We report a case of acrocephalosyndactyly, Pfeiffer syndrome type 1 with a mutation in FGFR2 c.758C>G (p.Ser253Trp) in a newborn with mild midfacial hypoplasia, significant brachydactyly and syndactyly in the hands and feet. One of the hallmark features of Pfeiffer syndrome is webbing or fusion (syndactyly) of the fingers and toes, which can vary in severity affecting both hands and feet. This variable expressivity of Pfeiffer syndrome makes classification of the condition challenging. Diagnosis was confirmed by genetic testing. Imaging investigations, clinical observation and physical examina

Introduction: Fraser syndrome (FS) is a rare autosomal recessive disorder. However, the clinical presentation remains variable. Diagnosis is based on a series of major and minor clinical criteria that can be supported by genetic tests. Prenatal diagnosis remains challenging. Methods: Herein, we reported a case of Fraser syndrome that was missed by ultrasound and diagnosed late at birth. The newborn presented with cryptophthalmos-syndactyly syndrome and absence of the right kidney. Based on a literature review of articles from the past 20 years, the authors found 40 cases, including indexed cas

Background: Congenital syndactyly is one of the most common congenital hand anomalies (1 in 3,000 live births). The ideal surgical technique must rebuild a satisfactory web space, restore digit abduction and avoid secondary contracture. The dorsal omega flap reduces the need for skin graft, thereby reducing web creep and contracture. There is a gap in literature on patient reported outcomes regarding the dorsal omega flap. We conducted a cross-sectional study to assess the outcomes patients operated with this technique. The aim of this study is to assess quality of life using Patient Reported

The Oberg, Manske and Tonkin (OMT) Classification of Congenital Hand and Upper Limb Anomalies was adopted as the official IFSSH classification in 2014 with recommendations for 3-yearly reviews and updates. This report examined the evidence and feedback from the scientific community to see whether changes to the 2020 OMT should be made. The Committee concluded that no current changes are required, but highlighted a number of areas where further research and discussions are needed. These areas include the conditions of symbrachydactyly vs transverse arrest, the ongoing challenge with classifying

Poland Anomaly is a rare congenital disorder typically characterized by hypoplasia or agenesis of pectoral muscle with or without ipsilateral limb hypoplasia. The association of central nervous system malformation with Poland Anomaly has been rarely reported and includes craniofacial dysplasia, microcephaly, and Dandy-Walker malformation. However, no anomalies in neural tube closure have been described in association with Poland Anomaly. Atretic cephalocele is another rare diagnosis, which is an involuted congenital herniation of intracranial contents through defects in the skull. Herein, we p

INTRODUCTION: Lenz-Majewski syndrome (LMS) is an ultra-rare congenital disorder with progressive skeletal dysplasia, cutis laxa, and intellectual disability, typically caused by pathogenic variants in the PTDSS1 gene.
METHODS: Our patient with multiple malformations and developmental delay who was treated at the Women and Children's Hospital of Ningbo University in September 2023 was selected as the research subject. Whole exome sequencing (WES) technology was used to test the child, and Sanger sequencing verification and pathogenicity analysis were carried out for the suspected variations.

We report a rare case of partial prenatal diagnosis of congenital lipomatous overgrowth, vascular malformations, epidermal nevi, and skeletal anomalies (CLOVES) syndrome in a fetus presenting extensive dorsal lymphatic malformation, bilateral polydactyly and syndactyly, hypertrophy on the left foot, and suspected cryptorchidism. Amniocentesis with comparative genomic hybridization (CGH) and trio-exome sequencing did not reveal any pathogenic variant. Postnatal clinical examination and imaging confirmed the malformations, including a multilocular macrocystic lymphatic malformation with retroper

Poland Anomaly (PA) represents pectoral muscle hypoplasia in combination with various forms of hand anomalies. We report a case of PA with pectoral hypoplasia, an upper limb deficiency and an accessory extremity/digit on the ipsilateral thoracic wall (thoracomelia). To our knowledge, this is the first reported case of thoracomelia in PA.

