C1A cysteine peptidases (CPs) are major contributors to host-parasite crosstalk and have been proposed as anti-trypanosomal chemotherapeutic targets. Trypanosoma evansi is the most widespread animal trypanosome of African origin causing the trypanosomosis known as "Surra" or "derrengadera". Gelatin zymography combined with peptidase class-specific inhibitors identified four CPs in T. evansi lysates with relative masses of 28, 31, 36 and 40 kDa (p28, p31, p36 and p40). The CPs present in the T. evansi lysate preferentially hydrolyzed the Z-Phe-Arg-AMC fluorogenic substrate over Z-Gly-Arg-AMC an

According to the World Health Organization vector-borne diseases account for more than 17% of all infectious diseases, causing more than 700,000 deaths annually. Vectors are organisms that are able to transmit infectious pathogens between humans, or from animals to humans. Many of these vectors are hematophagous insects, which ingest the pathogen from an infected host during a blood meal, and later transmit it into a new host. Malaria, dengue, African trypanosomiasis, yellow fever, leishmaniasis, Chagas disease, and many others are examples of diseases transmitted by insects. Both the diet and

BACKGROUND: Human African trypanosomiasis (HAT) is a fatal vector-borne disease caused by Trypanosoma brucei. Although HAT incidence has declined, meeting WHO's elimination targets remain difficult, particularly due to limited diagnostic sensitivity for low-parasite load-infections. Arboviruses such as dengue (DENV 1-4), chikungunya (CHIKV), and yellow fever (YFV) virus, present with nonspecific febrile symptoms similar to HAT and are underdiagnosed in Sub-Saharan Africa. Due to this overlap in symptoms and a suspected geographical overlap of vectors and pathogens in the Democratic Republic of

BACKGROUND: We aimed to establish an external quality assessment (EQA) programme for the yaws eradication campaign that would meet the needs of reference and district-level laboratories in low- and middle-income countries.
METHODOLOGY/PRINCIPAL FINDINGS: We designed proficiency testing items (PTIs) using a plasmid containing gene target sequences for Treponema pallidum (TP) and Haemophilus ducreyi (HD). The storage stability of the plasmids under different environmental conditions was then tested. A proficiency testing panel of seven swabs loaded with different concentrations of plasmids in d

Cystic fibrosis (CF) lung disease is characterized by chronic endobronchial infection resulting in progressive pulmonary destruction; this is a major cause of mortality and morbidity. Neutrophils are the primary effector cells responsible for the progressive deterioration of lung function. Peptido-leukotriene B4 antagonists, new anti-inflammatory agents that block the neutrophil-dominated inflammation, could have had the potential for long-term use. A trial on the pharmacokinetics of amelubant administered orally as a single dose of up to 75 mg in pediatric patients with CF and 300 mg in adult

Human African trypanosomiasis, Chagas disease, and leishmaniasis are neglected, debilitating diseases caused by protozoa of the Trypanosomatidae family. In search of new effective therapies, subtle modifications of purine nucleosides have previously afforded agents that demonstrate promising activity against these protozoa, also in vivo. Here, a library of imidazo[2,1-f][1,2,4]triazine ribosides with varying substituents at C6 (trivial purine numbering) was synthesized through late-stage diversification of a common propyl thioether precursor and screened against a panel of the aforementioned T

Malaria coinfections with other infectious agents represent a clinically relevant epidemiological challenge, but evidence remains fragmented across individual systematic reviews. This umbrella systematic review synthesized review-level evidence on the prevalence and epidemiological patterns of concurrent malaria coinfections in human populations. PubMed/MEDLINE, Scopus, Web of Science, Embase, and LILACS/BVS were searched for systematic reviews published from 1 January 2000 to March 2026. Risk of bias, certainty of evidence, and primary-study overlap were assessed using ROBIS, an adapted GRADE

Cardiogenic shock following an acute coronary syndrome (ACS) continues to be associated with significant mortality despite modern reperfusion strategies and inotropic support. There is mounting evidence that an acute inflammatory response accompanies the well documented decrement in left ventricular systolic function associated with cardiogenic shock and that this response may affect outcomes. In the past 2 decades it has also become apparent that nitric oxide (NO), a heteroatomic free radical has numerous biologic activities, among them the maintenance of vascular tone. The production of NO i

BACKGROUND: Understanding synapse-specific effects of neuroinflammation can provide mechanistic and therapeutically relevant insights across the spectrum of neurological diseases.
METHODS: We applied neuron-specific proteomic biotinylation in vivo, differential centrifugation of brain for crude synaptosome enrichment (P2 fraction) and mass spectrometry (MS) analysis of biotinylated proteins to derive native-state proteomes of Camk2a-positive neurons and their corresponding P2 synaptic compartments. Next, in an in vivo model of systemic lipopolysaccharide (LPS) dosing, we examined the effects

BACKGROUND: Human African trypanosomiasis (HAT) is an endemic neglected tropical disease in Sub-Saharan Africa targeted for elimination by 2030. However, asymptomatic human infections may sustain cryptic transmission. Nigeria has not reported confirmed HAT cases in over a decade, but emerging evidence suggests ongoing silent transmission. Infection during pregnancy raises additional concerns, including adverse outcomes and vertical transmission. This study therefore investigated the presence of trypanosome infection in pregnant women attending the antenatal clinic in Enugu, Nigeria.
METHODS:

