A Case of Polydactyly in a Roe Deer (Capreolus capreolus) From Germany.

Polydactyly is a rare anomaly that has been described in many vertebrate species including several deer species. The case described in this communication is believed to be only the second documented example of additional metatarsae in roe deer (Capreolus capreolus). In addition to normally developed digits III and IV and the dewclaws, a right hind foot of an adult female from Germany had two additional digits lateral to digit V. Both originated from additional metatarsal elements. This case offers significant insights into the developmental plasticity of the cervid limb and extends the morphol

Molecular and clinical heterogeneity in an Iranian case series of Joubert syndrome.

BACKGROUND: Joubert syndrome (JS) is a rare neurodevelopmental ciliopathy characterized by a distinctive midbrain-hindbrain malformation, manifested by hypotonia, ataxia, developmental delay, and variable multisystem involvement. The condition exhibits marked clinical and genetic heterogeneity, with over 40 genes implicated to date. However, the mutational spectrum and phenotypic presentation of JS in Middle Eastern populations, where consanguinity is prevalent, remain poorly characterized. METHODS: We retrospectively reviewed whole-exome sequencing (WES) data from patients referred to the Co

A Novel MID1 Mutation Identified in a Patient With Craniofacial Anomalies and X-Linked Intellectual Disability.

Genetic variations in MID1 were initially identified as the cause of X-linked Opitz G/BBB syndrome, which is characterized by hypertelorism, hypospadias, and a high incidence of intellectual disability. However, increasing evidence has shown that MID1 mutations are associated with a broader spectrum of phenotypes, and the genotype-phenotype correlation remains unclear. In this study, the authors report a 4-year-old boy presenting with congenital unilateral nostril hypoplasia, preaxial polydactyly, hemispheric brain asymmetry, and moderate intellectual disability. Whole-exome sequencing identif

Anatomic Classifications and Functional Outcomes in Preaxial Polydactyly of the Hand.

BACKGROUND: Preaxial polydactyly is a common congenital hand anomaly with substantial anatomic variability. Although several classification systems exist, the relationship between anatomic subclassification and postoperative outcomes remains incompletely defined. This study evaluated clinical and radiographic outcomes after surgical reconstruction, compared associations of the Wassel-Flatt (WF) and JK classifications with outcomes, and assessed the Japanese Society for Surgery of the Hand (JSSH) and Rotterdam scoring systems. METHODS: We retrospectively reviewed 58 patients (59 thumbs) treate

When Fetal Anomalies Lead to Prognostic Clarification: An Underrecognized Application of Exome Sequencing.

OBJECTIVES: To describe a cohort of fetuses with abnormal prenatal findings in whom exome sequencing (ES) identified genetic diagnoses that refined prognostic assessment and reduced prognostic uncertainty. METHODS: We retrospectively reviewed all fetal ES studies performed at our center between 2017 and 2025 for structural or biochemical abnormalities. Cases with a definitive molecular diagnosis were identified, and those with predicted mild, isolated, transient, or treatable phenotypes were classified as having comparatively favorable prognostic implications. Clinical findings and expected p

Polydactyly.

Polydactyly of the hand is among the most common congenital anomalies of the upper extremity and encompasses a broad range of anatomic and functional presentations. It is characterized by a duplicated digit, which may occur as an isolated finding or in association with genetic syndromes. Based on the location and characteristics of the duplication, polydactyly is commonly classified as preaxial, postaxial, or central. Proper evaluation requires an understanding of limb embryology, genetic influences, and classification systems, as these factors directly inform surgical planning. Treatment stra

Parental Aesthetic Anxiety, Disability Aesthetics, and the Ethics of Surgical Normalization in Congenital Hand Differences.

Children born with congenital hand differences (syndactyly, fusion of adjacent digits; polydactyly, duplication of digits; radial club hand, partial or complete absence of the radius; and symbrachydactyly, short, webbed, or absent fingers) occupy a unique and underexamined position at the intersection of pediatric surgery and disability studies. While surgical literature has meticulously documented operative techniques, functional outcomes, and complication profiles, it has almost entirely neglected the aesthetic and cultural dimensions of the decision to operate. This narrative review address

Familial multiple fetal cerebral arteriovenous malformations: a case report of maternal genetic susceptibility and fetal manifestation.

