Starving but surviving: a new RNA switch rewires parasite metabolism.

Intracellular Toxoplasma gondii parasites face severe metabolic stress during high-density infection. Giuliano et al. identify a novel mechanism involving the protein 'T. gondii parasite response to oxidation' (TgPRO). This RNA-binding protein regulates the translation of mRNAs vital for Fe-S cluster assembly and mitochondrial metabolism. TgPRO is crucial for oxygen-dependent adaptation and for ensuring optimal in vivo parasite replication.

Mitochondrial proteome of the apicomplexan Babesia divergens.

Babesia are tick-transmitted apicomplexan parasites of widespread medical and veterinary importance. Although atovaquone, a first-line therapy for human babesiosis, targets one of the three proteins encoded by mitochondrial DNA, a full characterization of the nuclear-encoded mitochondrial proteome is lacking. Here, we combined organelle immunoprecipitation and density gradient separation to enrich mitochondria from Babesia divergens. Using protein mass spectrometry and protein correlation profiling, we generated B. divergens MitoCarta (BdMitoCarta), a high-confidence inventory of 525 mitochond

Nationwide seroprevalence and spatial distribution of Toxoplasma gondii in wild boar (Sus scrofa) in South Korea, 2022-2025.

Toxoplasma gondii is a widespread zoonotic parasite that can infect nearly all warm-blooded vertebrates. Wild boars (Sus scrofa) are valuable indicator hosts because of their broad habitat use, omnivorous feeding behavior, and frequent contact with both natural and human-modified environments. However, comprehensive nationwide data on T. gondii exposure in wildlife in South Korea remain limited. This study aimed to investigate the seroprevalence, associated risk factors, and spatial distribution of T. gondii exposure in wild boars in South Korea using nationwide surveillance samples collected

Inducible dCas12a system expands CRISPRi/a capabilities for functional genomics in malaria parasite Plasmodium falciparum.

We developed inducible dead Cas12a (dCas12a)-based CRISPR interference/activation (CRISPRi/a) systems for Plasmodium falciparum, expanding the gene regulation toolkit beyond dCas9. Our previous systems fused dCas9 to the catalytic domains of PfSir2a or PfGCN5 under DiCre/loxP regulon, enabling conditional gene regulation but remaining constrained by Cas9's NGG protospacer adjacent motif (PAM) requirement. Unlike dCas9, dCas12a recognizes T-rich PAMs and processes a single transcript into multiple sgRNAs via its RNase activity, allowing multiplexed regulation of one or multiple genes. Functiona

Evaluation of a recombinant matrix antigen 1-based indirect enzyme-linked immunosorbent assay for serodiagnosis of Toxoplasma gondii infection in goats.

Toxoplasma gondii is an important zoonotic parasite affecting humans and livestock, and reliable serological tools are needed for disease surveillance in small ruminants. This study developed and evaluated a recombinant T. gondii matrix antigen 1 (TgMAG1) based indirect ELISA (iELISA) for detecting anti T. gondii antibodies in goat sera from northeastern Thailand. A truncated TgMAG1 fragment (amino acids 31-93) was selected to minimize cross-reactivity with Neospora caninum, expressed in Escherichia coli, and confirmed by SDS-PAGE, western blotting, and mass spectrometry. Overall, 213 goat ser

Molecular recognition of myosin L by the Toxoplasma gondii calmodulin.

Calmodulin is a conserved EF-hand Ca2+ sensor that transduces intracellular Ca2+ signals through interactions with target proteins. Recent cryo-electron microscopy studies revealed the Toxoplasma gondii calmodulin (TgCaM) as a light chain associated with the neck of the unconventional myosin MyoL within the preconoidal P2 ring , a key component of the conoid involved in parasite motility and invasion. Here, we used NMR spectroscopy, isothermal titration calorimetry, and complementary biophysical approaches to characterize TgCaM and its interaction with a synthetic peptide spanning the predicte

Broad-spectrum molecular detection of zoonotic agents in wild micromammals: a nationwide surveillance study in the Republic of Korea.

BACKGROUND: Wild micromammals serve as key hosts for diverse zoonotic pathogens, maintaining these agents in natural ecosystems and facilitating potential spillover to humans. Assessing pathogen prevalence in micromammals is essential for evaluating zoonotic risk and informing public health interventions. METHODS: In this study, we applied the TaqMan Array Card (TAC) platform, a high-throughput molecular diagnostic tool, to micromammal-borne zoonotic pathogen surveillance. From October to November 2022, a total of 160 wild micromammals, including Apodemus agrarius (Striped field mouse) (90.6,

Masquerade Syndrome Presenting as Optic Neuritis and Retinitis Due to Pituitary Germinoma.

