid magnetic resonance (MR) sequences allows more precise characterization of these lesions and also assists in therapeutic decision-making. OBJECTIVE: To identify fetal abdominopelvic cysts and assess their characteristics, ranging from the most common presentations to less frequently occurring cases. METHOD: In th

Genomic analyses implicate hormonal and metabolic dysregulation in polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) and its underlying features remain poorly understood. In this genetic study (n = 544,513), we expand the number of genetic loci from 16 to 29, and additionally identify 31 associated plasma proteins. Many risk-increasing loci were associated with later age at menopause, underscoring the reproductive longevity related to an increased oocyte number and/or availability across the lifespan. Hormonal regulation in the etiology of this condition, through metabolic and reproductive features, was emphasized. The proteomic analysis highlighted metabolic biology known to

Ecdysone receptor autonomously controls germ cell differentiation in the Drosophila ovary.

BACKGROUND: Brucellosis is a zoonotic disease that causes substantial public health problems and endangers the development of animal husbandry in endemic areas. Early diagnosis of infected animals and humans is a crucial step in reducing the incidence of brucellosis. In this study, we designed different combinations of Brucella major outer membrane proteins (omps) including omp10, omp16, omp19, omp25, omp31 and BP26 as antigens and evaluated their efficiency in serodiagnosis for brucellosis. The efficiency assay was conducted using the method of indirect enzyme-linked immunosorbent assay (iELI

Single-cell transcriptomic atlas of ovarian endometriosis patient's follicular fluid: unraveling novel insights into abnormal follicular development.

In brief: We present the first single-cell transcriptomic atlas of follicular fluid from an ovarian endometrioma (OMA) patient, revealing a novel TCR+ macrophage subtype, aberrant granulosa cell developmental trajectories, immune dysregulation, and enhanced MIF/MK signaling, providing new insights into mechanisms underlying impaired follicular development in OMA. Abstract: Ovarian endometrioma (OMA), an endometriosis subtype, involves hemorrhagic cysts formed by ectopic endometrial-like tissue, causing pelvic pain, inflammation, and infertility by disrupting ovarian function. Despite its impac

Case Report: Rare triple-line chromosome 9 mosaicism (47,XX,+del(9)(q13)/47,XX,+9/46,XX) associated with severe neurodevelopmental impairment and congenital anomalies.

BACKGROUND: Chromosome 9 abnormalities are rare. Full trisomy 9 is mostly incompatible with postnatal survival, whereas mosaic trisomy 9 permits survival with highly variable clinical manifestations. Triple-line mosaicism involving both full trisomy 9 and structural chromosome 9 abnormalities is exceptionally rare. CASE PRESENTATION: A 1-year-old girl, conceived via IVF, had normal nuchal translucency and NIPT. Prenatal ultrasound showed a single umbilical artery, lower abdominal cyst, borderline ventriculomegaly at 28 weeks, and late polyhydramnios. She was born at 40 weeks, 3,430 g, 49 cm,

Divergent functions of the RNA-binding protein, Mei-P26 in germline and somatic lineages of Drosophila testis.

The Drosophila TRIM-NHL family RNA-binding protein Mei-P26 has been implicated in suppressing tumorigenesis in the female germline. By restricting growth and proliferation while promoting differentiation within the ovarian stem cell lineage, Mei-P26 maintains germline homeostasis. Whether it plays comparable or distinct roles in the testis during spermatogenesis, however, remains unclear. Here we show that overexpression of mei-P26 in germline cells leads to truncated, agametic testes. These testes display germline cell cycle arrest and defective somatic cyst cell proliferation and differentia

Prenatal Androgenization Modifies H3K9me3 Binding in the Promoter of the Androgen Receptor Gene in the Arcuate Nucleus of the Adult Female Mouse.

Animal models have shown that prenatal exposure to excess androgens is associated with the development of polycystic ovary syndrome (PCOS) features. We have identified that prenatally androgenized (PNA) mice modelling PCOS show suppressed androgen receptor mRNA (Ar) expression in the arcuate nucleus (ARC) across development. This could contribute to PCOS-related impaired gonadal steroid hormone feedback to GnRH neurons. However, the mechanism of Ar mRNA suppression following PNA is not determined. We performed a chromatin immunoprecipitation (ChIP) assay coupled with quantitative PCR (qPCR) to

Ectopic adrenal tissue in the mesosalpinx: a rare incidental finding in an adult woman.

