Genomic evidence of positive selection highlights functional divergence associated with environmental resilience in central and West African indigenous cattle.

African indigenous cattle harbor rich genetic diversity shaped by long-term adaptation to tropical climates and endemic diseases, notably trypanosomiasis. To investigate the genetic basis of natural disease resistance, we analyzed 33,147 SNPs in 1,047 individuals from 17 breeds using complementary selection scans (iHS, XP-EHH, and Rsb) in a multi-cohort framework. Fifteen pairwise comparisons across six breed groups enabled robust detection of recent positive selection, capturing both within-breed and breed-specific adaptive signals. We identified 229 selective regions, with 47 outlier SNPs co

Antiprotozoal Aminosteroid Alkaloids from Buxus obtusifolia (Mildbr.) Hutch.

Human African Trypanosomiasis (HAT; sleeping sickness) and Malaria are life-threatening protozoan infections in tropical regions, with limited treatment options. As part of our ongoing efforts to discover new aminosteroid alkaloids from the Buxaceae family with antiprotozoal activity, which might serve as leads to new drugs against these infections, we investigated the dichloromethane extract from the leaves of Buxus obtusifolia (Mildbr.) Hutch. collected in Kenya, a species native to Kenya and Tanzania. To the best of our knowledge, and based on the most recent comprehensive literature review

Unexpected Clinical Drug-Drug Interaction of Fexinidazole on Midazolam Pharmacokinetics: Insights into Underlying Mechanisms from Clinical Phase I Study Results and Supporting In Vivo/Vitro Evidence.

BACKGROUND: Fexinidazole, a nitroimidazole antiparasitic, has been approved to treat human African trypanosomiasis (HAT) worldwide. In vitro studies have shown that fexinidazole inhibits and weakly induces CYP3A4/5. In silico predictions indicated that fexinidazole, which has a significant intestinal and liver first-pass metabolism, could increase the exposure of a sensitive probe substrate of CYP3A4 by two-fold. Therefore, this study investigated the potential clinical drug-drug interactions (DDIs) of fexinidazole with CYP3A4 substrates. OBJECTIVE: To assess the effect of fexinidazole on the

Combined toxicity of polyethylene micro/nanoplastics and PFOA in zebrafish (Danio rerio): Impacts on antioxidant, neurotransmission, and gut microbiota.

Polyethylene micro/nano plastics (PE-MPs, PE-NPs), being the predominant plastic pollutants in aquaculture environments, frequently coexist with emerging contaminants such as perfluorinated compounds (PFOA) within closed aquaculture systems. Nevertheless, the combined biotoxicological impacts and underlying mechanisms of PE-MPs, PE-NPs, and PFOA co-pollutants remain poorly understood in zebrafish. This study revealed induced oxidative stress in zebrafish, as evidenced by aberrant levels of antioxidant enzymes (CAT, SOD, GST) and lipid peroxidation markers (LPO, GSH) as well as neurotoxicity (A

Outbreak of Trypanosoma vivax in Dairy Cows: Hematologic, Immunological and Antioxidant Responses Before and After Treatment with Isometamidium Chloride.

Trypanosoma vivax infection is an emerging condition that causes damage and mortality among cattle and is transmitted by mechanical vectors or contaminated fomites. This disease has been spreading in southern Brazil, causing anemia, weight loss, diarrhea, abortion, and infertility; however, its behavior and host-parasite relationships are not yet fully understood. To clarify this issue, animals that presented clinical signs were subjected to an immunochromatographic screening test. An indirect immunofluorescence test was then performed on samples collected before treatment (the gold standard t

Optimization of Physicochemical Properties and ADME for 2,4-Substituted 1H-Pyrrolo[2,3b]pyridines Inhibitors of Trypanosome Proliferation.

Neglected tropical diseases, such as human African trypanosomiasis (HAT), require novel treatments that can populate the clinical pipeline in the event of rising resistance and treatment recrudescence. Following a high-throughput screen of almost 42,000 human kinase inhibitor chemotypes, we identified a 2,4-disubstituted azaindole that had good antiparasitic activity, selectivity, and predicted brain exposure, but that failed to meet our criteria for advancement into an efficacy study. Following hit-to-lead optimization, we arrived at 18a, which offered the best combination of potency, propert

Species distribution modeling to predict tsetse fly (Glossina spp.) habitat suitability in Kenya.

BACKGROUND: African animal trypanosomosis (AAT) and human African trypanosomosis (HAT) are transmitted and spread primarily by tsetse flies (Glossina spp.) in sub-Saharan Africa. The animal disease poses significant challenges to agropastoral systems, including in Kenya, where 38 out of 47 counties are infested with eight species of Glossina. Climate change and human activities can also aggravate these infestations, putting rural-scale farmers who rely on agropastoral systems at a greater risk. Geographical gaps in existing entomological datasets limit a comprehensive understanding of tsetse f

N-Myristoyltransferase Inhibition in Parasitic Pathogens: Insights from Computer-Aided Drug Design.