Masquerade syndrome is a collective term for various ocular diseases presenting with clinical features resembling intraocular inflammation. Delays in definitive diagnosis and treatment can not only lead to irreversible vision loss but also impact the patient's prognosis regarding survival. We report the case of a 26-year-old male patient who presented with impaired vision in the right eye, which had persisted for six months; neuroretinitis was diagnosed. Although infectious neuroretinitis was suspected, tests for pathogens such as Mycobacterium tuberculosis, Toxoplasma, Toxocara, and Bartonell

Manipulation of host response and trajectory of parasite survival: A damage-response framework perspective.

The Damage-Response Framework (DRF) provides a conceptual model for understanding the pathogenesis of parasitic infections by distinguishing biological infection from clinical infectious disease. Rather than viewing parasite virulence as an intrinsic property, the DRF proposes that disease results from dynamic host-parasite interactions, with host damage determined by the magnitude and quality of the immune response. Both insufficient immune responses, permitting parasite proliferation, and excessive inflammation, causing immunopathology, can lead to disease. Using representative examples, inc

Highly Selective Thiazole Derivatives as Promising Anti-Toxoplasma gondii Agents.

Toxoplasmosis, caused by the protozoan Toxoplasma gondii, remains a significant global health challenge, particularly for immunocompromised individuals. Current therapies, based on pyrimethamine and sulfadiazine, are limited by toxicity, suboptimal efficacy, and the emergence of resistant parasite strains, highlighting the need for safer and more effective alternatives. In this study, we evaluated the antiparasitic potential of six 2-pyridyl 1,3-thiazole derivativesprivileged scaffolds previously reported for potent antitumor activity but never investigated against T. gondii. ATP Synthase-Associated Proteins TgASAP5 and TgASAP6 Sustain Mitochondrial Homeostasis in Toxoplasma gondii.

Toxoplasma gondii is an obligate intracellular apicomplexan parasite that infects a wide range of warm-blooded animals and causes a globally prevalent zoonotic disease. Mitochondrial ATP synthase is the terminal complex of oxidative phosphorylation and plays an essential role in parasite energy metabolism. However, the functions of many parasite-specific ATP synthase-associated proteins remain largely unknown. Here, we identified and characterized two novel ATP synthase-associated proteins, TgASAP5 and TgASAP6, in T. gondii and generated conditional knockdown parasite strains to investigate th

Zinc Metalloproteases of Apicomplexan Parasites: Conserved Targets for Broad-Spectrum Antiparasitic Drug Discovery.

The phylum Apicomplexa includes major protozoan pathogens of humans and animals (Plasmodium falciparum, Toxoplasma gondii, Cryptosporidium parvum, Babesia spp., Eimeria spp., and Neospora caninum) with substantial global health and economic impact. Resistance to key chemotherapeutics, including the partner and antifolate drugs used against several of these diseases, is narrowing a limited arsenal of target-specific treatments, creating an urgent need for mechanistically distinct drug targets. Proteolytic enzymes are attractive candidates, yet antiprotease drug discovery in the Apicomplexa has

A dedicated partitioning machinery ensures faithful inheritance of the apicoplast in Toxoplasma gondii.

Toxoplasma gondii tachyzoites divide by endodyogeny, an internal budding process that requires precise machinery for equal partitioning of single-copy organelles into daughter cells. The essential apicoplast is partitioned by temporal association with the centrosome, which is orchestrating various cell division processes. However, the identity of the molecular infrastructure facilitating anchoring of the apicoplast to the centrosome is still unknown. Here, we describe the apicoplast partitioning protein 1 (APP1), which first assembles in a specific basal complex (BC) section and then localizes

Identification and characterization of factors that govern apicoplast segregation in Toxoplasma gondii.

The apicoplast is an essential plastid organelle of apicomplexan parasites that supports indispensable metabolic pathways, and its successful inheritance during cell division is critical for parasite survival. Here, we identify two previously uncharacterized proteins, APP1 and APP2, as key components of the apicoplast segregation machinery in Toxoplasma gondii. We show that APP1 is an inner membrane complex associated protein that is expressed during daughter bud development and localizes near the centrosomes and apicoplast. Loss of APP1 results in apicoplast loss and missegregation and growth

Immune signatures of common exposures through co-occurrence of T-cell receptors in tens of thousands of donors.

BACKGROUND: Memory T cells are records of clonal expansion from prior immune exposures such as infections, vaccines and chronic diseases. Some of the receptors of these expanded T cell clones in a typical immune repertoire are highly public (present in many individuals) because they respond to the same peptide from a prevalent immune exposure, presented by the same Human Leukocyte Antigen (HLA) allele. Only a tiny fraction of public T-cell receptor β sequences (TCRs) have known associations with exposures or specific peptides. METHODS: We mined the TCR repertoires of tens of thousands of dono