The aim of this study was to assess the possibility of deep learning-based object detection models for the early and comprehensive detection of animal trypanosomosis. We constructed deep learning models for the early detection of Trypanosoma parasites including Trypanosoma congolense from in vitro and in vivo thin blood smears. Our models were based on YOLO (You Look at Once), one of the most common and high-performance object detection models. The method was applied to 14, 380 thin blood smear images with 47, 276 parasites (cells). For the T. congolense, the possibility for the early detectio

African trypanosomes are a group of pathogens that cause disease in humans (human African trypanosomiasis; HAT) and animals (African animal trypanosomosis; AAT) across sub-Saharan Africa, leading to profound impacts on human health as well as economic instability. Achieving sustainable disease control requires a One Health approach, yet the development of urgently required pan-species chemotherapeutics is severely hindered by a lack of unified biological understanding of the causative species. A major bottleneck in drug discovery is the distinct in vitro requirements of the clinically relevant

Microalgal food supplements are becoming increasingly popular due to their promising biological effects and high nutritional value, evidenced in in vitro, in vivo, and human studies. Some products of this kind have, however, raised controversies concerning their safety. At the same time, not much is known about the frequency of adverse events following the use of microalgal supplements, potential factors that may influence them, and general characteristics and behaviours of the consumer group. The present study aimed to fill this gap and surveyed a group of Polish consumers of microalgal produ

Neglected tropical diseases (NTDs) are a group of infectious diseases that mainly affect the population living in poverty and without basic sanitation, causing severe damage to countries' economies. Among them, Leishmaniasis, Chagas disease, sleeping sickness, and related diseases such as Malaria stand out, which, despite being well known, have limited treatments based on old drugs and have high rates of parasite resistance. In addition, current drugs have an uncertain mechanism of action, and there is a need to identify new mechanisms to overcome problems related to side effects and resistanc

BACKGROUND AND OBJECTIVE: This study focuses on the potential impact of genetic screening technologies on healthcare. Genetic screening for asthma in children was chosen as a case study to explore the cost effectiveness of applying early genetic screening to infants, and preventive treatment to the population at risk. Early intervention could prevent progression and facilitate clinical management of the disease. From the elite group of genetic markers that have been associated with asthma-related phenotypes, ADAM33 was the first published candidate gene detected by a positional cloning approac

BACKGROUND: Neglected tropical diseases (NTDs) are parasitic and bacterial diseases that affect approximately 149 countries, mainly the poor population without basic sanitation. Among these, Human African Trypanosomiasis (HAT), known as sleeping sickness, shows alarming data, with treatment based on suramin and pentamidine in the initial phase and melarsoprol and eflornithine in the chronic phase. Thus, to discover new drugs, several studies point to rhodesain as a promising drug target due to the function of protein degradation and intracellular transport of proteins between the insect and ho

Synthetic drugs currently prescribed for the treatment of Human African Trypanosomiasis (HAT) are non-specific, toxic, demand extended therapeutic regimes and are of varying efficacy. Along with the challenging demographic and socio-economic hurdles, the everincreasing risk of drug resistance is another major problem to be addressed. Cysteine protease, Heat shock proteins (HSP-90), Trypanothione reductase (TR), Farnesyl diphosphate synthase, Glucose-6-phosphate dehydrogenase, UP-4-galactose epimerase, and Cytidine triphosphate synthetase are potential enzymatic targets for the development of n

SUMMARYPrior to 2019, when the orally available drug fexinidazole began its clinical use, the treatment of human African trypanosomiasis (HAT) was complex and unsatisfactory for many reasons. Two sub-species of the Trypanosoma brucei parasite are responsible for HAT, namely the rhodesiense form found in East and Southern Africa and the gambiense form found in Central and West Africa. Diseases caused by both forms manifest in two stages: stage 1 before and stage 2 after central nervous system involvement. Prior to 2019, different drugs were required for each of the two parasite sub-species at e

BACKGROUND: Leishmaniasis, Chagas disease (CD), and Human African Trypanosomiasis (HAT) are neglected tropical diseases in humans caused by intracellular parasites from the class Kinetoplastida. Leishmaniasis is one infectious disease that exhibits sex-bias not explained solely by behavioral or cultural differences. However, HAT and CD have less well documented and understood sex-related differences, either due to a lack of differences or insufficient research and reporting.
METHODS: This paper reviews the rate of disease and disease severity among male and females infected with CD, HAT, and

UNLABELLED: Trypanosoma brucei is a protozoan parasite that causes human and animal African trypanosomiases (HAT and AAT). Cardiac symptoms are commonly reported in HAT patients, and intracardiac parasites with accompanying myocarditis have been observed in both natural hosts and animal models of T. brucei infection. Despite the importance of T. brucei as a cause of cardiac dysfunction and the dramatic socioeconomic impact of African trypanosomiases in sub-Saharan Africa, there are currently no reproducible murine models of T. brucei-associated cardiomyopathy. We present the first clinically r
