Vasoproliferative Tumors of the Retina: Pathophysiology, Clinical Features, and Treatment Approaches.

BACKGROUND: Retinal vasoproliferative tumors (RVPTs) are rare, benign lesions appearing as elevated, pink masses in the peripheral retina. Initially considered acquired retinal capillary hemangioblastomas, RVPTs are now recognized as distinct entities, with idiopathic and secondary forms. Though primarily affecting individuals between 30 and 50 years of age, their pathogenesis remains under investigation. SUMMARY: Recent histopathological evidence suggests RVPTs have a predominantly glial rather than vascular origin. Clinically, RVPTs cause visual deterioration, floaters, and photopsia, often

Expiration Date of Extemporaneous Pyrimethamine Formulations for Treating Congenital Toxoplasmosis: Physicochemical and Microbiological Evaluation.

This study evaluated the stability of extemporaneous pyrimethamine oral suspensions prepared from commercial tablets using 85% sucrose or 70% sorbitol as vehicles. A validated HPLC method was employed to quantify pyrimethamine and assess its chemical stability. Physicochemical parameters including macroscopic appearance, pH, particle size, and drug content were monitored for 90 days at 5 and 25 °C. Both formulations maintained acceptable pH values and pyrimethamine content within pharmacopeial limits. Particle size increased over time, likely due to aggregation, while appearance remained uncha

Treatment of congenital toxoplasmosis with pyrimethamine combined with sulfadiazine in a Chinese neonate: a case report.

This work reports a case of neonatal congenital toxoplasmosis in which the standard regimen of pyrimethamine combined with sulfadiazine was administered under conditions of limited drug accessibility in China, with multidisciplinary collaboration and long-term follow-up. The infant was a male born at 36⁺⁶ weeks of gestation. Postnatal serological screening and cerebrospinal fluid (CSF) next-generation sequencing (NGS) confirmed the diagnosis of mildly symptomatic congenital toxoplasmosis. In the initial phase of treatment, due to the lack of routine access to standard therapeutic drugs in Chin

The Impact of Vitreoretinal Surgery in Patients with Uveitis: Current Strategies and Emerging Perspectives.

Uveitis constitutes a heterogeneous group of intraocular inflammatory pathologies, including both infectious and non-infectious aetiologies, often leading to substantial morbidity and permanent loss of vision in up to 20% of the affected cases. Visual impairment is most prominent in intermediate, posterior, or panuveitis and is commonly associated with cystoid macular oedema, epiretinal membranes, macular holes, and retinal detachment. In the context of uveitis, these complications arise as a result of recurrent flare-ups or chronic inflammation, contributing to cumulative ocular damage. Pars

Ocular Toxoplasmosis Infection Leading to Uveitis or Chorioretinal Lesions: A Systematic Review.

INTRODUCTION: Toxoplasma gondii (T. gondii) infections affect approximately 30% of the population worldwide. This systematic review of ocular sequelae from toxoplasmosis explores in greater depth the outcomes found in our previous scoping review. Uveitis and ocular lesions can be acute or recurrent following infection and result in long-term and often irreversible effects. METHODS: We followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines and used the Cochrane risk-of-bias tool to evaluate articles for inclusion. Data extraction included the frequency and p

Nosocomial oral myiasis caused by Chrysomya bezziana in Iran: A new case and review of human myiasis in the country.

Chrysomya bezziana can potentially cause myiasis, a condition in which fly larvae infest living tissue in humans and animals. Contributing factors to this rare ailment include poor oral hygiene, alcohol abuse, and infected wounds. Hospital-acquired myiasis, a rare type of myiasis, occurs in patients following hospital admission. Herein, we report a case of C. bezziana oral myiasis in an 89-year-old female patient hospitalized in northeastern Iran. In this article, we also present an updated review of reported human myiasis cases in Iran up to 2025. According to our findings, C. bezziana has be

An overview of risk factors for clinical toxoplasmosis.

Clinical toxoplasmosis is a rare but often fatal disease that impacts several medical fields, including obstetrics, ophthalmology, transplantation, oncology and the context of the AIDS pandemic. The prognosis for patients with toxoplasmosis largely depends on timely treatment, which makes early diagnosis a priority for clinicians. However, diagnosing toxoplasmosis is a significant challenge due to the lack of specific clinical symptoms. This issue is further complicated by the high seroprevalence of Toxoplasma in the general population, which is far higher than the incidence of the disease. Th

Is Toxoplasma gondii-secreted Protein With an Altered Thrombospondin Repeat (TgSPATR) a Potential Candidate for Immunisation? An Immunoinformatics-Based Analysis.

