Tissue-Specific Parasite Persistence Following Chemotherapy in a Drug-Resistant Trypanosoma cruzi Preclinical Model.

Therapeutic failure in Chagas disease remains a major unmet medical challenge, often associated with intrinsic variability in drug susceptibility among Trypanosoma cruzi strains. However, the biological basis of incomplete parasite clearance after treatment remains poorly understood. Here, we investigated the tissue distribution of an intrinsically drug-resistant T. cruzi strain in a murine model of acute infection and evaluated the effects of short-term treatment with benznidazole or E1224 on parasite burden and host immune responses. Parasite loads were quantified by qPCR in multiple tissues

Orally transmitted Chagas disease outbreaks in Latin America: a comprehensive systematic review and public health implications.

OBJECTIVE: Food-borne Chagas disease is an emerging public health concern in South America, yet key aspects of its epidemiology and clinical impact remain poorly characterized. This systematic review aimed to systematically synthesize the available evidence on food-borne Chagas disease outbreaks in order to characterize their epidemiological, clinical, laboratory, treatment, and follow-up features, and to identify major knowledge gaps. METHODS: A systematic review of food-borne Chagas disease outbreaks reported between 1965 and 2024 was conducted following PRISMA-2020 guidelines (PROSPERO-CRD

Intranasal Vaccination With Recombinant Human Adenovirus Encoding trans-Sialidase Gene Protects Mice From Lethal Subcutaneous and Oral Trypanosoma cruzi Challenges.

BACKGROUND: The majority of new cases of Chagas disease, caused by Trypanosoma cruzi, have been associated with oral infection. Thus, it is reasonable that any vaccine formulation developed should elicit systemic and mucosal immunity. METHODS: Antigen-specific T-cell, B-cell, antibody-secreting cell, IgG, IgA, cytokine, and parasite DNA were assessed in Balb/c mice immunized with a recombinant adenovirus replication-deficient vector encoding the parasitic trans-sialidase (rAdTS) and challenged with T. cruzi trypomastigotes. RESULTS: A 2-dose intranasal (IN) rAdTS administration protected mic

Trypanosoma cruzi retinitis following donor-derived Chagas disease after heart transplantation.

Donor-derived Trypanosoma cruzi infection is a recognized complication of solid organ transplantation, but ocular involvement has never been reported. We described a case of necrotizing retinitis due to T. cruzi in a heart transplant recipient. A 62-year-old man developed unexpected donor-derived Chagas disease 3 months posttransplant and completed a 12-week course of nifurtimox with near-complete clearance of parasitemia. Thirty-eight days after nifurtimox discontinuation, he presented with progressive unilateral vision loss and clinical findings consistent with acute retinal necrosis. Vitreo

Disporum cantoniense suppresses non‑small cell lung cancer through AMPK/mTOR/ULK1‑mediated autophagy‑dependent apoptosis.

PURPOSE: Advances in drug discovery and early clinical development depends fundamentally on robust pre-clinical research and a comprehensive understanding of cancer biology. There is an international effort to reduce the use of animals in research and product development, with a focus on technical advances to build better in vitro models. Three-dimensional (3D) cell culture mimics the architecture of solid tumors and could be validated as an alternative method to animal use in research and development, promoting faster and more. Chagas disease in a naturally-infected dog from Northeast Brazil: a case report.

BACKGROUND: Dogs are the main domestic reservoir host of the parasite Trypanosoma cruzi, the causative agent of Chagas disease, and they are considered sentinel animals for the detection of new cases of human infection. Canis familiaris is also a well-established experimental T. cruzi infection model, but little is known about the progression of Chagas disease in naturally-infected dogs, especially during the acute phase in these hosts. Triatomine species infected with discrete typing units (DTUs) I, II and III of T. cruzi have been previously found in many of the municipalities of the state o

Effect of 12-wk dietary nitrate supplementation on carotid arterial stiffness in postmenopausal females.

Menopause is associated with reduced nitric oxide (NO) bioavailability, a key contributor to increased arterial stiffness and, consequently, greater risk of cardiovascular disease-related mortality in postmenopausal females. Even though dietary nitrate has been shown to increase NO bioavailability in postmenopausal females acutely, previous studies showed no impact of dietary nitrate supplementation on arterial stiffness in postmenopausal females. Their findings were likely limited by the acute and/or short-term design. Thus, this study aimed to determine whether 12 wk of dietary nitrate suppl

Serial 'deep-sampling' PCR of fragmented DNA reveals the wide range of Trypanosoma cruzi burden among chronically infected human, macaque, and canine hosts, and allows accurate monitoring of parasite load following treatment.

Infection with the protozoan parasite Trypanosoma cruzi is generally well-controlled by host immune responses, but appears to be rarely eliminated. The resulting persistent, low-level infection results in cumulative tissue damage with the greatest impact generally in the heart in the form of chagasic cardiomyopathy. The relative success in immune control of T. cruzi infection usually averts acute phase death but has the negative consequence that the low-level presence of T. cruzi in hosts is challenging to detect unequivocally. Thus, it is difficult to identify those who are actively infected

Sympathetic neural overdrive, vascular dysfunction, and diminished exercise capacity in patients with long COVID-19: a long-term study of cardiovascular sequelae.

