Toxoplasma gondii prevention literacy and self-reported preventive practices among online respondents in China: a cross-sectional survey.

Toxoplasma gondii is a widespread zoonotic parasite, yet prevention literacy and protective practices among online respondents in China remain poorly characterized. We conducted a cross-sectional survey from April to November 2025 using a purpose-designed questionnaire distributed through WeChat and QQ. Knowledge and preventive-practice items contributed 23 and 10 points, respectively; a composite score of ≥20/33 represented meeting the prespecified 60% threshold. Among 622 valid respondents, 56.8% had heard of Toxoplasma gondii, the mean composite score was 14.22 ± 6.69, and 21.4% met the thr

TGIF is a golgin-like protein required for Golgi structural maintenance and function in Toxoplasma gondii.

The Golgi is an essential organelle that serves as a central hub for endomembrane trafficking. In the protozoan parasite Toxoplasma gondii, a single Golgi stack is essential for parasite survival; however, the molecular determinants governing Golgi structure and function remain poorly understood. Here, we characterize a Golgi-associated protein that is required for Golgi integrity and function, which we named Toxoplasma Golgi Integrity Factor (TGIF). Loss of TGIF disrupts parasite replication and natural egress and is lethal to the parasite. To investigate the impact of TGIF depletion on secre

Molecular survey of zoonotic protozoa in free-ranging marmosets (Callithrix penicillata) from Central Brazil: implications for One Health surveillance.

Urban expansion and environmental fragmentation have intensified contact among wildlife, domestic animals, and humans, reshaping infectious disease dynamics at the human-wildlife interface. Non-human primates (NHP), particularly the black-tufted marmoset (Callithrix penicillata), readily adapt to anthropogenic landscapes and may serve as sentinel hosts for zoonotic pathogens. This study investigated the occurrence of Leishmania spp., Plasmodium spp., Toxoplasma gondii, and Trypanosoma cruzi. in 232 free-ranging C. penicillata found dead in urban and peri-urban areas of the Federal District, ce

Preliminary study on expression and functions of Toxoplasma gondii SRS67 and SRS20A proteins.

BACKGROUND: Toxoplasma gondii surface proteins play critical roles in parasite invasion, proliferation, and host immune modulation, and represent promising targets for anti-toxoplasmosis intervention. SRS67 and SRS20A are poorly characterized surface proteins of T. gondii, and their biological functions and host regulatory effects remain largely unclear. METHODS: In the present study, we prepared recombinant SRS67 and SRS20A proteins and generated corresponding polyclonal antibodies to explore their functional characteristics and protective potentials. RESULTS: Immunofluorescence analysis lo

Repurposing of Milciclib as a Potential Inhibitor of Toxoplasma gondii in Human Trophoblasts and Placental Explants.

Congenital toxoplasmosis persists as a significant public health challenge due to the limited efficacy and safety of current treatments during pregnancy. In this study, we assessed the antiparasitic activity and mechanism of action of milciclib (MCB), a cyclin-dependent kinase inhibitor from the Medicines for Malaria Venture Pathogen Box, against Toxoplasma gondii using in vitro and ex vivo models of the maternal-fetal interface. MCB exhibited no cytotoxic effects in human trophoblastic BeWo cells or placental villous explants at concentrations up to 1.62 μM. The compound significantly reduced

A Toxoplasma gondii-derived ROP38 protein engages Ser15/Thr18-linked p53 activation and DNA damage-associated apoptotic signaling in human cancer cells.

BACKGROUND: The tumor suppressor p53 integrates diverse cellular stresses to regulate DNA damage responses, cell-cycle progression, and apoptosis-associated signaling. Toxoplasma gondii secretes rhoptry effectors that remodel host signaling networks, but the host targets and signaling functions of the rhoptry kinase ROP38 remain poorly defined. METHODS: GFP-tagged T. gondii ME49 ROP38 full-length protein (ROP38 Full) and its kinase-domain fragment (ROP38 KiD) were stably expressed in HCT116 and HepG2 human cancer cells. Kinase-associated activity was assessed in anti-GFP immunoprecipitates, a

4‑O‑Allyl Derivative of Natural Licarin A Exhibits Selective In Vitro Activity against Toxoplasma gondii Tachyzoites.

Toxoplasma gondii is an obligate intracellular protozoan responsible for toxoplasmosis, a widespread parasitic disease that poses a serious health threat, especially to immunocompromised patients and pregnant women. Current therapeutic options remain unsatisfactory because they are associated with adverse effects and do not eliminate latent infections, emphasizing the need for new antitoxoplasmic agents. In the present study, the natural neolignan (-)-licarin A (1), isolated from the Brazilian species Nectandra oppositifolia, together with three semisynthetic derivatives, 4-O-methyl (1a), 4-O-

Influence of ecological and management factors on Toxoplasma gondii infection in small ruminants in Algeria.

Toxoplasma gondii is a zoonotic protozoan of major concern for small-ruminant production and public health. This study is among the first to simultaneously compare T. gondii seroprevalence across contrasting Algerian bioclimatic zones. In summer 2024, a cross-sectional study was conducted across 82 herds comprising 4,022 small ruminants in three provinces of central Algeria (Blida, Médéa, Djelfa), representing contrasting bioclimatic zones (humid, semi-arid, and arid). Blood samples were collected from 1,052 animals older than six months to assess the apparent seroprevalence of T. gondii and a

Identification of Novel CD52+ Fibroblasts in Nodular Prostatic Hyperplasia.

