Conflicts within the tsetse fly belt revealed a strong correlation between the dynamics of bovine trypanosomosis and the insurgency involving farmers and herders in Nigeria and parts of West Africa. This study examined the history, causes and influence of farmers-herdsmen conflicts on banditry, terrorism and food security as it relates to the epidemiology of African animal trypanosomosis (AAT). A combination of literature database searches, semi-structured questionnaires, and mathematical modeling was employed. The study found that transhumance contributes significantly to conflicts between fa

Surra and Dourine are widespread diseases caused by two protozoan parasites Trypanosoma brucei evansi and Trypanosoma brucei equiperdum, respectively. A wide range of animals including camels, horses, cattle and buffaloes are susceptible to infection. These diseases pose a significant socio-economic burden, primarily due to the limited therapeutic options and the complications associated with toxicity and drug resistance, making disease management particularly challenging. This study evaluated the potential of 3'-deoxytubercidin, a previously identified antitrypanosomal nucleoside, as a therap

African animal trypanosomosis (AAT) in cattle is primarily managed through trypanocide administration and insecticide application. Trypanocides can be used for both treatment and prophylaxis, but failure is often reported; this may occur due to resistance, substandard drugs, or inappropriate administration. This study in Tanzania aims to quantify reasons for trypanocide failure. An observational year-long longitudinal study was conducted in high-risk AAT areas in Serengeti District between June 2021-October 2022. Purposive sampling targeted herds with high utilization of the prophylactic trypa

Human African trypanosomiasis (HAT) is one of the most lethal of the neglected tropical diseases. While the discovery of a novel antitrypanosomal drug is highly desired, the creation of a superior lead compound is challenging. Herein we report ukabamide (1), which was isolated from a marine Moorena sp. cyanobacterium, as a selective and potent antitrypanosomal lipopeptide (IC50 = 34 ± 18 nM against Trypanosoma brucei rhodesiense). Its structure was elucidated by spectroscopic analyses and degradation reactions and confirmed by total synthesis. In addition, we prepared two modified analogs and

BACKGROUND: African animal trypanosomosis (AAT) is one of the most serious diseases with ongoing detrimental effects on animal health and food production.
METHODS: A cross-sectional study was conducted in the Bedele and Dedesa districts of Buno Bedele Zone, Southwest Ethiopia, to determine the prevalence of trypanosomosis and its vector distributions in small ruminants. Blood samples collected from a total of 384 small ruminants were examined for trypanosomosis via hematological analysis. For the tsetse distribution survey, 72 traps were deployed in the designated districts.
RESULTS: Among t

Canine African trypanosomosis is endemic in sub-Saharan Africa. Chemotherapy remains the commonly employed approach to trypanosomosis control. However, it is beleaguered by the absence of new drugs, treatment failures, relapse infection and resistance. The efficacy of a high dose of isometamidium chloride (ISM) in single and mixed infections of T. congolense and T. brucei brucei therapy was assessed in dogs. Fifteen dogs employed in this study were allocated into four groups at random, each with four dogs except group I which had three dogs. Group I dogs were not infected while groups II and I

OBJECTIVES: Zimbabwe is endemic for lymphatic filariasis (LF) with 39 districts identified for mass drug administration (MDA) in 2014. The objective of this study was to reassess the current population requiring MDA.
METHODS: The LF confirmatory mapping method was used to evaluate the prevalence of circulating filarial antigens among school-aged children (9-14 years) in previously endemic districts. This was achieved through a cross-sectional survey between 2021 and 2023, applying a cluster sampling approach across 39 districts, targeting 18,720 children to assess ongoing LF transmission.

To regain infectivity, Trypanosoma brucei, the pathogen causing Human and Animal African trypanosomiasis, undergoes a complex developmental program within the tsetse fly known as metacyclogenesis. RNA-binding protein 6 (RBP6) is a potent orchestrator of this process, however, an understanding of its functionally important domains and their mutational constraints is lacking. Here, we perform deep mutational scanning of the entire RBP6 primary structure. Expression of libraries containing all single-point variants of RBP6 in non-infectious procyclic forms and subsequent purification of infectiou

Tsetse flies (Glossina spp.) vector African trypanosomes that cause devastating diseases in humans and domestic animals. Within the Glossina genus, species in the Palpalis subgroup exhibit greater resistance to trypanosome infections compared to those in the Morsitans subgroup. Varying microbiota composition and species-specific genetic traits can significantly influence the efficiency of parasite transmission. Notably, infections with the endosymbiotic bacterium Spiroplasma have been documented in several Palpalis subgroup species, including Glossina fuscipes fuscipes (Gff). While Spiroplasma

OBJECTIVES: Sleeping sickness, also known as human African trypanosomiasis (HAT), is a tsetse fly-borne neglected tropical disease that affects underserved rural communities in sub-Saharan Africa. Despite the advancements in diagnostics, the actual status of sleeping sickness in Nigeria remains unclear. In our quest for clarity, we conducted a human population survey to ascertain the prevalence of HAT in tsetse fly-infested remote rural hamlets.
METHODS: Blood samples were collected from 72 consenting participants by finger pricking. Blood samples were blotted on a Flinders Technology Associa