Ectopic adrenal tissue is most often identified incidentally on histopathological examination. It is reported mainly in male children and is usually located within the abdominopelvic cavity. Although adrenal cortical rests have been described in the female genital tract, involvement of the mesosalpinx (paratubal region) is exceptionally rare. We report a microscopic adrenal cortical rest in the left mesosalpinx of a 47-year-old woman who underwent total abdominal hysterectomy with bilateral salpingo-oophorectomy for abnormal uterine bleeding and evaluation of bilateral complex ovarian cysts. Hypoadiponectinemia does not enhance anxiety-like behaviour in a lean PCOS-like mouse model.

Polycystic ovary syndrome (PCOS) is characterized by reproductive and metabolic disturbances and associated with higher prevalence of anxiety and depression. Circulating adiponectin, an insulin-sensitizing adipokine, is reduced in women with PCOS, and low adiponectin has been linked to impaired mental health, particularly in females. We investigated whether low serum adiponectin is associated with impaired mental health in women with PCOS and whether adiponectin deficiency exacerbates anxiety-like behaviour in a PCOS-like mouse model. Serum adiponectin was measured in women with (n = 179) and

Hereditary PCOS and the environmental toxicant F-53B converge on lipid metabolic reprogramming to impair fetal brain development.

Polycystic ovary syndrome (PCOS) is associated with an increased risk of neurodevelopmental disorders in offspring, yet how maternal PCOS interacts with environmental toxicants to influence fetal brain development remains unclear. We hypothesized that an AMH-programmed PCOS-like background increases neurodevelopmental vulnerability, which is worsened by gestational F-53B exposure through lipid metabolic reprogramming. Our study reveals how endocrine-metabolic dysfunction and environmental toxicants interact to impact fetal brain development. F0 dams were exposed to anti Müllerian hormone in lat

A rare case of canal of Nuck cyst in a nulliparous woman of reproductive age: A case report and literature review.

The cyst of the canal of Nuck is a rare developmental disorder of the female inguinal region. It arises from the failure of the obliteration of the peritoneal fold accompanying the round ligament through the inguinal canal. Although predominantly encountered in the pediatric population, its occurrence in women of reproductive age presents a significant diagnostic challenge, given the overlap with more common inguinal pathologies. This report describes the case of a 33-year-old nulliparous woman who presented with a six-month history of progressive left inguinal swelling. Pelvic magnetic resona

Approach to the diagnosis and treatment of polycystic ovarian syndrome in adolescent patients.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders affecting women of reproductive age. Its diagnosis during adolescence is particularly challenging, as physiological pubertal changes overlap with key features of PCOS; nevertheless, this developmental stage often represents the earliest window for clinical manifestation. Accordingly, a definitive diagnosis should be established only in the presence of clear and persistent evidence of both hyperandrogenism and menstrual irregularity. Adolescents presenting with a single feature should be considered at increased risk

The Ontogeny of Distinct Reproductive Phenotypes in Girls at Risk for PCOS During the Postmenarchal Transition.

CONTEXT: Daughters of women with polycystic ovary syndrome (PCOS-d) and girls with overweight/obesity (OW-g) have hyperandrogenemia (HA) beginning in childhood. However, other features of their early reproductive phenotypes differ, suggesting that there are distinct mechanisms conferring increased risk for PCOS. OBJECTIVE: We performed a cross-sectional study of adolescent girls during the early postmenarchal transition. DESIGN, SETTING, AND PARTICIPANTS: PCOS-d (n = 15), OW-g (n = 12), and lean control girls (LC; n = 17) were studied within .2 to 1.2  years of menarche. Metabolic and reprod

Effects of peripubertal onset of hyperandrogenism on sociosexual behavior of mice modeling polyendocrine metabolic ovarian syndrome.

Sex differences in circulating androgens are well-known to organize and activate neural and behavioral sex differences of traditional animal models, but much less is known about the consequences of intrasexual differences in androgens. One approach to this question is studying the effects of hyperandrogenism in females. Hyperandrogenism is associated with several endocrine conditions affecting women, with polyendocrine metabolic ovarian syndrome (PMOS) being the most common during reproductive years. PMOS symptoms often emerge around puberty, raising the possibility of organizational effects o

Amniotic-fluid metabolomics identifies phospholipid remodeling as a metabolic signature of intrauterine exposure in pregnancies with polycystic ovary syndrome.

BACKGROUND: Polycystic ovary syndrome is associated with metabolic and hormonal disturbances during pregnancy, but whether these alterations are reflected in the fetal intrauterine exposure environment remains incompletely understood. This study aimed to identify polycystic ovary syndrome-related intrauterine metabolic signatures using late-gestation amniotic fluid. METHODS: Untargeted metabolomic profiling was performed on late-gestation amniotic fluid samples from women with polycystic ovary syndrome and controls. Differential metabolite analysis, pathway-level analysis, and multilevel sens