BACKGROUND: Benign prostatic hyperplasia (BPH) and lower urinary tract symptoms are strongly associated with chronic prostate inflammation and stromal remodeling, yet the stromal cell programs that emerge during inflammatory, nodular remodeling remain incompletely defined. A Toxoplasma gondii infection model produces persistent inflammation and nodular formations, providing an experimentally tractable system to map inflammation-linked stromal states. METHODS: We performed single-cell RNA sequencing of prostates from control and chronically infected male CBA/J mice harvested at 28 days post-in

Toxoplasmosis: Epidemiological Significance, Economic Burden, and Emerging Directions in Drug Discovery and Artificial Intelligence.

Toxoplasmosis, a parasitic disease caused by Toxoplasma gondii, affects a wide range of warm-blooded hosts and remains most prevalent in tropical regions, with distribution shaped by climate, cultural practices, and environmental conditions. Clinical severity varies across susceptible populations, generating substantial economic burdens through medical costs and agricultural losses. Given this global health impact, a comprehensive, up-to-date understanding of the disease is essential. However, significant gaps persist. While diagnostic techniques and epidemiological surveillance have advanced

Associations of Antibodies Against Several Infections With Alzheimer Disease Neuropathology: A Prospective Cohort Study Analysis.

BACKGROUND AND OBJECTIVES: Associations of common infections with Alzheimer disease (AD) risk have been reported. A hypothesized mechanism for these is cerebral β-amyloid (Aβ) aggregation as a defense to infection. We studied whether serologic measures of several infections in the UK population are associated with measures of AD neuropathology. METHODS: This prospective association study used data from the 1946 British birth cohort. Circulating antibodies against 14 pathogens, measured at age 60-64 years, were modelled as pathogen serostatus (indicating lifetime exposure to an agent), pathoge

Presumed herpes simplex virus keratouveitis in a 2-year-old child: a case report.

BACKGROUND: Ocular herpes simplex virus (HSV) disease is uncommon in very young children and may be difficult to recognize because history and examination are limited and early manifestations can resemble bacterial conjunctivitis. Pediatric disease may involve multiple corneal layers, extend into the anterior chamber, recur, and threaten visual development. We describe presumed HSV keratouveitis in a 2-year-old child. CASE PRESENTATION: A 2-year-old girl presented with a two-month history of right-eye pain, photophobia, and tearing after an episode diagnosed elsewhere as HSV blepharoconjuncti

Oocyst wall protein EtOWP6 is critical for sporocyst formation and developmental progression in Eimeria tenella.

Transmission of apicomplexan parasites-including Eimeria, Toxoplasma, and Cryptosporidium-relies on the environmentally resilient oocyst wall, a bilayered structure whose integrity is maintained by cysteine-rich oocyst wall proteins (OWPs). Yet the functional roles of these conserved proteins remain poorly defined. Here, we characterize EtOWP6, a conserved OWP in Eimeria tenella, the causative agent of avian coccidiosis. Immunofluorescence reveals that EtOWP6 localizes specifically to wall-forming bodies and oocyst/sporocyst walls during sexual development. CRISPR/Cas9-mediated knockout demons

Type III Interferons (lambda) in Parasitic Infections: Balancing Mucosal Defense and Immune Modulation.

Parasitic infections are a significant global health issue, with pathogens often entering the hosts through mucosal surfaces. Type III interferons (IFN-λ) are key mediators of immune responses in these infections. Produced mainly at epithelial barriers, IFN-λ induces a specific set of IFN-stimulated genes (ISGs) that boost antiviral and antimicrobial defense without causing widespread systemic inflammation. This review examines functions of IFN-λ diverse in parasitic infections, focusing on roles in antimicrobial responses and immune modulation. Understanding these effects may lead to new ther

Toxoplasma serology: Breaking the diagnostic boundary of schizoaffective disorder? Insights from a comparative peripheral immune marker analysis.

BACKGROUND: Toxoplasma gondii (T. gondii) infection has been implicated in the pathophysiology of psychiatric disorders, yet no study has compared its seroprevalence between schizoaffective disorder (SZA) and other psychiatric conditions. This study aimed to compare T. gondii seroprevalence between patients with SZA, schizophrenia (SZ), and bipolar disorder (BD), and to examine whether seropositivity differentially affects innate immune markers across these diagnoses. METHODS: Overall 144 patients were included in this cross-sectional study. T. gondii seroprevalence was determined and leukocy

Temporal Neuroimmune Changes During Primary and Persistent Experimental Toxoplasmic Encephalitis.

Toxoplasma gondii persists in the central nervous system and may cause severe toxoplasmic encephalitis, particularly in immunocompromised hosts. Our previous studies demonstrated that T. gondii-induced neuropathology involves oxidative and nitrosative stress, apoptosis, glial activation, blood-brain barrier injury, and GMF-β- and SAP/NF-κB-associated responses. This study investigated the temporal immunoreactivity of CD68, S100B, Foxp3, and IL-5 during early and chronic experimental infection. Twenty-four female Swiss albino mice were assigned to one uninfected control group and three ME49-inf
