INTRODUCTION: Parasitic infections are more common in low hygiene regions but can occur in developed countries. They often cause significant morbidity and death, especially in the central nervous system. Some spinal infections are caused by Taenia solium, which causes spinal neurocysticercosis. Echinococcus, which causes spinal echinococcosis, frequently infects the lumbar and thoracic spines. Spinal cord schistosomiasis caused by Schistosoma infection has serious clinical consequences. Toxoplasmosis is common in immunocompromised patients, whereas Toxocariasis is generally more common as a ch

Simulated microgravity (SMG or μG) influences Toxoplasma gondii growth and modifies the cellular structure and function of the host cells. As an obligate intracellular parasite, Toxoplasma gondii depends on the metabolic activity and mitochondrial function of its host. However, the impact of SMG on host-pathogen interactions remains unclear. We investigated the effect of SMG on host cell structure and mitochondrial functions and its impact on Toxoplasma gondii infection. Cultured human foreskin fibroblasts (HFF) and Vero cells under SMG formed stratified cell layers and exhibited morphological

BACKGROUND: Toxoplasmosis infection affects at least 30% of the human population globally. Transmission primarily occurs through the ingestion of Toxoplasma gondii oocysts. Various studies have documented the seroprevalence of T. gondii among children with neurological manifestations such as autism, epilepsy, and hydrocephalus; however, data on asymptomatic infants remain limited. Therefore, this study is aimed at determining the seroprevalence of T. gondii infection and identify associated risk factors among children in Mwanza, Tanzania.
METHODS: This was a hospital-based cross-sectional stu

Toxoplasmosis caused by the Apicomplexan parasite Toxoplasma gondii is a significant health threat to immunocompromised individuals and newborns. This parasite has a complex life cycle that is well controlled to achieve optimal transmission and pathogenesis. Acute phase of the disease is caused by rapid proliferation of tachyzoites, which has a highly coordinated and tightly regulated cell cycle to allow parasite propagation. Tachyzoite cell cycle has five partially overlapping phases (G1, S, G2, M and C) with distinct gene expression patterns and cellular activities, yet the underlying regula

Central nervous system (CNS) toxoplasmosis represents one of the most common opportunistic complications in patients with advanced HIV/AIDS infection. A wide range of neurological symptoms may contribute to the clinical presentation of the disease. This paper reports the case of a 29-year-old man with untreated HIV infection, in whom neurotoxoplasmosis led to rapidly progressive, bilateral sensorineural hearing loss and severe disability. Appropriate neuroimaging diagnostics enabled the initiation of effective treatment; however, neurological sequelae, including hearing impairment, persisted.

Medication errors remain a leading cause of patient harm, with significant consequences for both health outcomes and healthcare systems. In this case report, we presented a human immunodeficiency virus (HIV) positive patient who was treated with pyrimethamine due to Toxoplasmosis encephalitis. He unaware of the critical role of folinic acid, neglected to take it, resulting in severe hematologic complications, including disseminated intravascular coagulation (DIC). This report highlights the importance of effective patient education and clear communication regarding the potential risks of medic

Cerebral toxoplasmosis is a major opportunistic infection caused by Toxoplasma gondii, particularly in individuals with advanced HIV/AIDS and other forms of immunosuppression. This narrative review synthesizes current knowledge on the parasitology, diagnosis, treatment, and prevention of cerebral toxoplasmosis, with emphasis on clinical relevance and public health implications. We outline the parasite's life cycle and mechanisms of central nervous system invasion, distinguishing latent CNS infection in immunocompetent hosts from reactivated disease in immunocompromised patients. Diagnostic cha

Secondary central nervous system involvement by extranodal NK/T-cell lymphoma is rare and presents significant diagnostic challenges, particularly in immune compromised patients. We report a case of a 29-year-old man with chronic active Epstein-Barr virus infection and prior extranodal NK/T-cell lymphoma status post allogeneic hematopoietic stem cell transplantation who presented with a new-onset seizure and a ring-enhancing intracranial lesion. Initial imaging findings closely mimicked cerebral toxoplasmosis, leading to empiric antimicrobial therapy and delayed diagnosis. Despite treatment, t

Bromodomains (BRDs) are protein interaction modules that exclusively recognize acetylation motifs, and are evolutionarily conserved and present in diverse nuclear proteins. Toxoplasma gondii is the causative agent of toxoplasmosis, with the tachyzoite stage driving pathogenesis through rapid invasion and replication within nucleated cells. Bromodomain-containing protein 4 (BDP4) is a conserved BRD protein across apicomplexans. To address the roles of T. gondii bromodomain-containing protein 4 (TgBDP4) in the lytic circle of T. gondii, the clustered regularly interspaced short palindromic repea

Toxoplasma gondii is a significant foodborne protozoon responsible for human toxoplasmosis, typically transmitted via undercooked meat, contaminated water, or contact with oocysts in cat feces. This study aimed to evaluate the prevalence and genetic diversity of T. gondii in retail meats within Songkhla province to assess public health risks. A total of 304 meat samples-comprising chicken (N = 108), pork (N = 107), and beef (N = 89)-were collected from 30 fresh markets across seven regions of Songkhla. DNA was extracted from 100 μl of meat juice and analyzed using GRA7 nested-PCR assays. Posit

Toxoplasmosis, caused by Toxoplasma gondii, is a widespread zoonotic disease associated with reproductive losses in livestock and important public health consequences. Despite the recognized veterinary and public health importance of toxoplasmosis, data on T. gondii infection in goats in Zambia are currently lacking. Therefore, a cross-sectional seroepidemiological study was conducted among goats in eight veterinary camps in Choma District to determine the seroprevalence and associated risk factors of T. gondii infection. A total of 324 goat serum samples were examined for IgG antibodies again

Separation of cis/trans isomers is critical for SAR studies but challenging due to similar properties. To address this, we designed a rigid triple-connected heterocyclic scaffold locking stereochemical geometry, enabling reliable separation and unambiguous assignment. From a library of cis/trans oxadiazole derivatives, cis compound 6 emerged as the most potent anti-Toxoplasma agent, displaying superior selectivity, lower cytotoxicity, and enhanced intracellular parasite clearance compared to its trans counterpart and reference drug spiramycin. Mechanistic investigations revealed compound 6 ind

Toxoplasma gondii is an obligate intracellular apicomplexan parasite that poses serious risks to immunocompromised individuals and the global livestock industry. Our previous study showed that AP2X-1 regulates stage conversion and virulence of T. gondii; however, whether AP2X-1-deficient strains can serve as live-attenuated vaccine candidates is unknown. Here, we evaluated the in vivo virulence, protective efficacy, and induced immunity of the ap2X-1 knockout strain PruΔap2X-1. In Kunming mice, infection with even the highest tested dose (5 × 106 tachyzoites) of PruΔap2X-1 resulted in 100% sur

Curcumin derivatives (curcuminoids) exhibit considerable antiparasitic and antiviral activities. In this study, EF24-like bis-2-fluorobenzylidene piperidone derivatives were synthesized and evaluated for antiprotozoal activity and inhibition of the human immunodeficiency virus (HIV)-1 reverse transcriptase (RT)-associated RNase H (HIV-1 RNase H). A series of bis-arylidene piperidones and tetrahydro(thio)pyranones bearing halogen or nitro substituents were tested against Leishmania major (promastigotes and amastigotes) and Toxoplasma gondii. L. major promastigotes were the most sensitive parasi
