In vitro assessment of integrated apex locator systems: working length accuracy, operational parameters, and mechanical effects on NiTi instruments.

The past 20 years have seen considerable advances in the field of organ transplantation that have together led to a notable increase in survival rates and a reduction in postoperative morbidity of transplant recipients. However, these advances have been accompanied by the appearance of other complications of transplantation, such as post-transplant hyperlipidaemia, hypertension and graft coronary vasculopathy (GCV). GCV is an accelerated form of atherosclerosis in transplanted hearts that has proven to be one of the most important late complications of heart transplantation and is the single m

Deciphering the Role of ZEB1 as a Central Regulator of Epithelial-Mesenchymal Transition and Hormonal Resistance in Endometriosis.

Endometriosis is a chronic inflammatory and sex steroid hormone-dependent gynaecologic disease, characterised by endometrial cell proliferation outside the uterine cavity, often accompanied by fibrosis, progesterone resistance, and high recurrence despite hormonal and surgical interventions. Epithelial-mesenchymal transition (EMT) is important for establishment of lesions and invasiveness, but the upstream transcriptional regulators that integrate hormonal, inflammatory and epigenetic signals are incompletely defined. This review emphasises ZEB1 as a molecular integrator at the systems level i

Coral rubble facilitates feeding opportunities for invertivorous reef fishes on tropical coral reefs.

BACKGROUND: Endoscopic stapling to treat pharyngeal pouch is a relatively new technique with the potential to reduce the morbidity associated with the open approach for pharyngeal pouch. Despite enthusiasm for the endoscopic approach there have been no series reported in Australia, and descriptions of outcomes and benefits are currently anecdotal. The aim of the present study was to determine the outcome associated with endoscopic stapling of pharyngeal pouch in an Australian setting. METHODS: All patients admitted for endoscopic stapling for a pharyngeal pouch between 1998 and 2002 by surgeo

Eutopic Endometrium Immune Changes Involved in Development and Progression of Endometriosis: A Review.

Numerous abnormalities of the endometriosis eutopic endometrium contribute to the initiation and development of ectopic lesions. It is also believed that among the complex causes of the disease, systemic immunological disorders play a significant role. Therefore, this literature review aims to summarize the current knowledge on immunological alterations in the endometriosis eutopic endometrium and the impact of these changes on the progression of this disease. The reviewed studies mostly indicated a pro-inflammatory immunological profile within this tissue. This was evidenced by a predominance

Women with polycystic ovary syndrome exhibit impaired endometrial receptivity with excessive ERα and histone lactylation.

Polycystic ovary syndrome (PCOS) is one of the most common reproductive disorders in women and severely impairs fertility. Extant clinical studies can only provide indirect and plausible evidence to support endometrial dysfunction as an ovary-independent contributor to PCOS infertility, considering heterogeneous confounders in their phenotypes, comorbidities, and severities. By strictly controlling embryonic factors and potential confounders, our retrospective cohort study reports an adverse implantation rate in women with PCOS, confirming abnormalities in the endometrium, which are accompanie

An endometrial tissue-based predictive model for polycystic ovary syndrome constructed from immuno-metabolic dysregulation features mediated by ACO1.

OBJECTIVE: Polycystic ovary syndrome (PCOS) is a multifactorial endocrine disorder characterized by reproductive and metabolic abnormalities. This study aimed to identify key immunometabolic regulators in endometrial tissue and construct a predictive model for PCOS using machine learning approaches. METHODS: Three endometrial transcriptomic datasets (GSE277906, GSE193123, GSE199225) were integrated and analyzed for differentially expressed genes (DEGs), immune cell infiltration, and metabolic pathway enrichment. Core genes were identified via protein–protein interaction networks and functional

Construction and initial validation of key gene network for progesterone resistance in endometrial cancer based on genome-wide CRISPR screening.

