BACKGROUND: Oral-ocular synkinesis is a frequent sequela of idiopathic facial paralysis (IFP), yet objective, dynamic methods to quantify associated palpebral fissure changes remain limited.
OBJECTIVE: To objectively measure palpebral changes in IFP using automated facial tracking.Design Type: Case-control.
METHODS: A novel, video-based facial tracking software assessed facial expressions in 12 controls and 30 IFP cases with synkinesis at least 12 months from onset of IFP. Palpebral fissure dimensions were measured at rest and during smile, laugh, and pucker. Bilateral differences across exp

Biallelic FKBP10 variants cause autosomal recessive osteogenesis imperfecta(OI) type XI (OI-XI) and Bruck syndrome type 1 (BS-1), both characterized by bone fragility. However, BS-1 is additionally marked by joint contractures, leading to diagnostic overlap with OI-XI. To present two FKBP10-related cases illustrating the phenotypic continuum and diagnostic challenges between BS-1 and OI-XI. Case 1, a 3.5-month-old male, had multiple fractures, progressive joint contractures, and scoliosis. Genetic testing revealed a novel homozygous FKBP10 variant, c.603T>A (p.Tyr201Ter), confirming BS-1. Case

BACKGROUND: Neurofibromatosis type 1 (NF1) is an autosomal dominant neurocutaneous syndrome. NF1-related vasculopathy represents a clinically significant yet underrecognized complication. Moyamoya syndrome, a rare cerebrovascular manifestation of NF1, has been rarely reported in the paediatric population. Epilepsy and hydrocephalus are also uncommon neurological manifestations of NF1. However, the co-occurrence of the above three symptoms in NF1 cases is rarely found.
METHODS: Here, we report a 2-year-old boy with moyamoya syndrome, epilepsy and hydrocephalus caused by de novo NF1 pathogeneti

Rubinstein-Taybi syndrome (RTS) is a rare congenital neurodevelopmental disorder associated with several ocular and lacrimal anomalies. A 40-year-old female patient with RTS presented with persistent right-sided epiphora following an earlier unsuccessful endoscopic dacryocystorhinostomy performed elsewhere. The patient underwent revision external dacryocystorhinostomy with fistulectomy and intubation. Intraoperatively, a focal polypoidal lacrimal sac lesion was noted, and histological analysis revealed significant immunoglobulin G4 (IgG4)-plasma cell infiltration. However, the diagnostic crite

Accessory liver lobes (ALLs) are rare anatomical variations characterized by supernumerary lobes of normal hepatic parenchyma in continuity with the liver. They are typically asymptomatic and most often discovered incidentally during imaging or surgical procedures. We report a unique case of a three-month-old premature infant with a history of gastroschisis and bladder herniation, who developed clinical and biochemical signs of progressive cholestasis. Imaging studies raised suspicion for an ALL, which was confirmed intraoperatively as a pedunculated ALL in continuity with the gallbladder. The

Congenital linear scleroderma (CLS) is a rare connective tissue disorder characterized by unilateral linear induration that can lead to restricted mobility and limb deformities. Various treatments have been proposed, including D-penicillamine, phototherapy, oral vitamin D, and immunosuppressants such as methotrexate (MTX), cyclosporine, and interleukin-6 (IL-6) inhibitors. We present the case of a 14-year-old female patient with refractory CLS, whose condition continued to worsen despite treatment with tocilizumab, mycophenolate mofetil (MMF), and MTX. She was subsequently started on a glucago

Jeune syndrome (JS), first described by Jeune as asphyxiating thoracic dystrophy, is an autosomal recessive osteochondrodysplasia with characteristic skeletal abnormalities and variable renal, hepatic, pancreatic, and ocular complications. Approximately 1 in every 100,000 to 130,000 babies is born with JS. Most patients with JS have respiratory distress due to inadequate lung development and many lose their lives due to respiratory failure. Those who survive have serious comorbidities. In terms of ophthalmological diseases, JS is classified among the hereditary syndromic retinopathies. Most, i

Parry-Romberg syndrome (PRS) is a rare acquired disorder characterized by progressive hemifacial atrophy involving the skin, subcutaneous tissue, muscles, cartilage, and bone. The disease usually progresses for two to 20 years before stabilizing, with extracranial involvement of the trunk and extremities occasionally reported. The etiology remains uncertain, with proposed immunological, genetic, and neurovascular mechanisms. We present the case of a 24-year-old female with PRS who exhibited intraoral and extraoral soft tissue atrophy, localized alopecia, ocular changes, and otologic involvemen

BACKGROUND: Rubinstein–Taybi syndrome (RSTS) is an autosomal-dominant neurodevelopmental disorder caused by heterozygous loss-of-function variants in CREBBP or EP300. EP300-related cases often show milder clinical features, but their full phenotypic spectrum and the potential role of oligogenic inheritance remain unclear. METHODS: We retrospectively studied seven Chinese pediatric RSTS patients. Genomic analyses included whole-exome sequencing (WES), chromosomal microarray analysis (CMA) and Sanger validation. Variants were classified by ACMG/AMP guidelines. Protein structural impacts were ass

BACKGROUND: The cranial base develops from multipotent mesenchymal cells through endochondral ossification. Genetic ablation of Sonic hedgehog (Shh) or Smoothened (Smo) leads to early apoptosis of cranial base cells, thus limiting the study of their role in the early development of cranial base. Our previous studies have shown that administration of 150 mg/kg Vismodegib (a Smo-specific small molecule antagonist) in E9.5- or E10.5-mice leads to premature mineralization of the skull base synchondroses. In the current study, we further investigated the molecular mechanisms underlying this model.

Parry-Romberg syndrome (PRS), also known as progressive facial hemiatrophy, is a rare neurocutaneous disorder characterized by progressive, unilateral atrophy of the facial tissues. It usually appears during childhood or early adulthood; however, adult-onset cases are increasingly being recognized. PRS significantly overlaps with localized scleroderma, especially the en coup de sabre (ECDS) subtype, indicating a common pathophysiological mechanism. We present a 52-year-old female patient with adult-onset PRS associated with ECDS. The patient was first diagnosed with localized scleroderma in 19
