Pathology and parasite distribution in mice challenged with Toxoplasma gondii from different geographical origins.

Toxoplasma gondii (T. gondii), a zoonotic parasite, can cause severe disease in warm-blooded animals. Pathological changes in murine tissues infected with different T. gondii isolates were studied to establish factors influencing lesion severity and parasite burden. In Study A, mice were orally inoculated with genotype #3, #6 or #8 oocysts. In Study B, mice were inoculated intraperitoneally with genotype #1, #3, #6, #13, #141 or #265 tachyzoites. Mice were euthanised serially and tissues processed for histopathology. In Study A, genotype #6 caused pathology in the liver, brain, lung, intestine

Wild bird parasites in the Atlantic Forest: diversity and prevalence across forested and anthropogenic landscapes.

Wild birds are important for public health, as they have epidemiological characteristics that make them host species for the transmission and maintenance of several pathogens, either as reservoirs or by the dispersal of infected arthropod vectors. Recently, several bird species have been used as model organisms in several scientific studies. Investigations have used wild bird populations as bioindicators and sentinels to identify changes in habitat quality, exposure to chemical pollutants, and disease surveillance. The objective of this study was to investigate the prevalence and diversity of

Apicortin defines the Plasmodium apical conoid body but is dispensable for the parasite life cycle.

Apicomplexan parasites such as Plasmodium spp. and Toxoplasma gondii possess unique tubulin-based structures, including subpellicular microtubules and apical polar rings, which are essential for parasite motility, host cell invasion, and replication. Apicortin, a microtubule-associated protein, contains a doublecortin (DC) domain and a partial tubulin polymerization-promoting protein (TPPP) domain, both implicated in microtubule binding and stabilization. How tubulin-based structures are maintained is poorly understood, but it may involve Apicortin, so far found only in apicomplexans and the p

Human-wildlife conflict and its impacts on livelihoods, health, and the environment in Chebera Churchura National Park, Southwest Ethiopia: a phenomenological study using a One Health approach with an urgent call for action.

BACKGROUND: Chebera Churchura National Park (CCNP), located in Southwest Ethiopia, was established to conserve biodiversity. However, its contribution to the livelihood and wellbeing of the proximal communities remains limited. Local people derive few tangible benefits from conservation while facing significant costs through crop losses, livestock predation, and restricted access to natural resources factors that intensify human-wildlife conflict (HWC). Previous studies have addressed HWCs focused mainly from ecological and socio-economic aspects with limited attention to the linkage between h

Ring-enhancing lesions in an immunocompetent patient.

Ring-enhancing brain lesions present a common radiological conundrum, especially in immunocompetent hosts where there is a broad differential diagnosis. Toxoplasma gondii, an obligate intracellular protozoan, typically causes encephalitis in immunocompromised patients, especially those with HIV/AIDS. Toxoplasma cerebral lesions in apparently immunocompetent people pose a diagnostic dilemma. The clinical features may mimic neoplastic or fungal causes, particularly when there are atypical radiological and histological presentations. We report an immunocompetent patient with toxoplasma encephalit

Protection Against Toxoplasma gondii Lethal ME49 Challenge Induced by Influenza Virus-like Particles Containing Dense Granule Protein 14.

Background/Objectives: Toxoplasma gondii (T. gondii) dense granule antigen 14 (GRA14) is a parasitophorous vacuole membrane protein that plays a critical role in the development of chronic-stage cysts. However, its potential as a vaccine antigen and long-term immunity have not been evaluated using a virus-like particle (VLP) platform. Methods: influenza matrix protein (M1)-based VLPs displaying GRA14 were generated. Female BALB/c mice were intranasally immunized with the VLP vaccine and orally challenged with lethal ME49 cysts either 10 weeks or 32 weeks after prime vaccination for short-term

Assessing the Toxoplasma Tachyzoite Cell Cycle Phases Using Fluorescent Ubiquitination-Based Cell Cycle Indicator.

Toxoplasma gondii is an apicomplexan parasite that infects a wide variety of eukaryotic hosts and causes toxoplasmosis. The cell cycle of T. gondii exhibits a distinct architecture and regulation that differ significantly from those observed in well-studied eukaryotic models. To better understand the tachyzoite cell cycle, we developed a fluorescent ubiquitination-based cell cycle indicator (FUCCI) system that enables real-time visualization and quantitative assessment of the different cell cycle phases via immunofluorescence microscopy. Quantitative immunofluorescence and live-cell imaging of

MiRNA and proteomic profiling of small extracellular vesicles secreted by Toxoplasma gondii cultured with two different human cervical epithelial feeder layers.