We have recently showed that patients with severe coronavirus disease (COVID) have neurovascular dysfunction, cardiac morphofunctional alterations, and attenuated exercise capacity. However, whether these alterations persist over time is unknown. Here, we tested the hypothesis that patients with long COVID, even 2 yr after severe acute respiratory syndrome coronavirus 2 (SARS-COV-2) infection, exhibit sympathetic overdrive, aortic stiffening, endothelium-dependent dysfunction, cardiac morphofunctional changes, and diminished exercise capacity. Eighteen patients with long COVID and 19 well-matc

Biological and Molecular Characterization of Five Trypanosoma cruzi (Chagas, 1909) (Kinetoplastida, Trypanosomatidae) Isolates from the State of Hidalgo, Mexico.

Trypanosoma cruzi, the causal agent of Chagas disease, exhibits great genetic diversity, which has been related to its biological properties. However, these are poorly known in strains from the endemic area of Hidalgo. To assess the parasite's virulence, we evaluated parasitemia, mortality, and tropism in thirteen organs of CD1 mice during the acute phase of infection. For genotyping, we amplified the mini-exon gene from T. cruzi DNA using PCR. All five isolates were identified as belonging to DTU TcI. The peak of parasitemia occurred between 25 and 29 days post-infection. The Tultitlán and Ol

Risk of Incidence and Lethality by Etiology of Severe Acute Respiratory Syndrome in Hospitalized Children Under 1 Year of Age in Brazil in 2024: A Cross-Sectional Study.

Severe Acute Respiratory Syndrome (SARS) represents a significant cause of morbidity and mortality in children under one year of age, a particularly vulnerable population due to immunological and respiratory immaturity. The diverse etiology includes multiple respiratory viruses such as Respiratory Syncytial Virus (RSV), influenza, rhinovirus, and SARS-CoV-2, each with distinct potential to cause severe illness and death. Understanding the specific incidence and lethality by etiological agents in the recent Brazilian context (2024), after the COVID-19 pandemic, is essential to guide surveillanc

Advanced Heart Failure Secondary to Chagas Cardiomyopathy: A Case of Successful Left Ventricular Assist Device Placement.

Chagas cardiomyopathy is a rare but critical cause of nonischemic heart failure, particularly in patients from endemic regions. We present a 48-year-old Spanish-speaking male from Guatemala with hypertension, hyperlipidemia, and heart failure with reduced left ventricular ejection fraction (LVEF, 10%) due to Chagas disease. Despite guideline-directed medical therapy and implantable cardioverter-defibrillator (ICD) placement, he developed cardiogenic shock. Right heart catheterization confirmed severely reduced cardiac output and elevated wedge pressure. He was ineligible for heart transplantat

Dynamic evaluation of a COVID-19 death prediction model using Extreme Gradient Boosting Predictive Model.

The COVID-19 pandemic has evolved dynamically with the emergence of new variants and an increase in vaccination coverage. Given the high fatality rate of severe COVID-19, disease severity prediction models must incorporate these temporal variations. In this light, the present study seeks to develop a model to predict COVID-19 mortality in hospitalized patients. The Extreme Gradient Boost model was used to predict COVID-19 mortality upon hospital admission, and the results were correlated with laboratory test results, vaccination status, comorbidities, and clinical signs and symptoms at the tim

Trypanosoma cruzi DTU TcI disproportionately associated with fatal T. cruzi myocarditis in dogs in Texas, USA.

Acute heart failure and sudden death are possible outcomes of Trypanosoma cruzi infection in domestic dogs. Understanding which discrete typing units (DTUs) are present in fatal canine Chagas disease cases may help explain variation in disease outcome among infected dogs. We conducted a retrospective study of cases from the Texas A&M Veterinary Medical Diagnostic Laboratory and sampled heart tissue from 62 dogs with a histopathologic diagnosis of Chagas disease from 2007 to 2013 for which formalin-fixed paraffin embedded heart tissue was available. Extracted DNA was subjected to qPCRs for T. c

Effective Ultraslow Low-Dose Thrombolysis for Aortic Bioprosthetic Valve Thrombosis.

BACKGROUND: Bioprosthetic valve thrombosis (BPVT) is rare, with an incidence of 0.2% to 6.0%. It often presents as heart failure, thromboembolism, or a new murmur. Diagnosis relies on transthoracic and transesophageal echocardiography, with treatment traditionally being surgical. CASE SUMMARY: We report a 74-year-old patient with valvular and Chagas-related heart disease who developed BPVT of the aortic valve. Owing to high surgical risk and lack of response to heparin, ultraslow low-dose thrombolysis was performed, achieving a successful outcome. DISCUSSION: The emerging therapies for BPVT

Haemodynamic response to peripheral and inspiratory resistance training in patients with coronary artery disease (HEMODYRET): study protocol of a prospective, single-centre, randomised crossover trial.

INTRODUCTION: Due to the documented benefits of peripheral resistance training (RT) and inspiratory resistance training, known as inspiratory muscle training (IMT), in patients with cardiovascular disease, both exercise forms are regularly used in cardiac rehabilitation. However, little is known about the haemodynamic responses to different intensities of dynamic RT, isometric RT, and IMT in patients with coronary artery disease (CAD). This study is designed to evaluate and compare the acute haemodynamic responses elicited by different RT and IMT modalities in patients with CAD. METHODS AND A

Efficacy of sulfonamides targeting malic enzyme in an animal model of Chagas disease.

INTRODUCTION: Even after a century since its discovery, Chagas disease remains a major public health concern. Benznidazole and nifurtimox are the only approved treatments, but their limited efficacy and adverse effects highlight the urgent need for new therapies. In the last decade, several phenotypic screenings performed by pharmaceuticals and academic groups revealed new promising compounds. In a previous study, we identified the T. cruzi malic enzyme (TcME) as the molecular target for the sulfonamide TCMDC-143108, a phenotypic hit made public available through the Chagas Box collection.
