Toxoplasmosis Acquired in French Guiana in Immunocompetent Adults: Case Series From 2002 to 2019, With a Focus on Outpatients.

BACKGROUND: Amazonian toxoplasmosis is classically associated with severe disease in hospitalized adults. However, little is known about milder outpatient forms in French Guiana. METHODS: We retrospectively analyzed 174 immunocompetent adults diagnosed with acute toxoplasmosis in French Guiana (2002-2019), comparing clinical and laboratory features between hospitalized (HPs) and outpatient (OPs) cases. RESULTS: Seventy-four patients (43%) were OPs. Fever (93%), lymphadenopathy (62%), and digestive symptoms (56%) predominated among OPs, while respiratory symptoms, anemia, and hyponatremia pre

A protein disulfide isomerase coordinates redox homeostasis and ER calcium regulation for optimal lytic cycle progression in Toxoplasma gondii.

The endoplasmic reticulum (ER) maintains an oxidative environment that facilitates the formation of disulfide bonds, a critical process for proper protein folding. Protein disulfide isomerases (PDIs) are ER-resident enzymes that facilitate the formation, breakage, and rearrangement of disulfide bonds between cysteine residues, thereby stabilizing protein structures. Although PDIs are functionally diverse, they all contain at least 1 thioredoxin-like domain and most mediate disulfide exchange through their conserved CXXC motifs. The apicomplexan parasite, Toxoplasma gondii, infects approximatel

Distinct motors, shared mechanics: unifying principles of microbial gliding.

Gliding motility is one of the more elegant tricks in the microbial playbook, allowing cells to move smoothly along an external surface. Unlike swimming in bulk fluid, gliding requires contact with a biotic or abiotic surface, imposing strong physical constraints on how forces are generated, transmitted, and dissipated. It is an active, energy-dependent process that operates without flagella, instead relying on specialized molecular machines that couple dynamic surface adhesins to motion. In this mini-review, we summarize recent advances that illuminate how distinct molecular motors converge o

Global translational and metabolic remodeling during iron deprivation in Toxoplasma gondii.

Iron is required to support essential cellular processes. Due to diverse and dynamic host environments, the obligate intracellular parasite Toxoplasma gondii must adapt to iron-limited conditions. To investigate the adaptations critical to parasite survival under these conditions, we conducted proteomic and metabolomic profiling of Toxoplasma cultured in iron-depleted conditions. We find that iron depletion results in remodeling of the parasite proteome and triggers swift translational repression, prior to decreases in the key translational factor ABCE1. In the context of repressed translation

Maternal immune activation perturbs intestinal niche through microbial glycerophospholipids and drives offspring behavioral abnormalities.

Maternal immune activation (MIA) contributes to neurodevelopmental deficits in offspring, yet the prenatal mechanisms remain elusive. Here, using a mouse MIA model induced by prenatal exposure to Toxoplasma gondii soluble tachyzoite antigen (STAg), we reveal that maternal gut dysbiosis, characterized by increased Firmicutes and Actinobacteria, drives offspring behavioral deficits. This dysbiosis disrupts intestinal barrier integrity and induces metabolic and inflammatory dysregulation, characterized by elevated M1 macrophages and IL-6, alongside a pronounced accumulation of the glycerophosphol

A single-cell atlas of Toxoplasma sexual development in the feline intestinal tract.

Toxoplasma gondii undergoes sexual development exclusively in the feline intestine, a process critical for genetic diversity and population expansion. Recent studies have identified genes critical in suppressing presexual development and metabolic differences in felines that may promote sexual development, but to date the gene regulatory networks driving development in the cat are unknown. To investigate this, we performed single-cell transcriptomics on parasites isolated from cat intestines, using fluorescent reporter strains and flow cytometry. From 15,068 cells across two experiments, we id

The CLAMP-Linked Invasion Protein (CLIP) plays an essential role in Plasmodium berghei zoites.

Apicomplexan parasites such as Toxoplasma and Plasmodium spp. rely on the sequential secretion of parasite apical organelles, called micronemes and rhoptries, to invade host cells. The claudin-like apicomplexan microneme protein (CLAMP) is a conserved protein that plays an essential role during host cell invasion in Toxoplasma and Plasmodium zoites. Previous studies have shown that CLAMP is essential in Plasmodium merozoites for erythrocyte invasion and also in sporozoites for the invasion of the mosquito vector salivary glands and of mammalian host hepatocytes. In Toxoplasma gondii tachyzoite

Kinase inhibitors in organoid media influence Toxoplasma gondii growth and development.

UNLABELLED: Toxoplasma gondii is a widespread parasite that impacts both human and animal health. Increasing use of organoid model systems has made previously challenging aspects of the T. gondii lifecycle more accessible. The media for these organoid systems are highly complex with many growth factors and pathway inhibitors that promote stem cell retention or differentiation of the cells. We noticed changes in T. gondii growth and development in our intestinal organoid system and wanted to determine if this was driven by cell type or media components. We found that low concentrations of SB202

Pharmacological inhibition of the MST2/Hippo signaling pathway mitigates Toxoplasma gondii-induced apoptosis and immunopathology in macrophage and mice liver.