Toxoplasma gondii has been reported to exert antitumor effects by inducing apoptosis and cell cycle arrest; however, the biological roles of small extracellular vesicles released by the parasite remain poorly understood. Small extracellular vesicles derived from parasite-host coculture systems may encapsulate molecular signals that reflect interactions between T. gondii and distinct epithelial microenvironments. In this study, tachyzoites of the T. gondii RH strain were cocultured with human cervical adenocarcinoma HeLa cells or normal cervical epithelial H8 cells, and small extracellular vesi

Public Health Implications of Latent Toxoplasmosis and Its Association With Type 2 Diabetes: A Case-Control Study in Qazvin, North-Western Iran.

INTRODUCTION: Toxoplasma gondii is a worldwide-distributed zoonotic parasite, causing latent infections in humans. Increasing evidence has suggested a possible link between toxoplasmosis and diabetes mellitus (DM), though data from Iran are limited. This study was conducted to determine the seroprevalence of latent T. gondii infection and its association with diabetes in Qazvin province, north-western Iran. MATERIALS AND METHODS: In this case-control study, 350 patients with type 2 diabetes mellitus and 350 non-diabetic controls were recruited from clinical laboratories in Qazvin province. An

Bioassay of the infectivity of heat-treated Toxoplasma gondii cysts in susceptible C57BL/6J mice.

Toxoplasma gondii is a significant foodborne parasite. However, the precise thermal conditions required to inactivate its tissue cysts in meat remain poorly defined. This study systematically determined the effects of temperature (45-70 °C) and time (10-30 min) on cyst viability. Cysts treated under each condition were orally administered to susceptible C57BL/6J mice, and infectivity was comprehensively assessed through survival, clinical signs, serology (IgG), qPCR, and histopathology. Results demonstrated that treatment at 60 °C for 10 min or under more stringent conditions completely abolis

Establishment of an ELISA for detecting oocyst-derived Toxoplasma gondii infections in sheep.

BACKGROUND: Ovine toxoplasmosis is a zoonotic disease that severely impacts the development of the sheep industry. The primary routes of Toxoplasma gondii infection in sheep are cyst infection and oocyst infection. However, current technologies are unable to distinguish between these two infection pathways. METHODS: In this study, we initially screened eight proteins that are highly specifically expressed during the oocyst stage. Through Western blot (WB) analysis, we identified a protein (TGME49_267410) that could serve as a diagnostic antigen. Subsequently, we optimized the conditions for a

Microtubule inner proteins in apicomplexan parasites.

Microtubule inner proteins (MIPs) are integral components within the microtubule lumen of various organisms, contributing to microtubule structural integrity and functionality. Apicomplexan parasites, including Plasmodium spp. and Toxoplasma gondii, exhibit a range of specialized tubulin structures, such as axonemal microtubules, subpellicular microtubules (SPMTs), and conoid fibers, playing critical roles in cellular morphology and motility. Yet, in contrast with model organisms, only a few MIPs have been characterized in apicomplexans so far. Recent advances in cryo-electron tomography and s

ZFT is the major iron and zinc transporter in Toxoplasma gondii.

Transition metals, such as iron and zinc, are indispensable trace elements for eukaryotic life, acting as co-factors in essential processes ranging from metabolism to DNA replication. These metals can be transported into cells by an evolutionary-conserved family of metal transporters; however, how the ubiquitous mammalian parasite Toxoplasma gondii acquires essential metals has been unknown. Here, we have identified and characterised the first iron and zinc importer in T. gondii. This transporter, named ZFT, localised to the parasite plasma membrane and is essential for the parasite's life cyc

IFN-γ protects the human blood-brain barrier from Toxoplasma gondii-mediated dysfunction via CCL2 suppression.

The obligate intracellular parasite Toxoplasma gondii infects nearly one-third of the global population, yet its impact on human blood-brain barrier (BBB) function remains poorly defined. In this study, we use human induced pluripotent stem cell-derived (iPSC) brain-like microvascular endothelial cells (BMECs), a physiologically relevant BBB model, to investigate if T. gondii infection directly compromises barrier integrity. We show that infection induces robust monocyte chemotactic protein-1 (also known as CCL2) secretion from BMECs, and that CCL2 itself serves as a previously unrecognized dr

Structural and regulatory principles underlying apicomplexan conoid function.

Zeng et al. and Qian et al. reveal in their cryogenic electron microscopy (cryo-EM) studies how apicomplexan parasites extensively remodel tubulin to construct the conoid, a specialized invasion organelle. Distinct microtubule assemblies integrate mechanical support, force generation, and secretion control, providing a structural framework that underpins parasite motility and host cell entry.
