Environmental chemicals in dogs: evidence review and new data linking dietary exposure and reproductive performance.

The domestic dog is proposed as a sentinel species for the adverse reproductive effects of environmental chemicals (ECs), sharing the human environment and some components of dietary exposure, whilst exhibiting similar reproductive changes as humans. Here we present a narrative review of published information incorporating new data that potentially link EC exposure to changes in reproductive function in the dog. We compare these findings with similar trends in humans and other species. Our conclusions include; (1) within a single UK assistance-dog breeding population, a decline in sperm motili

Clinical characteristics and genotype-phenotype correlation analysis in 279 patients with 5α-reductase type 2 deficiency.

OBJECTIVE: 5α-Reductase type 2 deficiency (5α-RD2) is an autosomal recessive differences/disorders of sex development caused by SRD5A2 gene mutations, characterized by impaired testosterone-to-dihydrotestosterone conversion and highly heterogeneous clinical phenotypes. The genotype-phenotype correlation of 5α-RD2 remains incompletely understood, and large-scale multicenter evidence is lacking for clinical management. METHODS: We systematically searched PubMed, Embase, CNKI, Wanfang, and Chinese Medical Journals databases from inception to December 2025. A total of 279 genetically confirmed pa

[A CASE OF RIGHT TESTICULAR SEMINOMA WITH REGRESSIVE CHANGES PRESENTING AS INGUINAL LYMPH NODE METASTASIS].

A 32-year-old male presented with a growing right inguinal mass. He had a history of right orchiopexy for cryptorchidism at age 8. Radiologic imaging, including CT and MRI, suggested a malignant soft tissue tumor. Needle biopsy confirmed seminoma, leading to referral to our department. Physical examination revealed a 12 cm tender mass in the right inguinal region. The right testis was atrophic, with no palpable intratesticular mass. Ultrasonography showed diffuse microcalcifications within the right testis parenchyma, with a small space-occupying lesion identified within these areas. FDG-PET/C

Comparative Analysis of Cytoskeletal Proteins and the Sarcoglycan Complex in the Cremaster Muscle of Patients with Retractile and Undescended Testis.

INTRODUCTION: The cremaster muscle plays a crucial role in testicular mobility and thermoregulation. Structural and functional alterations of this muscle have been implicated in the pathophysiology of undescended testis. Previous studies have demonstrated a significant impairment of the cytoskeleton in cremasteric smooth muscle cells in patients with cryptorchidism. This alteration may explain the muscular dysfunction involved in testicular descent. The aim of this study was to assess and quantify the expression of sarcoglycans, as well as the anchoring proteins vinculin and talin, in the crem

Case Report: Minipuberty hormonal profile in PPP1R12A-related persistent Müllerian duct syndrome.

Persistent Müllerian duct syndrome (PMDS) is a rare difference/disorder of sex development and maturation characterized by the retention of Müllerian structures during fetal development in individuals with a 46,XY karyotype. It is typically due to mutations in anti-Müllerian hormone (AMH) or its receptor, anti-Müllerian hormone receptor type 2 (AMHR2). Here, we report a novel de novo heterozygous loss-of-function frameshift variant in protein phosphatase 1 regulatory subunit 12A (PPP1R12A; p.Lys197IlefsTer13) identified in a 46,XY neonate presenting with bilateral cryptorchidism and a Mülleria

Testicular self-examination in adolescents following childhood orchiopexy for cryptorchidism.

INTRODUCTION: Cryptorchidism is associated with an increased lifetime risk of testicular malignancy. Although orchiopexy reduces this risk and facilitates examination, postpubertal vigilance remains important. We assessed adherence to monthly testicular self-examination (TSE) among adolescents and young men who underwent childhood orchiopexy. METHODS: We conducted a retrospective cohort study with a cross-sectional telephone questionnaire component. Patients who underwent orchiopexy for cryptorchidism at a tertiary children's hospital between 2010 and 2020 and were at least 13 years old at th

Undiagnosed Undescended Testis in Young Adults in Saudi Arabia: Examination-Based Prevalence, Sociocultural Barriers to Early Care and Public Health Implications.

BACKGROUND: Undescended testis (UDT) persisting into adulthood increases risks of infertility and testicular cancer. In Saudi Arabia, delayed diagnosis is common and population prevalence of undiagnosed adult UDT is unknown. This study estimated point prevalence of previously unrecognized UDT among healthy young adult males in Riyadh and explored sociocultural barriers to early care. METHODS: A cross-sectional, examination-based screening was conducted January - August 2025 at Al Kharj Armed Forces Hospital. Simple random sampling recruited 1349 Saudi males aged 18-28 from university, militar

Psychotic Features in Myhre Syndrome: Evidence for Broader Neuropsychiatric Surveillance.

