Case Report: Congenital absence of the fifth metacarpal with polydactyly and syndactyly.

Congenital hand malformations encompass various types, with polydactyly and syndactyly being common. However, congenital absence of the metacarpal is rarely reported, and literature on this condition remains limited. To date, no cases of congenital absence of the fifth metacarpal combined with polydactyly and syndactyly have been documented. Here, we present a case of a 7-month-old female infant with congenital absence of the fifth metacarpal accompanied by polydactyly and syndactyly. The intraoperative management strategy is described, and early postoperative outcomes are evaluated. This case

Expanding the Phenotypic and Genotypic Spectrum of Postaxial Polydactyly: A Study of Four Consanguineous Pakistani Families.

Background: Postaxial polydactyly (PAP) is characterized by the development of extra digits at the fifth finger. It can occur as an isolated disease or a part of a syndrome. The genetic basis of nonsyndromic PAP has been linked to sequence variants in different genes. The aim of the present study was to identify the causative genetic variants in four Pakistani families demonstrating PAP. Methods: Causative genetic variants were identified using whole-exome sequencing and microsatellite mapping, followed by validation through Sanger sequencing. Further analysis was carried out through conservat

Phenotypic Spectrum of KATNIP-Associated Joubert Syndrome: Possible Association with Esophageal Atresia and Review of the Literature.

Background: Joubert syndrome (JS) is a multi-systemic ciliopathy, characterized by intellectual disability and congenital anomalies involving the brain, kidney, heart, and eye. Even if clinical presentation is variable, most authors consider a brain abnormality known as the molar tooth sign (MTS) as mandatory for diagnosis. About 40 genes were identified to be associated with JS, usually with an autosomal recessive pattern. KATNIP variants represent a rare cause of JS; only six families were previously reported. Methods: We performed exome sequencing in a child with a syndromic phenotype, desc

Case Report: A case of Culler-Jones syndrome caused by GLI2 gene mutation.

Culler-Jones syndrome (CJS) (OMIM: 615849) is a rare genetic condition characterized by considerable phenotypic variability. This case reports a 5-day-old male neonate who presented with postaxial polydactyly, growth restriction, and recurrent epileptic seizures. A thorough clinical workup, including laboratory investigations, imaging, and genetic analysis, resulted in a confirmed diagnosis of Culler-Jones syndrome. Peripheral blood samples collected from the proband and his parents were used for DNA extraction. Whole-exome sequencing (WES) identified a heterozygous nonsense variant in the GLI

A Systematic Review of Digital Deformities in Pediatric Patients and Their Podiatric Treatments.

BACKGROUND AND OBJECTIVES: Digital deformities in pediatrics are one of the reasons why parents go to podiatry clinics with their children. There is a great diversity of digital alterations in the feet; the origin of the vast majority is genetic. Due to this diversity, different treatments are offered depending on deformity and age, ranging from monitoring progress and changing footwear to deformities that are directly evaluated for surgery. The main objective of this study is to describe and identify the different podiatric digital deformities present in pediatric patients, as well as the mos

A Rare Footprint: A Case Report of Isolated Pre-Axial Fully Developed Supernumerary Toe.

INTRODUCTION: Cleft hands and feet constitute a rare, congenital abnormality of limb bud development, manifesting as a cosmetically and, occasionally, a functional debility of the hands or feet. Patients often pursue expert advice regarding the surgical reconstruction of their deformities, which may pose an ethical dilemma to the treating practitioner. CASE REPORT: We present a case series of a family of three with bilateral cleft hands and feet, highlighting the dissimilarities in their phenotypic presentation. The clinical pointers indicate a possible syndromic association of ectrodactyly,

A Rare Foot Duplication; Mirror Foot - Case Report.

Polydactyly is a congenital extremity malformation defined by the presence of additional digits. The mirror foot anomaly, regarded as a rare variant of preaxial polydactyly, is characterized by a mirror image duplication of the foot. Due to the variability of mirror foot patterns reported in the literature, defining the term "mirror foot" remains challenging. In this case study, we present the treatment of a five-year-old male patient with a mirror foot anomaly who was admitted to our outpatient clinic. There is no consensus on the optimal surgical management strategy, and very few cases have

Truncating mutations in BBS10 and BBS12 impair proteostasis and ciliary architecture in Bardet-Biedl Syndrome.

Bardet-Biedl Syndrome (BBS) is a rare autosomal recessive ciliopathy characterized by genetic heterogeneity. Despite significant progress in understanding the BBSome-coding genes associated with ciliopathies, the pathogenesis linked to mutations in chaperonin-coding genes (BBS6, BBS10, and BBS12) remains poorly defined. This study aims to confirm the genetic diagnosis of BBS and elucidate the pathological mechanisms in causative genes of BBS10 and BBS12. Clinical evaluations were performed on BBS patients, followed by targeted next-generation sequencing (NGS) to identify disease-causing varian