Congenital hand malformations encompass various types, with polydactyly and syndactyly being common. However, congenital absence of the metacarpal is rarely reported, and literature on this condition remains limited. To date, no cases of congenital absence of the fifth metacarpal combined with polydactyly and syndactyly have been documented. Here, we present a case of a 7-month-old female infant with congenital absence of the fifth metacarpal accompanied by polydactyly and syndactyly. The intraoperative management strategy is described, and early postoperative outcomes are evaluated. This case

Bardet-Biedl syndrome is a rare, pleiotropic genetic disorder. Despite advances in genetic testing, the diagnosis of Bardet-Biedl syndrome remains primarily clinical, particularly in low-resource settings. Clinical features are classified as either primary or secondary. The primary features include retinal dystrophy, central obesity, renal abnormalities, male hypogonadism, and learning disabilities. Secondary characteristics include developmental delay, dental defects, diabetes mellitus, speech disorders, brachydactyly/syndactyly, and ocular abnormalities such as nystagmus, strabismus, and ast

Syndactyly is a common inherited and clinically heterogeneous malformation which can be syndromic or non-syndromic. It also varies phenotypically between the families. In this study, we present twins who experienced bilateral syndactyly and underwent reconstruction using the Gilbert method. These female twin toddlers were referred to Hazrat Fatemeh Clinic of Hand Surgery in Tehran, Iran, in 2022 with bilateral syndactyly of the middle and ring fingers. In both, fingers were fused along their entire length. One of them underwent the separation of the middle finger and ring finger of the right h

PURPOSE: First web space syndactyly presents a considerable reconstructive challenge, and because of the rarity and complexity of the condition, there is no consensus on the optimal approach to its reconstruction. This study therefore describes our experience with congenital first web space syndactyly, providing an overview of reconstructive techniques and outcomes.
METHODS: Sixty-nine cases (55 simple and 14 complex/complicated), from 2007 to 2022, were analyzed with descriptive statistics.
RESULTS: Dorsal commissural flaps were the initial operative approach in 100% of the complex/complica

Objectives: Monitoring pregnancies is essential for community well-being. However, not all pregnancies progress normally, and some require termination. The objective was to emphasize the importance of trust in the doctor-patient relationship during this challenging time for expectant parents. Case report: During fetal morphology examination, parents were warned of a poor fetal prognosis, prompting a request for pregnancy termination. They consulted another specialist, who reassured them that the fetus appeared normal, though slightly hypotrophic. The child was born at 35 weeks gestational age

BACKGROUND: Arrhythmogenic right ventricular cardiomyopathy (ARVC) is mainly caused by desmosomal variants. Nondesmosomal ARVC is rare.
CASE SUMMARY: A 31-year-old athlete experienced sudden cardiac death. Autopsy revealed ARVC and syndactyly. Genetic testing identified a heterozygous pathogenic KCNQ1 variant associated with long QT syndrome, and a heterozygous likely pathogenic GJA1 variant encoding connexin-43, a gap junction protein. No relative had ARVC or prolonged QTc interval; however, GJA1-positive relatives had either syndactyly or a gait disorder; both of these pathologies are assoc

INTRODUCTION: Heterozygous variants in MAP3K20 have been implicated in a recently identified syndromic form of ectodermal dysplasia, distinguished by a unique combination of ectrodactyly, craniosynostosis, bilateral sensorineural hearing loss, and skeletal anomalies such as transverse limb defect, and brachydactyly.
CASE PRESENTATION: We present an 11-year-old male with ectrodactyly, ectodermal dysplasia, bilateral sensorineural hearing loss, and cutaneous syndactyly in both hands. A de novo heterozygous variant, c.837_839del p.(Asn279del), in MAP3K20 was identified in his whole exome sequenc

INTRODUCTION: We assessed the long-term quality of life, cosmesis and pain after childhood syndactyly separation.
METHODS: A retrospective review identified 155 patients (⩾10-year follow-up) of whom 76 participated with a median follow-up period of 18.2 years (IQR 12.4 to 38.3). Patient-reported outcomes were assessed using the Quick Disabilities of the Arm, Shoulder, and Hand questionnaire, EuroQol-5 Dimension-5 Level, and visual analogue scales (0-10) for function, cosmesis and pain.
RESULTS: Uncomplicated cases had significantly better patient reported outcomes (0 vs. 11.4 and 1.0 vs. 0.8

OBJECTIVE: To examine the association between the introduction of the fetal anomaly scans in the Netherlands and termination of pregnancy (TOP) in cases of prenatally detected upper limb anomalies.
METHODS: We conducted a retrospective study among prenatally detected upper limb anomalies between 2000 and 2023. Anomalies were categorized as reduction defects, syndactyly, or polydactyly, and classified as isolated or non-isolated. We analyzed TOP rates across three periods (2000-2006, 2007-August 2021, September 2021-2023), including an interrupted time series (ITS) analysis to assess the impac