Neglected tropical diseases (NTDs) constitute a group of infectious diseases that severely affect the health of impoverished populations, and the health, economies, and health systems of affected countries. Leishmaniasis and human African trypanosomiasis (HAT) are particularly notable, and malaria, despite not being neglected, is part of the "big three" (HIV, tuberculosis, and malaria) with high incidence, increasing the probability of infection by NTDs. Therefore, efforts are ongoing in the search for new drugs targeting the enzyme N-myristoyltransferase (NMT), a potential drug target that ha

Passive surveillance of human African trypanosomiasis in the Democratic Republic of the Congo: clinical presentation and prospective evaluation of rapid diagnostic and reference laboratory test accuracy.

BACKGROUND: Passive screening of gambiense human African trypanosomiasis (HAT) is based on rapid diagnostic tests (RDT), but sensitivity of the currently commercialised RDTs has hardly been assessed prospectively. In view of the increasing importance of remote testing for HAT, the diagnostic performance of reference laboratory tests also needs further documentation. METHODOLOGY/PRINCIPAL FINDINGS: The study is registered in ClinicalTrials.Gov under identifier NCT03356665. Clinical suspects in 29 health facilities in DR Congo were screened consecutively between October 2017 and December 2020 w

Development of chimeric multivalent proteins for serological diagnosis of African animal trypanosomosis.

The control of African animal trypanosomosis (AAT) relies on accurate diagnostic tools. Serological diagnosis using ELISA is well-suited for surveillance due to its high-throughput capacity, low cost, and adaptability to rapid formats. However, the WOAH-recommended antibody ELISA for AAT, based on trypanosome lysates purified from rodent blood, can lack specificity and presents standardization challenges as well as ethical concerns. Recombinant proteins offer a solution to standardization, often improving specificity, though potentially at the expense of sensitivity. Combining multiple recombi

DNA Vaccine Encoding Trypanosoma brucei MSP-B Elicited IgG and IFN-γ Responses and Partial Protection in Immunised Mice.

As an effort towards vaccine development against African trypanosomiasis, we studied key parasite molecules that mediate VSG functions, specifically the major surface protease-B of Trypanosoma brucei that catalyses proteolytic removal of old VSGs for expression of new ones, an important stage-specific function that allows the parasite to survive in its host, thus making it an attractive candidate for vaccine development. Herein, the Tbmsp-b gene was cloned into a pVAX-1 plasmid to produce the pVAX-1-Tbmsp-b construct for DNA vaccine trials. BALB/c mice were immunised by intradermal injection w

Cure of experimental Trypanosoma vivax infection with a single dose of an unmodified antibody-based drug targeting the invariant flagellum cell surface protein IFX.

Animal African trypanosomiasis (AAT) is an infectious wasting disease of economically important livestock caused by Trypanosoma spp. parasites. The disease is primarily caused by two species: Trypanosoma congolense and Trypanosoma vivax, which are endemic in many African countries. AAT is managed by therapeutic and prophylactic drugs; however, resistance is now widely reported, and the development of new drugs has been impeded due to a chronic lack of investment. Recently, we identified an invariant flagellar-associated cell surface protein (IFX) that could elicit protective immune responses w

Fexinidazole results in specific effects on DNA synthesis and DNA damage in the African trypanosome.

Nitroaromatic drugs are of critical importance for the treatment of trypanosome infections in Africa and the Americas. Fexinidazole recently joined benznidazole and nifurtimox in this family when it was approved as the first oral monotherapy against Human African trypanosomiasis (HAT). Nitroaromatic prodrugs are bioactivated by the trypanosome-specific type I nitroreductase (NTR) that renders the compounds trypanocidal. The trypanocidal activity of nitroaromatic drugs has been proposed to arise from the formation of reactive oxygen species (ROS) and DNA damage. However, the specific cytotoxic

Development of a camelid single-domain antibody-based antigen detection assay for the pan-specific diagnosis of active human and animal Trypanosoma brucei infections.

Trypanosoma brucei infections cause African trypanosomiasis, also known as sleeping sickness in humans and nagana in animals, presenting a significant global health and economic burden, especially in sub-Saharan Africa. Animal trypanosomiasis also affects the economic development in Asia and South America. Accurate diagnosis of diseases caused by infection with the parasite of the T. brucei group remains a major challenge due to the persistence of infection-induced antibodies long after parasite clearance, complicating serological discrimination between active and past infections. PI3Kdelta-driven expansion of regulatory B cells impairs protective immune responses to Trypanosoma congolense parasite infection.

Phosphatidylinositol 3-kinase delta (PI3KCD) is a critical signaling enzyme for B cell development, activation, function and immune regulation. Gain-of-function mutations in PI3KCD result in the congenital immunodeficiency known as Activated PI3KCD Syndrome (APDS). APDS patients are prone to repeated infections and other serious clinical manifestations. Here, we determine how B cell-intrinsic expression of the APDS-associated PI3KCDE1021K mutation impacts immune responses to the protozoan parasite Trypanosoma congolense. PI3KCDE1021K/B mice exhibit a significant expansion of IL10-expressing B
