RUBACE profile of pediatric patients with prune belly syndrome undergoing renal transplantation: Findings from a case series.

INTRODUCTION: Prune Belly Syndrome extends beyond the classical triad of deficient abdominal wall musculature, urinary tract anomalies, and cryptorchidism. Urological abnormalities may be severe enough to require renal transplantation during their lifetime. The RUBACE score is currently the most comprehensive tool for categorising the multisystem manifestations of the syndrome. We hypothesized that higher RUBACE scores would be associated with poorer post renal transplantation outcomes. PATIENTS AND METHODS: We conducted a retrospective review of 500 paediatric renal transplantations performe

Clinical and molecular characterization of 5α-reductase deficiency type 2: First report of a family with homozygous deletion in exon 1 of SRD5A2.

OBJECTIVES: 5α-reductase deficiency type 2 (5αRD2) is the second most common cause of 46, XY disorders of sex development. The 5α-reductase type 2 isozyme, which is encoded by the SRD5A2 gene, catalyzes the conversion of testosterone (T) to dihydrotestosterone (DHT). This study aimed to evaluate the clinical, laboratory, and molecular characteristics of patients with 5αRD2. METHODS: We retrospectively analyzed the clinical, laboratory, and genetic features of thirteen patients diagnosed with 5αRD2. Age, gender assignment, presenting features, hormonal profiles, and SRD5A2 variants were evalua

Clinical, hormonal, and genetic spectrum of cryptorchidism: a comprehensive evaluation of patients referred for pediatric endocrinological assessment.

OBJECTIVES: Cryptorchidism is the most prevalent pediatric genital anomaly, yet its clinical significance often extends beyond simple anatomical maldescent. While surgical management is well-established, the necessity for holistic endocrine and genetic evaluation remains underexplored. We aimed to characterize the clinical, hormonal, and genetic landscapes of patients referred for pediatric endocrinological assessment following orchiopexy. METHODS: We retrospectively analyzed 99 boys referred for postoperative evaluation. Clinical findings, basal and dynamic hormonal profiles, and genetic res

Management of undescended testicle in patients with prune belly syndrome: 30 years of experience in the French cohort - A multicentric study.

BACKGROUND: Prune Belly Syndrome (PBS) is a rare congenital disorder characterized by bilateral intraabdominal undescended testes, abdominal wall flaccidity, and urinary tract dilation. Surgical management of cryptorchidism in PBS is technically challenging due to anatomical and urological anomalies. This study aimed to evaluate outcomes of different orchidopexy techniques, laparoscopic vs. open and one-stage vs. two-stage, in a national multicenter PBS cohort over 30 years. METHODS: We retrospectively reviewed 56 PBS patients treated between 1990 and 2023 in 22 French centers using data from

Spectrum of Congenital Anomalies in Myhre Syndrome-Insights Into Effects Brought by Altered TGF-β Signaling via Gain-of-Function Variants in SMAD4.

Myhre syndrome is a rare genetic disorder characterized by progressive multisystem involvement. Gain-of-function missense heterozygous variants affecting the Ile500 residue and Arg496 residue of the SMAD4 gene are implicated in this condition. In this article, we aim to understand the spectrum of congenital anomalies in Myhre syndrome by studying a cohort of previously unreported patients alongside published literature. Our analysis revealed that the musculoskeletal system was the most common system to be affected, followed by the cardiovascular system. Intrauterine growth restriction was the

A lethal form of ASCC3 disease: Severe global developmental delay, axial hypotonia, hypoplasia of corpus callosum, hypothyroidism, and micropenis.

Activating Signal Cointegrator 1 Complex, Subunit 3 (ASCC3) encodes a DNA helicase that is pivotal in transcriptional regulation, DNA damage response, and ribosomal quality control. Bi-allelic ASCC3 variants have been recently associated with a wide spectrum of non-lethal neuromuscular and neurodevelopmental disorders, with no consistent brain malformations or endocrine dysfunction. We describe a severe lethal form of ASCC3 disease: eight individuals of three consanguineous Bedouin families presented with a consistent phenotype of severe global developmental delay, axial hypotonia, and hypopla

From incidentaloma to actionable insight: a clinical-molecular-imaging framework for risk-stratified management of testicular microlithiasis.

OBJECTIVE: Testicular microlithiasis (TM) is common in infertile men, but its management is controversial due to an unclear link to testicular germ cell tumor (TGCT) risk. This scoping review synthesizes evidence to clarify the basis for personalized management of TM, focusing on infertile men. METHODS: Following PRISMA-ScR guidelines, we systematically searched PubMed, Embase, and Web of Science (2015-2025) for studies on TM pathogenesis, imaging, molecular mechanisms, management, and TGCT association. Data were charted and narratively synthesized. RESULTS: TM and TGCT share molecular pathw

Pediatric andrology: bridging pediatric urology and reproductive medicine.

PURPOSE OF REVIEW: Andrological disorders, including male infertility, hypogonadism, and sexual dysfunction, are often managed in adulthood despite their origins in early life. This review proposes the emerging concept of 'pediatric andrology', which integrates pediatric urology, endocrinology, and reproductive medicine, and aims to highlight its clinical significance from a life-course perspective. RECENT FINDINGS: Growing evidence suggests that common andrological conditions such as varicocele, cryptorchidism, and Klinefelter syndrome originate in childhood and have long-term implications f
