In vitro oogenesis breaks free of the ovary.

In this issue of Developmental Cell, Nosaka et al. differentiate mouse pluripotent stem cells to large germinal vesicle-stage oocyte-like cells in the absence of ovarian somatic cells. Their paradigm advances the field toward clinical translation and offers insights into oogonial cyst breakdown, X chromosome dynamics, and requirements for oocyte growth and meiotic resumption.

Multi-ancestry genome-wide association analyses of polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS), the leading endocrine disorder in women of reproductive age, is highly heritable, yet its polygenic architecture remains poorly understood. Here we conducted a genome-wide association study on 12,419 Chinese women with PCOS and 34,235 controls, followed by a multi-ancestry meta-analysis with up to 13,773 European cases and 411,088 controls, identifying 94 independent loci, 73 of which were previously unreported. Despite different evolutionary pressures, Chinese and European ancestries showed substantial genetic overlap. Integrative functional analyses prioriti

Preconception, gestation, and childhood exposure to air pollution and risk of polycystic ovary syndrome (PCOS) in a US girls cohort study.

BACKGROUND: Exposure to ambient air pollution (AP) during sensitive developmental periods could dysregulate the reproductive system, resulting in later in life menstrual disorders, including polycystic ovary syndrome (PCOS). Minimal research exists examining the relationship between AP exposure and PCOS. OBJECTIVES: To determine if residential ambient particulate matter (PM) and nitrogen dioxide (NO2) exposure during preconception, gestation, and childhood increases the risk of PCOS. METHODS: We used data from 3,321 female participants with gestational data available born between 1989 and 19

Acetylation reader BRD4-driven TXNIP transcription enhances NLRP3 inflammasome activation in PCOS.

Polycystic ovary syndrome (PCOS) is an endocrine disorder characterized by the developmental arrest and dysfunction of ovarian granulosa cells (GCs), serving as a major cause of infertility among women of reproductive age. Persistent activation of thioredoxin-interacting protein (TXNIP) due to aberrant histone acetylation modifications of transcription is a potential trigger; however, its precise upstream regulatory mechanism remains poorly understood. In this study, we found that TXNIP was aberrantly upregulated in both the dehydroepiandrosterone (DHEA)-induced PCOS-like rat model and the dih

Conditioned media and extracellular vesicles derived from human Wharton's jelly mesenchymal stem cells improve the in vitro maturation of immature oocytes in normal and PCOS mouse model.

BACKGROUND: The effects of conditioned medium (CM) and extracellular vesicles (EVs) derived from human Wharton's jelly mesenchymal stem cells (hWJMSCs) on in vitro maturation (IVM) of immature oocytes in both normal and polycystic ovary syndrome (PCOS)-induced mice were investigated. PCOS was induced in adult female NMRI mice by administering letrozole (90 μg/kg/day) via gavage for one week. Germinal vesicle (GV) oocytes were collected from both PCOS-induced and normal mice, while mature oocytes (MII) were obtained from superovulated normal mice to serve as controls. The experimental groups in

Advances, Mechanisms, and Clinical Perspectives for the In Vitro Maturation of Human Oocytes.

The in vitro maturation (IVM) of human oocytes represents a valuable assisted reproductive technology that bypasses the need for full ovarian stimulation, offering safer alternatives for patients with polycystic ovary syndrome (PCOS), resistant ovary syndrome, or those requiring fertility preservation before oncological treatment. Despite its potential, IVM efficiency remains lower than that of conventional in vitro fertilization (IVF) due to incomplete understanding of the molecular and metabolic mechanisms underpinning oocyte maturation. This review summarizes recent advances in IVM, includi

Mental freedom and body acceptance by others as correlates of embodied well-being in women with polycystic ovary syndrome in Australia.

Polycystic ovary syndrome (PCOS) is a common reproductive-endocrinological-metabolic condition in women, causing bodies to diverge from cis-heteronormative feminine expectations. Greater body image concerns have been identified among women with PCOS, but further research is needed to identify drivers and mitigating factors. The ability to inhabit the body positively may be one such mitigating factor, and the developmental theory of embodiment is a novel framework allowing exploration of an integrated range of protective (and risk) sociocultural factors that may contribute to embodied well-bein

Endocrine-disrupting chemicals and reproductive health: With focus on the developmental window of susceptibility.

Sexual differentiation is highly dependent on the fetal hormonal environment, guiding sexual development and establishing the foundation for lifelong reproductive health. This short communication highlights critical windows of sex determination and differentiation, emphasizing the role of steroid sex hormones, alongside other factors, in orchestrating these processes in early life. Growing evidence suggests that endocrine-disrupting chemicals (EDCs) can disrupt these tightly regulated pathways, leading to developmental disturbances that manifest as reproductive disorders at birth or later in l

PCOS endometrium-derived epithelial organoids as a novel model to study endometrial dysfunction.