Tropical parasitic diseases affect millions of people around the world, particularly in poor countries. The human parasitic diseases that will be covered in this review are malaria and neglected diseases, such as leishmaniasis, Chagas disease, and African trypanosomiasis. The current treatments for these diseases present several problems, such as the development of drug resistance, very limited drugs available in the clinic, significant side effects of the drugs, and a long treatment period. For these reasons, there is an urgent need to develop new chemotherapeutics to eradicate or eliminate t

Bovine trypanosomosis remains a significant challenge in Nigeria due to the widespread presence of tsetse and biting flies. Despite numerous control interventions, livestock owners continue to face the burden of high treatment costs year-round. Holistic management has been elusive, largely due to the interconnected roles of the "Big-Six" animal hosts in sustaining transmission dynamics. This study conducted a systematic review of publications from Web of Science, Ovid MEDLINE, PubMed, Google Scholar, and AJOL Online databases, following PRISMA guidelines between 2000 and 2024. Neglected tropical diseases such as Chagas disease, human African trypanosomiasis, leishmaniasis, and schistosomiasis have a significant global health impact in predominantly developing countries, although these diseases are spreading due to increased international travel and population migration. Drug repurposing with a focus on increasing antiparasitic potency and drug-like properties is a cost-effective and efficient route to the development of new therapies. Here we identify compounds that have potent activity against Trypanosoma cruzi and Leishmania donovani, and the latter were progresse

BACKGROUND: The chronic form of human African trypanosomiasis (HAT) caused by Trypanosoma brucei gambiense and commonly referred to as gambiense-HAT (gHAT) is endemic in 7 of Angola's 18 provinces. Major epidemics of the disease occurred in the country in the 1920s to 1940s and 1990s -mid 2000s, and current estimates are that up to a third of the country's population is at risk of infection. Whereas gHAT was first reported in Angola in 1871, control efforts did not begin until 30 years later in 1901. This case study describes the history of gHAT in Angola, outlines the policies and strategies

The protozoan parasites Trypanosoma brucei and Trypanosoma cruzi, which cause human African trypanosomiasis (HAT) and Chagas disease, respectively, are responsible for considerable human suffering. Reduced case numbers and improved treatment options for HAT provide hope, but the outlook for Chagas disease is less promising, and safer, more efficacious chemotherapy is sorely needed. We previously reported the discovery and optimisation of a novel class of potent and selective trypanosomacidal 2-[(2-phenylthiazolyl)ethyl]ureas active against both T. brucei brucei and T. cruzi. In the current wor

In the mammalian host, Trypanosoma congolense cytoadheres, or sequesters, to the vascular endothelium. Although sequestration influences clinical outcome, disease severity and organ pathology, its determinants and mediators remain unknown. Challenges such as the variability of animal models, the only-recently developed tools to genetically manipulate the parasite, and the lack of physiologically-relevant in vitro models have hindered progress. Here, we engineered brain and cardiac 3D bovine endothelial microvessel models that mimic the bovine brain microvasculature and the bovine aorta, respec

Elimination of gambiense human African trypanosomiasis (gHAT) as a public health problem has been reached or is in sight in a number of endemic foci and the next step is now to reach the elimination of transmission. The ability to detect Trypanosomabruceigambiense (T.b.gambiense) in both the last human cases and in a suspected animal reservoir becomes increasingly important to reach this goal. We have evaluated here the diagnostic performance of the AnTat A/B and LiTat A/B primers in comparison with the TBR, TgsGP and nested TgsGP PCRs that are currently used for the molecular diagnosis of gHA

Extreme weather events can lead to infectious disease outbreaks, especially those spread by hematophagous flies, and The Gambia is particularly vulnerable to climate change. To the best of our knowledge, no one has ever documented the relationship between climate variability and change and the distribution of the hematophagous flies belonging to the families Glossinidae, Tabanidae, and Stomoxyinae. This paper aims to study the association of temperature and humidity on the distribution of the above species and their families in The Gambia in the recent past and to provide predictions of specie

Background/Objectives: Many tropical diseases such as malaria, Chagas, human African Trypanosomiasis, and Lymphatic filariasis coexist in endemic countries, affecting more than 1 billion people worldwide, and are recognised as major global vector-borne diseases. Tackling this disease requires an accurate diagnosis that is sensitive, specific, and rapid. This study aimed to describe and validate a new highly sensitive and specific multiple-analysis system that can effectively detect numerous etiological agents in a single test. Methods: A total of 230 human blood samples were assessed retrospec

Trypanosoma brucei rhodesiense (Tbr) and Plasmodium falciparum (Pf) are protozoan parasites that cause severe diseases, namely, Human African Trypanosomiasis (HAT) and Malaria. Due to limited treatment options, there is an urgent need for new antiprotozoal drugs. Pachysandra terminalis (P. terminalis), a plant belonging to the family Buxaceae, is known as a rich source of aminosteroid alkaloids, and a previous study of our working group already showed that the alkaloid-enriched fraction of P. terminalis aerial parts showed promising activity against protozoan parasites. In the present study, t
