Toxoplasmosis is a parasitic infection commonly related to ocular lesions and neonatal malformations. Since the early 1950s, the first-line treatment of this disease has relied on the combination of sulfadiazine, pyrimethamine, and folinic acid. Over those years, only a few alternative regimens have been introduced. This highlights the need for discovering new bioactive molecules against Toxoplasma gondii. Given that this pathogen is an obligate intracellular parasite, traditional drug screening in the laboratory typically requires expensive materials and equipment, such as assays using fluore

Congenital toxoplasmosis results from transplacental transmission of Toxoplasma gondii during maternal primary infection. While the classic triad-hydrocephalus, intracranial calcifications, and chorioretinitis-defines severe forms, hypothalamic-pituitary axis involvement is extremely rare. Endocrine complications, particularly central hypothyroidism, are seldom described in this context. We report a preterm newborn at 35 weeks' gestation, representing the first documented case of isolated central hypothyroidism associated with severe congenital toxoplasmosis. Neuroimaging revealed major triven

PURPOSE: To describe the therapeutic efficacy of intravitreal faricimab in a refractory case of macular neovascularization (MNV) secondary to toxoplasmic chorioretinitis.
METHODS: Case report.
RESULTS: A 29-year-old woman presented with decreased vision in her right eye. She had been diagnosed with ocular toxoplasmosis four years earlier and subsequently developed secondary MNV, which was treated with intravitreal anti-vascular endothelial growth factor (anti-VEGF) injections. Previous intravitreal bevacizumab and aflibercept treatments resulted in minimal functional and anatomical improveme

Toxoplasmosis is a leading cause of death from foodborne illness. Its causative agent, Toxoplasma gondii, forms cysts in skeletal muscle, contributing to parasite persistence and transmission. Cases of myositis have been reported in T. gondii-infected immunocompetent individuals, and impaired myogenesis has been demonstrated in mice. Herein we investigated the effects of T. gondii infection on glycolytic tibialis anterior (TA) and oxidative soleus (SOL) muscles in mice, and whether prior physical exercise could prevent infection-induced myopathology. Functional, metabolic, histological, inflam

The novel work by Upadhyaya et al. suggests a potential use of intravitreal clindamycin for treating congenital toxoplasma retinochoroiditis. We share our clinical experience of encountering a sudden spike in intraocular pressure (IOP) after administering intravitreal clindamycin (5 mg/0.5 mL) in a neonate. Intravitreal injections are known to cause an increase in IOP immediately, which reduces in a few minutes. However, a smaller vitreous volume in neonates can cause an exponential increase in IOP immediately post-injection, posing a risk of optic nerve damage and compromise in retinal perfus

BACKGROUND: Ocular toxoplasmosis is a leading cause of infectious posterior uveitis worldwide. The retinal pigment epithelium (RPE), a key barrier and immunomodulatory layer in the eye, is directly targeted by Toxoplasma gondii during infection. However, its role in orchestrating the local immune response remains unclear.
OBJECTIVES: To investigate whether RPE cells actively drive macrophage migration during T. gondii infection in vitro, and to identify associated cytokine profiles.
METHODS: Adult retinal pigment epithelial cells (ARPE)-19 and primary RPE cells were exposed to tachyzoites, s

ABSTRACT: PURPOSE: Toxoplasmosis is the most common cause of infectious uveitis of the posterior segment. Current literature regarding this parasitic ocular infection in children is sparse. In this study, we focused on localisation of retinal infiltration and visual outcome in children and adolescents.
ABSTRACT: MATERIAL AND METHODS: Single-centre, retrospective study from 2012 to 2021. Patients were divided into three subgroups depending on foveal involvement: (1) direct foveal infiltrate, (2) with extrafoveal infiltrate, either with or (3) without fovea-involving inflammation. The primary o

Ocular toxoplasmosis is a relapsing infectious eye disease that carries an increasing risk of vision loss with each reactivation episode. Antimicrobial drug prophylaxis has been used to reduce the rate of recurrence. This review aims to summarize the current literature regarding expert clinician preferences, as well as the effectiveness and safety of prophylaxis. A literature search was conducted using the PubMed platform of the National Library of Medicine of the National Center for Biotechnology Information and relevant pre-specified search terms. Four professional surveys indicated that app

Toxoplasma gondii-the causative agent of toxoplasmosis-is a zoonotic pathogen of warm-blooded hosts. Infection causes mild-to-severe symptoms, including lethargy, fever, muscle pain, abortion, ocular disease, and encephalitis. Toxoplasma affects many vertebrate species, although felids are the only known definitive hosts. Seroprevalence in wildlife is often assessed using cross-sectional data, but few studies have tracked individual-level infections through time. We present a 4-yr dataset from white-tailed deer (Odocoileus virginianus) with repeated sampling of individuals that highlights chal

Toxoplasmosis is a globally significant infectious disease, affecting approximately one-third of the world's population. Ocular toxoplasmosis (OT) is a major and vision-threatening clinical manifestation. However, its pathogenesis remains elusive, and effective targeted therapies are unavailable. This study demonstrates that Toxoplasma gondii infection induces epithelial-mesenchymal transition (EMT)-like changes in both human retinal pigment epithelial (RPE) cells (ARPE-19) and murine ocular tissues. Mechanistic investigations, employing rhoptry protein 16 (ROP16) knockout parasites and ROP16

Toxoplasmosis affects approximately one-third of the global population. Despite the high seroprevalence in Brazil, contemporary data on the incidence and clinical spectrum of congenital toxoplasmosis (CT) in tertiary-care settings remain limited. Few studies have specifically addressed the incidence of symptomatic CT, limiting the identification of risk factors, preventive strategies, and optimal patient management, particularly in the context of recent healthcare disruptions and environmental changes. This study aims to determine the incidence of exposure to gestational toxoplasmosis and CT a

BACKGROUND: Amazonian toxoplasmosis (AT) is a severe form of acute toxoplasmosis occurring in the Guiana Shield and characterized by a high frequency of multivisceral involvement, even in immunocompetent individuals. Pulmonary, ocular, hepatic, and lymphatic manifestations are well documented, but cardiac involvement remains exceptionally rare and likely underdiagnosed. The diagnosis is challenging because the early symptoms are nonspecific and cardiac abnormalities may be clinically silent. We present a case of severe AT with documented myopericarditis in a previously healthy young adult (one

Toxoplasmosis, caused by the protozoan Toxoplasma gondii, affects nearly one-third of the global population and may result in severe congenital, ocular, and neurological manifestations. Current therapies are limited by toxicity, poor efficacy against chronic infection, and lack of activity against tissue cysts, highlighting the need for new therapeutic strategies. Aspartic proteases represent promising but underexplored drug targets in T. gondii. In this study, we evaluated the anti-T. gondii potential of a panel of aspartic protease inhibitors initially developed for inhibition of Plasmodium.

Congenital toxoplasmosis rarely occurs in infants born to mothers with evidence of prior immunity. We report the case of a preterm infant with severe ocular and neurological manifestations of congenital toxoplasmosis, despite maternal seropositivity (IgG+/IgM-) during pregnancy. This case highlights the potential for reactivation or reinfection with virulent strains and reinforces the need for vigilance and preventive strategies throughout pregnancy.