Toxoplasma gondii (T. gondii) is an obligate intracellular parasite that manipulates host cell apoptosis and inflammation to support its survival and replication. Our previous work demonstrated that T. gondii activates the Hippo signaling pathway through phosphorylation of mammalian STE20-like protein kinase 2 (MST2), thereby promoting host cell apoptosis. However, the impact of Hippo pathway inhibition on apoptosis, inflammation, and host protection during infection remains unclear. Here, we demonstrate that T. gondii infection induces apoptosis in RAW264.7 macrophages and causes acute liver

An in vivo fitness gene of Toxoplasma, MIC11, is essential for PLP1-mediated egress from host cells.

After invasion and replication, intracellular pathogens must egress from infected host cells. Toxoplasma gondii facilitates this process by permeabilizing host cells through induced secretion of perforin-like protein 1 (PLP1). However, the precise mechanism of host cell permeabilization remains enigmatic. Here, we identify the secretory microneme protein MIC11 as a key factor for membrane disruption. A CRISPR-based in vivo screen identifies MIC11 as the top in vivo fitness-conferring gene. Deletion of MIC11 results in severe defects in membrane rupture and egress. Scanning mutagenesis identifi

Toxoplasma gondii effector GRA35 mediates neuronal damage via ER stress and mitochondria-associated apoptosis.

Encephalitis resulting from acute reactivation of chronic Toxoplasma gondii infection in the central nervous system poses a significant mortality risk in immunodeficient individuals. However, the specific molecular mechanisms underlying this process remain elusive. We constructed the GRA35 gene knockout ME49 strain and compared the differences with wild type ME49 strain. We used the GST-pull down experiment to explore the mechanism of GRA35 promoting neuronal cell apoptosis. We used immunofluorescence, flow cytometry and CCK8 experiments to verify the pathway of GRA35 promoting neuronal cell a

Integrating design-of-experiments (DOE) optimization and risk assessment towards a safe and simplified electroporation protocol for Toxoplasma gondii.

Genetic manipulation of Toxoplasma gondii presents unique challenges due to its obligatory intracellular nature and relatively rapid growth. Electroporation is the main technique used to introduce genetic modifications into T. gondii. However, the existing protocols require an electroporation buffer comprised of eight components and involving multiple steps of preparation. Optimizing electroporation protocols, including a readily available buffer is crucial for achieving efficient transfection while simplifying the overall process. In this study, we present a modified Opti-MEM I based electrop

A streamlined integrated system integrating lysate release, freeze-dried reagents for multiplex polymerase chain reaction, and intelligent analysis for TORCHes pathogen identification.

INTRODUCTION: Current Toxoplasma gondii (TOX), Epstein-Barr virus (EBV), rubella virus, human cytomegalovirus (HCMV), and herpes simplex virus types I and II (HSV I and II) (TORCHes) diagnostic methods are limited by challenges, such as multi-step workflows and cold-chain dependency. This study aimed to develop an integrated "sample-in-result-out" system combining rapid nucleic acid release, room-temperature-stable multiplex polymerase chain reaction (PCR), and automated interpretation to improve diagnostic accuracy. METHODS: We utilized a self-prepared lysis buffer for fast nucleic acid extr

The human parasite, Toxoplasma gondii, is paralyzed without two components of the apical polar ring.

The phylum Apicomplexa contains ∼ 6000 known species of unicellular eukaryotic parasites. A unifying feature among the apicomplexans is the apical complex, which varies in complexity in different lineages, but always contains an annulus (a.k.a. the apical polar ring) into which the minus ends of an array of cortical microtubules are embedded. In Toxoplasma gondii, the apical complex also includes the conoid, which contains several signaling and structural proteins critical for parasite motility. The conoid extends and retracts through the apical polar ring in a calcium-dependent manner. Here w

Legacy 4(1 H )-quinolone scaffolds activity against acute and chronic Toxoplasma gondii infection.

Toxoplasma gondii is a protozoan parasite capable of infecting most warm-blooded animals, including humans, and can cause severe disease in immunocompromised individuals and the developing fetus. Current treatments for toxoplasmosis are effective only against the acute stage of infection and have limited or no activity against the latent bradyzoite stage found within tissue cysts. The mitochondrion of T. gondii is a validated drug target, and the clinically used drug atovaquone acts by inhibiting the mitochondrial electron transport chain (ETC) at the coenzyme Q:cytochrome c oxidoreductase ( b

A Suite of Eight Toxoplasma gondii Effectors Cooperates to Activate the Non-canonical NF-κB Pathway.

As a master of host-cell reprogramming, Toxoplasma gondii (T. gondii) tachyzoites manipulate diverse signaling networks to establish a niche permissive for long-term infection. While the parasite's subversion of canonical NF-κB signaling (p65/p50) is well established, how infection impacts the non-canonical NF-κB pathway has been largely unexplored. Here, we report that T. gondii infection induces robust nuclear accumulation of the non-canonical NF-κB subunits RelB and p52 in both human and murine cells. This activation follows a gradual kinetic profile and is conserved across both Type I and

Phosphoproteomic Analysis of GAP50-Deficient Parasites Reveals the Inner Membrane Complex Is Involved in Ion Regulation in Toxoplasma gondii.

The Alveolata group of organisms is characterized by a vesicular structure located beneath the plasma membrane. In apicomplexa, this structure is known as the inner membrane complex (IMC). The IMC acts as a scaffold during the budding of the daughter parasite and is utilized as a rigid base for the glideosome. In this study, we discovered that the phosphatase activity of GAP50 is crucial for its function in the biogenesis of the IMC. Through further phosphoproteomic analysis of parasites lacking GAP50, we identified that an NIPA family transporter is localized in the IMC. Knocking out TgNIPA1
