INTRODUCTION: Mutations in TCF12 are identified in 10-20% of coronal craniosynostoses negative for TWIST1 and FGFR mutations, with autosomal dominant inheritance, incomplete penetrance, and wide phenotypic variability. The recognized phenotypic spectrum includes coronal synostosis, craniofacial dysmorphism, ptosis, strabismus, mild syndactyly, and neurocognitive alterations. However, axial skeletal segmentation anomalies have not been previously reported in association with TCF12 mutations.
CASE PRESENTATION: We present the case of a 9-year-old male with right unicoronal craniosynostosis trea

BACKGROUND: Fibroblast growth factor receptors (FGFRs) play a crucial role in tissue homeostasis and organ development by regulating cellular processes, including proliferation, differentiation, and survival. Dysregulation of FGFRs contributes to developmental disorders and carcinogenesis. As membrane-bound receptors, they represent promising targets for therapeutic intervention and drug development.
METHODS: This study employed a systematic in silico analysis of publicly available phosphoproteomics datasets to provide a comprehensive overview of the phosphorylation regulatory network of the

Congenital upper limb differences occur in approximately 20-30 per 10,000 live births. These differences frequently prompt reconstructive surgery and may present in isolation or as markers of systemic syndromes. Embryologically, limb development depends on coordinated signaling along the proximal-distal (apical ectodermal ridge/fibroblast growth factor [FGF]), radial-ulnar (zone of polarizing activity/sonic hedgehog), and dorsal-ventral (wingless-related integration site 7) axes, whose disruptions lead to specific phenotypic patterns. Among upper limb differences, those with a well-established

Synpolydactyly is a rare limb deformity characterized by a unique combination of syndactyly and polydactyly. Synpolydactyly has an autosomal dominant mode of inheritance often with reduced penetrance. Variants in HOXD13, FBLN1, GLI3, and TTC30B have been reported to underlie the etiology of synpolydactyly. Here, we describe a three-generational Pakistani family segregating a form of synpolydactyly which has not previously been reported that includes, severe hand involvement characterized by bilateral syndactyly of the third, fourth, and fifth fingers, accompanied by preaxial polydactyly in all

BACKGROUND: Trifid epiglottis (TE) is an extremely rare congenital laryngeal anomaly. To the best of our knowledge, only two cases have previously been reported in the literature. Both of them have been incidentally detected and associated with polydactyly.
CASE PRESENTATION: We report the third known case of TE in a two-day-old female neonate who was admitted to the emergency department due to abdominal distension, oliguria, and poor feeding within the first 24 h of life. Examination revealed stridor during crying, left postaxial polydactyly, right syndactyly, and an anteriorly displaced anu

BACKGROUND: Apert syndrome is a rare congenital malformation caused by a mutation in the fibroblast growth factor receptor 2 (FGFR2) gene. Characteristic imaging findings include bilateral premature closure of the coronal suture, midfacial hypoplasia, and symmetrical syndactyly of the fingers and toes. These features may be accompanied by lateral ventricle dilation, absence of the corpus callosum, cervical vertebral fusion, and other anomalies. However, in the second trimester, syndactyly and extracranial anomalies may be the only detectable signs, which complicate early diagnosis. The purpose

Streeter's dysplasia, also known as amniotic band syndrome (ABS), is a rare congenital condition characterized by a wide spectrum of malformations, deformities, and, in severe cases, limb amputations. Other manifestations associated with ABS include clubfoot, distal finger or toe hypoplasia, and syndactyly. Although several theories have been proposed, the exact etiology of this syndrome remains unclear. In this case report, we present the 14-year follow-up of a patient diagnosed with ABS at birth who has been managed through a multidisciplinary approach since infancy. This report highlights t

Cranial sutures are essential for skull growth and tissue homeostasis. Among them, the coronal suture is most frequently affected in syndromic craniosynostosis, yet the mechanisms underlying this preferential involvement remain unclear. Here, we show that the coronal suture mesenchyme undergoes a postnatal lineage transition from mesodermal to cranial neural crest origin, facilitated by dural cell migration into the suture. Mechanistically, this migration is regulated by suture TGFβ signals to Tgfbr2+ dural cells. Loss of dural Tgfbr2 impairs this cell migration into the suture, reduces the Gl

Floating-Harbor syndrome (FLHS) is a rare neurodevelopmental and skeletal disorder caused by truncating variants in exons 33 and 34 of the SRCAP gene. It is characterized by distinctive facial features, delayed bone age, short stature, and moderate intellectual disability. While digital anomalies have been reported in approximately half of the more than 100 known cases, the phenotypic spectrum continues to expand. Here, we describe a family in which two individuals were identified with FLHS. Both the proband and her mother presented with typical manifestations, including classic facial dysmorp

During the past decade, advancements in CryoEM have afforded significant insight into transmembrane regions of ion channels. Portions of cytosolic amino acids have been structurally and biophysically characterized; however, understanding structural relationships and how components integrate into a functional ion channel required clarification. Here, we have calculated an all-atom model for the complete sequence of the cardiac voltage-gated calcium ion channel (CaV1.2 alpha-1C and beta-3 subunits) in a lipid bilayer with explicit salt and water. Four one-microsecond molecular dynamics (MD) simu

BACKGROUND: Multiple finger skin defects with exposed bone or tendon are challenging to reconstruct while preserving finger independence. Traditional approaches require multiple operations, secondary finger separation, or multiple vascular anastomoses. We systematically evaluated the use of free poly-foliate perforator flaps for one-stage reconstruction of multiple finger defects and report the largest focused case series to date.
METHODS: Thirty-nine patients (27 men, 12 women; mean age 36.6 years) with multiple finger skin defects underwent reconstruction using free poly-foliate perforator

BACKGROUND: Holt-Oram syndrome, also known as atrio-digital syndrome, is a rare autosomal dominant genetic disorder primarily characterized by upper limb malformations and congenital heart defects, associated with mutations in the TBX5 gene. Diagnosis is typically based on clinical presentation, and the prognosis for affected individuals is closely related to the severity of cardiac involvement. Our study aims to highlight some key features of Holt-Oram syndrome to improve the level of clinical diagnosis.
METHODS: We collected 11 patients with clinical features strongly suggestive of Holt-Ora

BACKGROUND: Polydactyly and syndactyly are common congenital hand malformations in children, with surgical correction as the primary treatment. Postoperative pain in children can trigger pain catastrophizing in parents, which may impair family care quality and children's rehabilitation. This study aimed to evaluate the current status of pain catastrophizing and its associated factors in parents of children after polydactyly or syndactyly surgery.
METHOD: A cross-sectional survey was conducted from February 2024 to October 2025, enrolling parents of children who underwent polydactyly or syndac

Oculodentodigital dysplasia (ODDD) is a rare genetic disorder caused by mutations in the GJA1 gene, which encodes connexin 43 (Cx43). The condition presents with a broad phenotypic spectrum including ocular, dental, craniofacial, neurological, and skeletal abnormalities. We report the case of a 24-year-old female patient with genetically confirmed ODDD, carrying the c.226C > T (p.Arg76Cys) variant in the GJA1 gene, who presented with compatible clinical signs. Ophthalmologic evaluation revealed epicanthal folds, microcorneas, nystagmus, hyperprolate corneal profiles, and corneal topography con
