Myhre syndrome is a rare genetic disorder caused by pathogenic variants in SMAD4. The neurocognitive profile may include intellectual disability and developmental delay across a wide spectrum of domains. Four individuals with Myhre syndrome and psychosis and/or schizophrenia have been reported. We discuss three additional individuals (one briefly mentioned in a review of patients with Myhre syndrome) with SMAD4 pathogenic variants at codon Arg496Cys (two cases) and codon Ile500Val (one case). All had social challenges and variable developmental delays. They presented with acute-onset psychosis

Myhre syndrome (MYHRS) is an ultra-rare, progressive multisystem disorder caused by recurrent heterozygous missense variants in the SMAD4 gene, a central mediator of TGF-β and BMP signaling. Skeletal abnormalities-including postnatal short stature, brachydactyly, thickened calvarium, and craniofacial dysmorphism-are cardinal features, often accompanied by joint contractures and progressive soft tissue fibrosis. Extensive cellular and genetic evidence supports a gain-of-function (GoF) mechanism wherein mutant SMAD4 displays increased protein stability and prolonged nuclear localization, enhanci

Caudal regression syndrome (CRS) refers to a spectrum of congenital structural defects of the caudal region. We present a case of caudal regression syndrome with the objective of correlating the symptoms of caudal regression syndrome with the detailed imaging findings. A 05-year-old boy who was admitted to Mymensingh Medical College Hospital in August 2025 , presented with a post-urethroplasty state of hypospadias, complaining of urinary incontinence through the normal meatus and a urethrocutaneous fistula. He had dribbling of urine through hypospadias and deformed feet since birth. He also de

Biomolecular interactions are central to many essential cellular processes, but RNA-containing complexes remain challenging to resolve structurally, even as experimental methods and AI-based prediction have expanded structural coverage. Tools for the integrated analysis of interfaces within such complexes remain limited. We present ProNA3D, a tool that provides a unified platform for analyzing protein-nucleic acid and nucleic acid-only complexes, bridging the gap between structure prediction and functional interpretation. ProNA3D supports both experimental and computationally predicted structu

Human epidermal growth factor receptor 2 (HER2) is an established therapeutic target in multiple solid tumors, particularly breast and gastric cancers. Significant advancements have been made in the development of HER2-targeted therapies, including monoclonal antibodies, tyrosine kinase inhibitors, antibody-drug conjugates (ADC), and novel bispecific antibodies. These agents have revolutionized the treatment landscape for HER2-positive metastatic cancers, resulting in improved progression-free and overall survival, and quality of life for patients. Beyond breast and gastric cancers, HER2 expre

BACKGROUND: Unilateral mandibular hypoplasia affects not only the involved side but also the contralateral hemimandible. Thus, treatment must address bilateral deformities. The authors present a novel bilateral CAD/CAM titanium onlay implant design for unilateral mandibular hypoplasia and evaluate outcomes using objective and patient-reported measures.
PATIENTS AND METHODS: Thin-slice CT scans were processed with Mimics® software. The unaffected hemimandible was repositioned to the midline using the TMJ as a pivot, and its mirror image was used to create the contralateral side. Bilateral cust

Guided bone regeneration (GBR) is a critical technique in craniomaxillofacial surgery, significantly advancing the management of bone defects resulting from trauma, tumor resection, congenital malformations, and insufficient bone volume for dental implants. Autologous GBR capitalizes on the body's inherent regenerative capabilities in conjunction with strategic interventions, benefiting from enhanced biocompatibility and integration with osteogenic processes. This review delineates recent advancements in autologous GBR within the context of craniomaxillofacial surgery. First, it elucidates the

Congenital Radio-Ulnar Synostosis (CRUS) causes difficulty in forearm rotation, which is treated using osteotomy. Effective preoperative planning of osteotomy requires automatic and objective quantification of deformity angles. In this paper, an automatic Congenital Radio-Ulnar Synostosis deformity evaluation method (CRUS-DE) is proposed. Initially, a method using threshold-layer tracking (TLT) and the segment anything model (SAM) is designed to recognize and segment the forearm from CT images. Subsequently, the Gaussian Process Morphable Model, in conjunction with refinement based on anatomic

Syndactyly, a congenital hand deformity caused by failed phalangeal separation, often requires surgical correction to restore function and aesthetics. Traditional dorsal rectangular flap techniques involve skin grafting, increasing the risk of web creep and residual deformity. This study evaluates islanded perforator-based flaps, which offer better vascularization for improved outcomes. Preoperative Doppler ultrasound was used for vascular mapping to enhance surgical precision. This prospective observational study analyzed 31 webspaces in 26 patients undergoing syndactyly release. BACKGROUND: Split-hand/foot malformations (SHFM) have both dominant and recessive inheritance patterns, but the autosomal recessive forms (SHFM1D and SHFM6) are much rarer and often present with more severe and asymmetrical limb defects compared to the dominant forms. This study aimed to investigate the genetic basis of SHFM in patients presenting with limb anomalies and associated features, contributing to the understanding of autosomal recessive SHFM subtypes (SHFM1D and SHFM6).
METHODS: Clinical and radiological assessments were performed on the patients, followed by exome sequencing and s