Myhre syndrome is a rare genetic disorder caused by pathogenic variants in SMAD4. The neurocognitive profile may include intellectual disability and developmental delay across a wide spectrum of domains. Four individuals with Myhre syndrome and psychosis and/or schizophrenia have been reported. We discuss three additional individuals (one briefly mentioned in a review of patients with Myhre syndrome) with SMAD4 pathogenic variants at codon Arg496Cys (two cases) and codon Ile500Val (one case). All had social challenges and variable developmental delays. They presented with acute-onset psychosis

Father and son with a pathogenic variant c.614dup p.(Gln206Thrfs*20) in the NR5A1 gene: a case report.

BACKGROUND: Steroidogenic factor 1 (SF-1), encoded by the NR5A1 gene, is a critical transcriptional regulator of adrenal and gonadal development. Pathogenic NR5A1 variants lead to a broad phenotypic spectrum characterized by insufficient virilization, including gonadal and testicular dysgenesis, ambiguous genitalia, hypospadias, micropenis, cryptorchidism, anorchia, and male infertility. This case report characterizes the clinical and genetic findings in a boy and his father with differences of sex development (DSD) caused by a heterozygous NR5A1 frameshift variant, thereby expanding our under

Unraveling the Mechanistic Spectrum of Myhre Syndrome: SMAD4 Signaling Disruption, Skeletal Phenotypes, and Translational Innovation.

Myhre syndrome (MYHRS) is an ultra-rare, progressive multisystem disorder caused by recurrent heterozygous missense variants in the SMAD4 gene, a central mediator of TGF-β and BMP signaling. Skeletal abnormalities-including postnatal short stature, brachydactyly, thickened calvarium, and craniofacial dysmorphism-are cardinal features, often accompanied by joint contractures and progressive soft tissue fibrosis. Extensive cellular and genetic evidence supports a gain-of-function (GoF) mechanism wherein mutant SMAD4 displays increased protein stability and prolonged nuclear localization, enhanci

Microdeletion and microduplication syndromes, including recurrent rearrangements at 16p11.2 and 22q11.21, are enriched in unexplained male infertility.

STUDY QUESTION: What is the impact of undiagnosed microdeletion and microduplication syndromes (MMS) for men with idiopathic low sperm count? SUMMARY ANSWER: Among idiopathic male infertility, ∼2% of cases harbour known disease-causing microdeletions and duplications linked to clinically well-established syndromes, representing ∼2.5-fold higher prevalence than in the general population. WHAT IS KNOWN ALREADY: While infertility affects up to 10% of men, a substantial proportion of cases remain with no identifiable underlying cause. Recurrent submicroscopic losses or gains cause MMS, some of w

Risk of testicular cancer and exposure to welding fumes.

The incidence of germ-cell testicular cancer (TC) has increased in recent decades. Current evidence does not indicate a unanimous association between occupational exposure to welding fumes and TC risk. However, most publications do not provide information on exposure levels to welding fumes. We investigated the association between occupational exposure to welding fumes and the risk of TC in a German case-control study (268 cases and 797 control subjects). A measurement-based welding-exposure matrix was used to estimate year-specific exposure values which were linked to job-task descriptions. O

Disorders of sex development associated with MPI and RSPH1 variants expand the phenotypic spectrum of CDG and PCD in Morocco.

BACKGROUND: Human sex development is a highly regulated process guiding undifferentiated gonads toward a testicular or ovarian fate. Disruptions in this pathway result in disorders of sexual development (DSD), characterized by atypical chromosomal, gonadal, or anatomical sex. These conditions usually appear as ambiguous genitalia at birth or as atypical pubertal development during adolescence. Different etiologic, phenotypic, and genotypic factors can cause DSD. Advances in next-generation sequencing (NGS) have significantly accelerated the identification of genetic variants through targeted p

Elevated anti-müllerian hormone concentrations in blood serum of dogs with oligozoospermia or azoospermia.

Reduced semen quality such as oligozoospermia or azoospermia is a frequent reason for subfertility in dogs. Anti-Müllerian hormone (AMH), produced by Sertoli cells, is known to play a role in spermatogenesis. Elevated serum AMH concentrations have been reported in dogs with Deslorelin-induced testicular atrophy resulting in azoospermia. This led us to hypothesize that congenital or acquired oligozoospermia or azoospermia often due to testicular degeneration or atrophy may also result in increased AMH serum levels, and that AMH could potentially serve as a diagnostic marker. The study included

A Hemizygous MED12 Variant in Three Brothers with Hypomasculinized Genitalia and Additional Clinical Features: A Case Report.

INTRODUCTION: MED12 is a causative gene for congenital malformation syndromes. The association between MED12 variants and 46,XY disorders/differences of sex development (DSD) remains unclear, although several variant-positive patients exhibited genital abnormalities. CASE PRESENTATION: Three siblings manifested hypomasculinized genitalia, including hypospadias and cryptorchidism, with normal or mildly increased gonadotropin levels. Two patients showed normal spontaneous puberty. Whole exome sequencing identified a maternally derived hemizygous MED12 variant (c.3064A>G, p.Met1022Val). One pati
