Recent Progress in Thiazole, Thiosemicarbazone, and Semicarbazone Derivatives as Antiparasitic Agents Against Trypanosomatids and Plasmodium spp.

Neglected tropical diseases (NTDs), including Chagas disease, human African trypanosomiasis (HAT), leishmaniasis, and malaria, remain a major global health challenge, disproportionately affecting low-income populations. Current therapies for these diseases suffer from significant limitations, such as reduced efficacy, high toxicity, and emerging parasite resistance, highlighting the urgent need for new therapeutic strategies. In response, substantial efforts have been directed toward the synthesis of new molecules with improved potency, selectivity, and pharmacokinetic profiles. However, despi

A cytidine deaminase-like protein modulates pyrimidine nucleotide homeostasis in Trypanosoma brucei.

Pyrimidine nucleotide homeostasis is critical for DNA replication and cell viability, yet its regulation in Trypanosoma brucei, the causative agent of African trypanosomiasis, remains poorly understood. Here, we characterize a T. brucei cytidine deaminase-like protein (TbCDA-like), a kinetoplastid-specific enzyme absent in mammals, that harbors a deaminase domain and a zinc-finger CCCH motif. Using RNA interference (RNAi) and overexpression approaches, we demonstrate that TbCDA-like modulates pyrimidine nucleotide pools, influencing both ribonucleotide and deoxyribonucleotide profiles. Cytological profiling of trypanocidal principles from Aloe barbadensis and Taraxacum officinale.

The use of herbal medicines to treat ailments is a common practice in several regions in Africa, relying on knowledge systems that have evolved over several generations. These herbal remedies are often based on anecdotal claims, many of which lack scientific validation. This study investigates the mode of action of two bioactive fractions, F1 (IC50: 8.5 µg/mL) and F5 (IC50: 7.4 µg/mL), derived from a dichloromethane extract of a herbal mixture, consisting of Aloe barbadensis and Taraxacum officinale, that is commonly used in Ghana to treat parasitic fevers. Both fractions exhibited trypanocida

Modelling motility of Trypanosoma brucei.

African trypanosomiasis, or sleeping sickness, is a life-threatening disease caused by the protozoan parasite Trypanosoma brucei. The bloodstream form of T. brucei has a slender body with a relatively long active flagellum, which makes it an excellent swimmer. We develop a realistic trypanosome model and perform mesoscale hydrodynamic simulations to study the importance of various mechanical characteristics for trypanosome swimming behavior. The membrane of the cell body is represented by an elastic triangulated network, while the attached flagellum consists of four interconnected running-in-p

The "Gredouno" Cross Target: a new tool adapted to control Glossina palpalis gambiensis in the mangrove forests of Guinea.

BACKGROUND: In the mangroves of Guinea, where the most active foci of human African trypanosomiasis in West Africa are located, vector control against tsetse flies using insecticide-impregnated Tiny Targets was first introduced in 2012. While annual deployments of Tiny Targets have resulted in an overall 90% reduction in tsetse fly densities in control areas, managing tsetse densities in specific biotopes such as mangrove channels, which are susceptible to significant climatic disturbances, presents greater challenges. Thus, a new three-dimensional model called the Cross Target was designed to

Antitrypanosomal secondary metabolites from medicinal plants: a review.

Human African trypanosomiasis (HAT) or sleeping sickness is caused by two subspecies of extracellular protozoan parasites, namely, Trypanosoma brucei gambiense and T. brucei rhodesiense. On the other hand, Chagas disease is caused by Trypanosoma cruzi. There are currently six drugs available for the treatment of African sleeping sickness, namely, pentamidine, suramin, melarsoprol, nifurtimox, eflornithine, and fexinidazole. Besides, benznidazole and nifurtimox are drugs that are currently used for the treatment of Chagas disease. Most of the current chemotherapies for the treatment of HAT and

Chemical and Genetic Validation of an Essential Calcium Entry Channel of Trypanosoma brucei as a Therapeutic Target.

The Trypanosoma brucei group of parasites causes Nagana in cattle and human African trypanosomiasis, or sleeping sickness, in humans. Current drugs against these parasites have severe toxicity, vaccines are not available, and development of drug resistance makes finding new chemotherapeutic targets imperative. Ion channels, which are involved in several biological processes, are targets of many therapeutically useful agents, and they remain significantly underexplored as therapeutic targets in parasites. Here, we report the presence of a voltage gated Ca2+ channel (VGCC, TbCav), which is local

Uncovering the Unusual Inhibition Mechanism of a Trypanosome Alternative Oxidase Inhibitor Displaying Broad-Spectrum Activity against African Animal Trypanosomes.