BACKGROUND: Toxoplasmosis is a widespread zoonotic disease that poses risks to pregnant women and immunocompromised individuals. Despite considerable efforts, no licensed vaccines are currently available for humans or animals. Rational vaccine design increasingly relies on immunoinformatics approaches to identify immunodominant epitopes and key immunological features. OBJECTIVES: This study aimed to characterise the Toxoplasma gondii-secreted protein with an altered thrombospondin repeat (TgSPATR) using immunoinformatics tools to evaluate its suitability as a vaccine candidate. METHODS: A co

Pathology and parasite distribution in mice challenged with Toxoplasma gondii from different geographical origins.

Toxoplasma gondii (T. gondii), a zoonotic parasite, can cause severe disease in warm-blooded animals. Pathological changes in murine tissues infected with different T. gondii isolates were studied to establish factors influencing lesion severity and parasite burden. In Study A, mice were orally inoculated with genotype #3, #6 or #8 oocysts. In Study B, mice were inoculated intraperitoneally with genotype #1, #3, #6, #13, #141 or #265 tachyzoites. Mice were euthanised serially and tissues processed for histopathology. In Study A, genotype #6 caused pathology in the liver, brain, lung, intestine

Protection Against Toxoplasma gondii Lethal ME49 Challenge Induced by Influenza Virus-like Particles Containing Dense Granule Protein 14.

Background/Objectives: Toxoplasma gondii (T. gondii) dense granule antigen 14 (GRA14) is a parasitophorous vacuole membrane protein that plays a critical role in the development of chronic-stage cysts. However, its potential as a vaccine antigen and long-term immunity have not been evaluated using a virus-like particle (VLP) platform. Methods: influenza matrix protein (M1)-based VLPs displaying GRA14 were generated. Female BALB/c mice were intranasally immunized with the VLP vaccine and orally challenged with lethal ME49 cysts either 10 weeks or 32 weeks after prime vaccination for short-term

Assessing the Toxoplasma Tachyzoite Cell Cycle Phases Using Fluorescent Ubiquitination-Based Cell Cycle Indicator.

Toxoplasma gondii is an apicomplexan parasite that infects a wide variety of eukaryotic hosts and causes toxoplasmosis. The cell cycle of T. gondii exhibits a distinct architecture and regulation that differ significantly from those observed in well-studied eukaryotic models. To better understand the tachyzoite cell cycle, we developed a fluorescent ubiquitination-based cell cycle indicator (FUCCI) system that enables real-time visualization and quantitative assessment of the different cell cycle phases via immunofluorescence microscopy. Quantitative immunofluorescence and live-cell imaging of

Public Health Implications of Latent Toxoplasmosis and Its Association With Type 2 Diabetes: A Case-Control Study in Qazvin, North-Western Iran.

INTRODUCTION: Toxoplasma gondii is a worldwide-distributed zoonotic parasite, causing latent infections in humans. Increasing evidence has suggested a possible link between toxoplasmosis and diabetes mellitus (DM), though data from Iran are limited. This study was conducted to determine the seroprevalence of latent T. gondii infection and its association with diabetes in Qazvin province, north-western Iran. MATERIALS AND METHODS: In this case-control study, 350 patients with type 2 diabetes mellitus and 350 non-diabetic controls were recruited from clinical laboratories in Qazvin province. An

Establishment of an ELISA for detecting oocyst-derived Toxoplasma gondii infections in sheep.

BACKGROUND: Ovine toxoplasmosis is a zoonotic disease that severely impacts the development of the sheep industry. The primary routes of Toxoplasma gondii infection in sheep are cyst infection and oocyst infection. However, current technologies are unable to distinguish between these two infection pathways. METHODS: In this study, we initially screened eight proteins that are highly specifically expressed during the oocyst stage. Through Western blot (WB) analysis, we identified a protein (TGME49_267410) that could serve as a diagnostic antigen. Subsequently, we optimized the conditions for a

Hemichorea-Hemiballismus As Presentation of Toxoplasma gondii Encephalitis in an Immunocompromised Host.

Toxoplasma gondii encephalitis is a common but serious opportunistic infection in people living with human immunodeficiency virus (PLHIV), particularly those with advanced disease and low CD4 counts. Its classical manifestations include fever, altered mental status, seizures and focal neurological signs. Movement disorders are a less common consequence, with hemichorea-hemiballismus (HCHB) representing a rare but striking manifestation. We describe a 36-year-old man with advanced HIV disease who had defaulted on treatment. He presented with bizarre, involuntary movements of his left upper and

First report of viable Toxoplasma gondii isolation from African lions (Panthera leo): Evidence for their status as intermediate hosts.

Toxoplasma gondii can cause lifelong infection in most definitive and intermediate hosts. Serological evidence of T. gondii infection has been documented in both captive and free-ranging lions. We collected samples from 40 captive African lions (Panthera leo) in China from 2017 to 2025 and investigated T. gondii in these animals via serological, bioassay, and molecular methods. The overall prevalence of T. gondii in the 40 lions was 45.0% (95% CI: 30.70-60.18%). Sex, age, phenotype, and sampling date were not risk factors for susceptibility to T. gondii infection. A viable T. gondii isolate (d