Endometrial carcinoma, a prevailing malignancy of the female reproductive system, exhibits escalating incidence and a trend towards early onset. Hormone therapy serves as a primary choice for fertility preservation and is also considered for advanced and recurrent cases. However, a considerable number of patients fail to respond favorably to progestin treatments. We employed CRISPR/Cas9 technology to establish a comprehensive human genome library in the Ishikawa cell line. Subsequent exposure to medroxyprogesterone was followed by high-throughput sequencing, and differential gene expression an

Electroacupuncture may improve endometrial receptivity in controlled ovarian hyperstimulation mice by activating AVP neurons.

BACKGROUND: Controlled ovarian hyperstimulation (COH) can lead to reduced endometrial receptivity during the window of implantation (WOI) and disrupted expression of endometrial clock genes. This study aimed to explore the potential mechanisms by which electroacupuncture (EA) improves endometrial receptivity in COH mice, focusing on the regulation of central and peripheral clock genes within the circadian system. METHODS: Experiment 1 included three groups of mice: control (CTR), COH, and EA. Each group was further subdivided into six subgroups according to sampling time. Tissue samples were

Study on the Role of the AMPK/PGC-1α Pathway in Cold-Induced Vascular Endothelial Cell Apoptosis and Uterine Damage.

Cold exposure may influence reproductive health through vascular changes, yet its mechanisms remain underexplored. This study aimed to investigate the impact of cold exposure on uterine blood vessels and the expression of the AMPK/PGC-1α gene and protein in adult female SD rats. A primary dysmenorrhea model was established in female Sprague Dawley rats and subjected to continuous cold exposure. Changes in body weight, ear temperature, and estrous cycle were observed. Superoxide dismutase (SOD) activity and adenosine triphosphate (ATP) levels were measured to assess oxidative stress. Uterine ti

Atractylenolide III improves ovarian dysfunction and endometrial receptivity in a PCOS rat model by regulating Cybb-mediated oxidative stress.

Polycystic ovary syndrome (PCOS) is an endocrine and metabolic condition affecting reproductive health, marked by disrupted ovulation, excessive androgen levels, and impaired endometrial receptivity. Oxidative stress is an important part of its pathophysiological process. Atractylenolide III (ATIII), a major sesquiterpene lactone derived from Atractylodes macrocephala Koidz., exhibits antioxidant properties; however, its role and molecular mechanisms in PCOS have not yet been explored. A PCOS rat model was established using dehydroepiandrosterone (DHEA) and subsequently treated with ATIII or m

Progesterone resistance in atypical endometrial hyperplasia: Expression and mechanisms of hormone-responsive molecules.

This study aimed to delineate the molecular profile underlying progesterone resistance in atypical endometrial hyperplasia (AEH), a precancerous condition associated with a high risk of malignant transformation. Using a retrospective cohort of 20 AEH patients who completed a 6-month progestin therapy, we compared protein expression between 10 progesterone-resistant and 10 progesterone-sensitive tissues using immunohistochemistry and Western blot analysis. The results revealed a distinct molecular signature in resistant tissues, characterized by significant upregulation of estrogen signaling co

Single-cell transcriptomic analysis of endometrium pre-receptivity and receptivity stages in porcine.

Endometrial receptivity is a key factor influencing embryo implantation and development, and is therefore of significant research importance. In this study, we constructed single-cell transcriptome maps of the porcine endometrium during the pre-receptivity and receptivity phases, and identified the major cell types within the endometrial tissue. It is worth noting that there are significant differences in the composition of epithelial cell subpopulations during the pre-receptivity and receptivity phases. We examined the functional characteristics of endometrial epithelial cells and stromal cel

Single-Cell Transcriptomic Landscape of Cervical Cancer Cell Lines Before and After Chemoradiotherapy.

Cervical cancer remains a significant global health burden, with chemoradioresistance representing a major obstacle to successful treatment. To elucidate the mechanisms underlying this resistance, we established a unique pair of isogenic primary cervical cancer cell lines, AdMer35 and AdMer43, obtained from a patient with squamous cell carcinoma of the cervix before and after radiation therapy. The aim of our study was to characterize the transcriptomic and cellular heterogeneity of these cells. We conducted an in-depth comparative analysis using single-cell RNA sequencing. Analysis of this pa