STUDY QUESTION: Are we able to establish endometrium epithelial organoids (EEOs) from endometrial samples obtained from women with PCOS, and do they differ from non-PCOS EEOs? SUMMARY ANSWER: We were able to establish, for the first time, PCOS EEOs which capture endometrial abnormalities present in women with PCOS, including increased inflammation and decreased receptivity-related gene expression. WHAT IS KNOWN ALREADY: Patient-derived EEOs could serve as a tool to study endometrial dysfunction, as diseased tissue-derived organoid models typically retain the disease-related traits. In PCOS,

Differential Methylation Signatures Associated with PCOS- A Systematic Review and In-Silico Analysis.

Polycystic ovary syndrome (PCOS) is a common endocrinopathy affecting 5-20% of reproductive-age women. Besides genetic factors, environmental triggers are considered major underlying causes that play a role through epigenetic alterations. Since epigenetic studies can be a possible link to explain the complexity of multifactorial disorders, it is worthwhile to investigate differentially methylated regions (DMRs) associated with PCOS. Therefore, a systematic review was conducted to identify epigenetic modifications associated with PCOS and analyse their functional role in the pathogenesis of the

BACKGROUND: Polycystic ovary syndrome (PCOS) is a common endocrine-metabolic disorder affecting 5-15% of women of reproductive age and is a leading cause of female infertility. It is a complex, multifactorial condition in which genetic factors play a significant role and may affect siblings within a family. Here, we present the case of two infertile sisters from an Iranian family. METHODS AND RESULTS: Two sisters with treatment-resistant infertility and a history of PCOS underwent comprehensive genetic evaluation. Randomized controlled trial to evaluate the impact of follicle priming on IVM outcomes in women with polycystic ovaries: CFA versus FSH-B.

STUDY QUESTION: Do corifollitropin alfa (CFA) and follitropin beta (FSH-B) have different effects on oocyte yield and live birth rates (LBRs) after IVM in women with polycystic ovaries? SUMMARY ANSWER: In patients who underwent IVM, one injection of CFA resulted in lower oocyte retrieval rates, but similar cumulative LBRs compared to three injections of FSH-B. WHAT IS KNOWN ALREADY: IVM involves the maturation of cumulus-oocyte complexes (COCs) from antral follicles and has been offered to women with polycystic ovaries as an alternative for conventional ovarian stimulation (OS). In vitro maturation in subfertile women with polycystic ovarian syndrome undergoing assisted reproduction.

BACKGROUND: Polycystic ovarian syndrome (PCOS) occurs in 8% to 13% of all women of reproductive age and 50% of women presenting with infertility (i.e. inability to reach a pregnancy after 12 months or more of regular unprotected sexual intercourse). A proportion of these women ultimately need assisted reproductive technology. In vitro fertilisation (IVF)/intracytoplasmic sperm injection (ICSI) are assisted reproduction techniques used to raise the chances of a pregnancy. In women with PCOS, the supra-physiological doses of gonadotrophins used for controlled ovarian hyperstimulation (COH) often

Endocrine-disrupting chemicals and female reproductive health: a growing concern.

Female fertility and reproductive health depend on a series of developmental steps from embryogenesis through puberty, in addition to the proper functioning of the reproductive system in adulthood. Two important steps are the establishment of the ovarian reserve and development of the hypothalamic-pituitary-ovarian axis. During reproductive years, maintaining an adequate ovarian reserve of follicles as well as balanced neuroendocrine control of reproductive organs is crucial for fertility. Dysregulation of either of these events, during development or in adulthood, can lead to reproductive dis

Metformin use in women with polycystic ovary syndrome (PCOS): Opportunities, benefits, and clinical challenges.

Metformin, a synthetic biguanide, is widely used to manage type 2 diabetes, and is commonly prescribed in polycystic ovary syndrome (PCOS) to address insulin resistance and associated metabolic and reproductive disturbances. PCOS is characterised by hormonal imbalances such as hyperandrogenism and anovulation, metabolic abnormalities including insulin resistance and increased cardiometabolic risk, and higher rates of pregnancy complications. However, the role of metformin in the multifaceted nature of PCOS remains debated. This review synthesises the mechanisms of action of metformin and its e

Exosomal Communication Between Cumulus-Oocyte Complexes and Granulosa Cells: A New Molecular Axis for Oocyte Competence in Human-Assisted Reproduction.

Exosomal microRNAs (ex-miRs), encapsulated in extracellular vesicles (EVs), play a vital role in facilitating paracrine communication among granulosa cells (GCs), cumulus cells (CCs), and the oocyte inside follicular fluid (FF). These small non-coding RNAs are crucial for regulating folliculogenesis, oocyte maturation, and early embryonic development via modulating intracellular signaling networks. Dysregulation o has been associated with reproductive disorders such as polycystic ovarian syndrome (PCOS), diminished ovarian reserve (DOR), and inadequate ovarian response (POR), impacting oocyte
