INTRODUCTION AND IMPORTANCE: Congenital hallux varus (CHV) is a very rare foot deformity, defined by the medial deviation of the great toe at the metatarsophalangeal joint. It can appear in isolation or with other anomalies, such as polydactyly, metatarsus adductus, or a longitudinal epiphyseal bracket. However, an association with a ventricular septal defect (VSD) has not been described before. Early surgical correction is essential to restore function, improve cosmetic appearance, and support normal motor development.

BACKGROUND: The Wassel classification for thumb polydactyly relies on radiographs; however, invisible unossified cartilage in young children can lead to misclassification and suboptimal surgical planning. This study evaluated whether ultrasound improves the accuracy of polydactyly classification compared with radiographs.
MATERIALS AND METHODS: This prospective study included 31 cases of radial polydactyly initially classified as Wassel type IV or type II based on radiographs. All patients underwent preoperative ultrasound to identify potential cartilaginous connections between the proximal a

Postaxial polydactyly of the foot is a common form of digital duplication that often causes difficulty with footwear, metatarsalgia, and cosmetic concerns. Although it may present as an isolated anomaly, it can also be associated with genetic conditions. Surgical excision is the primary intervention, with generally excellent outcomes. A 5-year-old male presented with postaxial polydactyly of the left foot, characterized by an accessory articulating osseous structure associated with the fifth ray. The main complaints were difficulty with footwear and cosmetic appearance. Radiographic evaluation

PHF1 and PHF19 are key accessory components of polycomb repressive complex 2 (PRC2) which is responsible for trimethylation at histone H3 lysine 27, ultimately leading to gene repression. PHF1 and 19 are known for their methyl-lysine reader Tudor domains that recognize methylated lysine 36 on H3, and they have also been implicated as drivers of a range of cancers, including multiple myeloma. Here, we describe the first small-molecule antagonist of the Tudor domains of PHF1 and 19. We screened an internal library of ∼ 1 K compounds against the Tudor domains using a TR-FRET assay. Upon validatio

Considerable advances have been made in our understanding of Bardet-Biedl syndrome (BBS), particularly in its core clinical features and molecular genetics, warranting an update to the existing diagnostic criteria framework. Using a rigorous, evidence-based, and consensus-driven process, a multidisciplinary group of international experts and patient-led organizations developed an updated diagnostic algorithm. This algorithm provides practical, updated guidance for clinicians, including a pathway for accurately incorporating genetic findings into the diagnostic process. We recommend that a clin

BACKGROUND: Anemia continues to affect one-third of the global population and is one of the most common reasons for large-scale morbidity and mortality especially among women. The importance of iron-rich diet has always been the backbone of preventing iron deficiency anemia (IDA) in vulnerable age groups followed by oral iron therapy and parenteral iron therapy as the next options in management of iron deficiency.
OBJECTIVE: Objective of this survey was to assess the knowledge, attitude and practices of obstetricians and gynaecologists relevant to anemia in pregnancy and identify the practice

Floating-Harbor syndrome (FLHS) is a rare neurodevelopmental and skeletal disorder caused by truncating variants in exons 33 and 34 of the SRCAP gene. It is characterized by distinctive facial features, delayed bone age, short stature, and moderate intellectual disability. While digital anomalies have been reported in approximately half of the more than 100 known cases, the phenotypic spectrum continues to expand. Here, we describe a family in which two individuals were identified with FLHS. Both the proband and her mother presented with typical manifestations, including classic facial dysmorp

BACKGROUND: Bardet-Biedl syndrome is a pleiotropic ciliopathy marked by retinal degeneration, obesity, polydactyly, renal and reproductive anomalies, and cognitive impairment. BBS1, the most frequently mutated gene in Bardet-Biedl syndrome, encodes a key component of the BBSome complex, which is essential for ciliary membrane trafficking. Although BBS1 is known to be essential for proper BBSome function, the effects of disease-associated BBS1 variants on its activity remain incompletely understood.
METHODS: In this study, we examined how patient-derived BBS1 mutations affect BBSome integrity

BACKGROUND AND PURPOSE: Several European countries offer nationwide prenatal ultrasound screening for fetal anomalies. Few studies have examined prenatal detection of upper limb anomalies, and existing reports originate exclusively from high‑volume centers outside Europe. We aimed to assess the timing of diagnosis in patients with all Oberg-Manske-Tonkin diagnoses except tumorous dysplasias referred to the largest specialized unit in Norway, investigate prenatal detection rates, and explore associations between patient‑ and hospital‑related factors and prenatal detection.
METHODS: We extracte

Background/Objectives: Bardet-Biedl syndrome (BBS) is a rare autosomal recessive ciliopathy caused by variants in genes encoding components of the BBSome complex. Interpretation of rare variants in the BBS7 gene remains challenging, particularly for variants of uncertain significance. This study aimed to provide clinical and molecular genetic characterization of two siblings with a BBS phenotype harboring compound heterozygous variants in BBS7, including a previously undescribed missense variant. Methods: Two siblings (aged 11 and 8 years) presenting with obesity, postaxial polydactyly, retina

BACKGROUND: Reconstruction of elbow extension is an important aim after brachial plexus injury. In extended partial brachial plexus injuries affecting elbow extension, a variety of nerve transfer techniques have been described to neurotize triceps. Spinal accessory, ulnar fascicles, axillary branches, intercostal and phrenic nerves have all been reported. These donors have mostly been transferred to triceps brachii nerves; however, more proximal transfers have also been reported. Results with these techniques have been inconsistent. In extensive partial brachial plexus palsy, radial fascicular