The glucose-dependent respiration of bloodstream forms of the parasite Trypanosoma brucei depends on an unusual and essential mitochondrial electron-transport system, consisting of glycerol-3-phosphate dehydrogenase and the trypanosome alternative oxidase (TAO). We report here the discovery of an allosteric inhibitor of TAO that displays highly potent activity (EC50 values in the range 1-20 nM) against the important veterinary pathogens T. b. brucei, Trypanosoma evansi, Trypanosoma equiperdum, and Trypanosoma congolense, i.e., >5-fold greater potency than the standard drugs. Parasite-resistant variants in African genomes: implications for global genetic medicine.

African populations harbor the highest genetic diversity globally, shaped by over 300 000 years of adaptation to the continent's diverse ecology. Parasitic infections like malaria (Plasmodium falciparum) and sleeping sickness (Trypanosoma brucei) have driven genetic selection for resistance. For instance, the sickle-cell mutation (HBB:c.20A>T) offers malaria protection but causes sickle-cell disease. Similarly, APOL1 gene variants (G1/G2) protect against sleeping sickness but increase the risk of kidney disease. While these variants provide survival benefits, they also pose significant health

Syntheses of benzothiazole amidoximes and arylimidamides and evaluation of their ADME and DNA binding properties and activity against Trypanosoma brucei.

Novel 6-amidoxime- and 6-arylimidamide-substituted benzothiazoles were synthesised to investigate their activity against Trypanosoma brucei, the causative agent of African trypanosomiasis. Benzothiazole amidoxime 12b, with diethylaminoethyl and fluorine substituents on a phenoxymethylene linker exhibited pronounced (IC50 = 0.92 μM) and selective (SI = 18) antitrypanosomal activity. ADME profiling showed that the majority of compounds synthesised are metabolically stable and that arylimidamides have low membrane permeability, while amidoximes, including 12b, have moderate to high permeability.

CryoEM structure of the Trypanosoma brucei flagellum central apparatus.

Axonemes are cylindrical bundles of microtubule filaments that typically follow a 9+n pattern (where n ranges from 0 to 4). However, variations exist across species and cell types, including architectures with fewer (e.g., 3+0, 6+0) or more than nine doublet microtubules (e.g., 9+9+0, 9+9+3), reflecting diverse structural adaptations of cilia and flagella in eukaryotes. Trypanosoma brucei, the causative agent of African trypanosomiasis, relies on its single 9+2 flagellum to navigate through environments within the mammalian host and insect vector. Central to the T. brucei flagellum's function

Towards elimination: Challenges in community participation to a gHAT 'screen and treat' strategy using the new oral drug acoziborole in the Democratic Republic of the Congo.

Until recently, treatment options for gambiense human African trypanosomiasis (gHAT) have been limited and toxic, negatively impacting community participation to screening and treatment. A new, non-toxic, single-dose oral drug has shown efficacy in a Phase III trial and is being tested in a trial called 'STROGHAT', which aims to demonstrate cessation of HAT transmission using a 'screen and treat' strategy. This study aims to explore community perceptions about current and future screening and treatment strategies and identify what could act as barriers to participation in order to prevent them

Identification and initial characterisation of Trypanosoma Cox17 copper metallochaperone.

Copper plays an essential role in organisms as a catalytic co-factor for key enzymes like cytochrome c oxidase. Copper importation, distribution and secretion is carried out by copper transport and copper-binding proteins known as copper chaperones. Cox17 is a chaperone that conveys copper to Cox11 and Sco1 for metalation of the CuB and CuA of Cox1 and Cox2 respectively in eukaryotes. Cox17 from Trypanosoma brucei brucei and T. congolense were recombinantly expressed and affinity purified as MBP-fusion proteins. Chalcogen Derivatives for the Treatment of African Trypanosomiasis: Biological Evaluation of Thio- and Seleno-Semicarbazones and Their Azole Derivatives.

Human African trypanosomiasis (HAT) is caused by Trypanosoma brucei. Drug therapy remains challenging due to drug resistance and/or toxicity. New drugs are needed. Using thiosemicarbazones as a starting point, we employed a S to Se isosteric replacement strategy to design 44 analogs which were evaluated against T. brucei in vitro. Compounds were divided into 11 groups of four derivatives corresponding to thio-, selenosemicarbazones, and their cyclic counterparts, thio- and selenazoles. We selected three groups which contained a total of six derivatives that inhibited parasite growth by >70%. T

Detection of antibodies against the African parasite Trypanosoma brucei using synthetic glycosylphosphatidylinositol oligosaccharide fragments.

Trypanosoma brucei (T. brucei) parasites cause two major infectious diseases in Africa: African trypanosomiasis in humans (HAT) and Nagana in animals. Despite the enormous economic and social impact, vaccines and reliable diagnostic measures are still lacking for these diseases. The main obstacle to developing accurate diagnostic methods and an active vaccine is the parasite’s ability for antigenic variation, impairment of B cell maturation, and loss of B cell memory which collectively prevent the development of a long-lasting, effective immune response. The antigenic variation is sustained by