INTRODUCTION: Cerebral arteriovenous malformations (AVMs) are rare vascular anomalies that can present significant clinical challenges, especially when occurring in multiple sites. Hereditary hemorrhagic telangiectasia (HHT), a genetic disorder may be caused by mutations in genes such as ENG (encoding endoglin) or ACVRL1 (ALK1), and less commonly SMAD4, is one condition that predisposes individuals to multiple AVMs. This case report investigates the role of maternal genetic susceptibility in the fetal manifestation of multiple cerebral AVMs. CASE PRESENTATION: A multiparous woman with emboliz

Comparison of intra-articular injectable polydactyly-derived chondrocyte sheet-minis with direct transplantation of intact polydactyly-derived chondrocyte sheets in a rat full-thickness cartilage defect model.

INTRODUCTION: Polydactyly-derived chondrocyte (PD) sheets are being developed as an allogeneic cell sheet therapy for cartilage defects associated with knee osteoarthritis. Although intact PD sheets can promote hyaline cartilage repair, transplantation requires invasive open procedures. Injectable fragments of PD sheets (PD sheet-minis) may reduce invasiveness. However, their in vivo repair capacity after intra-articular administration remains unclear. METHODS: PD cells were isolated from discarded cartilage tissue obtained during polydactyly surgery. PD sheets and PD sheet-minis were fabrica

Meckel-Gruber syndrome: a rare and fatal congenital disorder (case report).

Meckel-Gruber syndrome is a rare congenital disorder characterized by multiple malformations. It transmits via a recessive autosomal mode. It is characterized by an occipital encephalocele, polydactyly, and polycystic renal dysplasia. The diagnosis could be established via ultrasound. In fact, it is actually the key method for the early screening of this lethal malformation with at least two of its main characteristics. However, the diagnosis is confirmed by karyotype analysis. Herein, we present a case of Meckel-Gruber syndrome diagnosed in a fetus from a consanguineous marriage in a 40-year-

A Case of Short-Rib Thoracic Dysplasia: Diagnostic Clues in Early Pregnancy and Genetic Analysis.

To investigate the diagnostic clues and genetic etiology of short-rib thoracic dysplasia (SRTD) in early pregnancy, we report a case of a fetus suspected of having SRTD based on ultrasound findings at 13 weeks of gestation. Prenatal genetic testing revealed a pathogenic variant in the DYNC2H1 gene, and post-abortion examination of the specimen along with X-ray findings confirmed the diagnosis as short-rib thoracic dysplasia 3 (SRTD3; OMIM #613091). Such cases are rarely reported in the early gestational period. The detection and subsequent confirmation in this case at 13 weeks demonstrate that

Biallelic variants in DNAH10 are associated with skeletal developmental abnormalities and ciliary dysfunction.

BACKGROUND: Primary cilia are essential for skeletal development by coordinating key signalling pathways in osteoblasts and chondrocytes. While pathogenic variants in approximately 40 genes have been linked to skeletal ciliopathies, additional causative genes remain to be identified. Although DNAH10 is well characterised in motile cilia, its role in primary cilia remains unclear. METHODS: Whole-exome sequencing was performed in two individuals with skeletal developmental abnormalities and identified biallelic DNAH10 variants. In vitro assays were used to assess the effects of these variants o

Delayed diagnosis of Townes-Brocks syndrome accompanied with kidney failure.

Townes-Brocks syndrome (TBS) is a rare autosomal dominant disorder typically characterized by the triad of anorectal malformations, external ear anomalies and hand malformations such as preaxial polydactyly, caused by mutations in the SALL1 gene. Kidney involvement, although less common, can progress to end-stage kidney failure. Early recognition of the characteristic features, particularly those related to SALL1, is crucial for accurate diagnosis, management, and genetic counseling. Here, we present a Turkish family with TBS in which the diagnosis was delayed due to the absence of the classic
