Toxoplasma gondii in Egyptian Blood Donors and Livestock: Seroprevalence, Risk Factors, and One Health Implications.

Toxoplasma gondii is a globally prevalent zoonotic parasite with major public health and veterinary impacts. In Egypt, One Health data on its distribution among blood donors and livestock are limited. This multicentric cross-sectional study aimed to determine the seroprevalence of Toxoplasma gondii among asymptomatic blood donors and livestock across six different Egyptian governorates and to identify the associated risk factors. Serum samples of 2000 asymptomatic blood donors and 500 animals (cattle and buffaloes) were tested for anti-Toxoplasma gondii IgG and IgM antibodies using ELISA. Soci

ROP64-Deficient Toxoplasma gondii Strain Confers Protection Against Acute and Chronic Toxoplasmosis in Mice.

Toxoplasmosis, caused by Toxoplasma gondii, remains an important zoonotic disease affecting both human and animal health, particularly in immunocompromised individuals and during pregnancy. Current treatments are unable to eliminate latent tissue cysts and are further limited by adverse effects and prolonged treatment regimens, highlighting the need for effective vaccines. In this study, we evaluated the virulence attenuation and protective efficacy of a CRISPR-Cas9-generated rhoptry protein 64 knockout strain (RHΔrop64) as a live attenuated vaccine candidate in Kunming mice. RHΔrop64 exhibite

Current Status of Avian Toxoplasmosis in the World: A Systematic Review and Meta-analysis.

INTRODUCTION: Toxoplasmosis is a food-borne disease that infects almost all warm-blooded mammals and birds. Birds are considered intermediate hosts for the parasite. The consumption of infected birds by humans and other animals plays a highlight role in the transmission of this parasite. Accordingly, the present review was conducted to find the global seroprevalence of T. gondii in birds. METHODS: A comprehensive search of five academic databases (ScienceDirect, PubMed, Web of Science, ProQuest, and Scopus) and Google Scholar was conducted for literature published from 1952 to December 13, 20

Genome-resolved analysis reveals disruption of gut microbial vitamin B and K2 biosynthesis during Toxoplasma gondii infection in mice.

UNLABELLED: Toxoplasma gondii infection remodels the gut microbiome, yet its impact on microbial vitamin biosynthetic potential and host redox metabolism remains unclear. Here, we integrated mouse gut metagenomes with publicly available metagenome-assembled genomes (MAGs) to construct a genome-resolved atlas of B-vitamin and vitamin K2 biosynthesis. From 45,697 MAGs, we curated 4,771 representative genomes, of which 2,682 met high-quality criteria (completeness ≥90%, contamination <5%). Functional annotation identified 229,717 vitamin-related genes corresponding to 177 Kyoto Encyclopedia of Ge

Interkingdom remodeling of the intestinal bacteriome and virome during Toxoplasma gondii infection in rats.

Toxoplasma gondii infection is associated with intestinal microbiome disruption, but its effects on genome-resolved bacterial populations, the gut virome, and bacteriome-virome relationships remain poorly understood. Using previously generated shotgun metagenomic datasets from 36 intestinal samples collected from 18 Sprague-Dawley rats across control, acute, and chronic infection groups, we reconstructed 294 quality-filtered, non-redundant bacterial metagenome-assembled genomes (MAGs) and identified 899 medium-to-high-quality viral operational taxonomic units (vOTUs) from assembled metagenomic

Targeting sexual stage-specific flagellar proteins to develop vaccines for feline toxoplasmosis.

Toxoplasma gondii mainly relies on feline-shed oocysts for environmental transmission, making the blockade of oocyst shedding an effective strategy for toxoplasmosis control. Although cilia- and flagella-associated proteins (CFAPs) are essential for flagellar assembly, maturation, and motility in mammalian male gametes, their biological roles and transmission-blocking vaccine potential in apicomplexan parasites remain unexplored. Here, we identified CFAP221 as a sexual stage-exclusive antigen of T. gondii. CRISPR/Cas9-mediated knockout of CFAP221 did not affect tachyzoite growth or mouse virul

Integrated Proteomic Profiling Reveals Dynamic Remodeling of the Intestinal Proteome in Toxoplasma gondii-Infected C57BL/6J Mice.

The intestinal mucosa is the primary site of Toxoplasma gondii (T. gondii) infection and interaction with the host. While the intestinal responses have been characterized through transcriptomic and histopathological studies, a comprehensive, system-wide analysis of the functional proteome remodeling by acute T. gondii infection is required to elucidate the underlying mechanisms. This study presents a temporal, quantitative proteomic profiling of T. gondii-infected C57BL/6J mouse intestine to define the protein-centric host response. Global analysis reveals profound reprogramming, characterized

Therapeutic Antitoxoplasmosis Potential of Garcinia cambogia: Evidence From In Silico and In Vivo Studies.

Toxoplasma gondii, an opportunistic protozoan, is widely prevalent and poses potential health risks. Its treatment remains challenging with several side effects, urging the exploration of new, safe, and efficient drugs. We aimed to explore the therapeutic efficacy of the medicinal plant Garcinia cambogia (GC) against both acute and chronic toxoplasmosis. Sixty Swiss albino mice were allocated into five groups: I (healthy), II (model), III (spiramycin [SPM] treated, IV (GC treated), and V (SPM + GC treated). Each group is further divided into two subgroups receiving treatment at 4 and 42 days p

A terpyridine derivative exhibits potent and selective anti-Toxoplasma gondii activity in BeWo cells.

Toxoplasmosis is a zoonosis widespread worldwide, although it primarily affects populations in underdeveloped countries. In humans, infection can lead to serious complications, particularly in immunocompromised individuals and pregnant women. Despite advances in the diagnosis of this disease, therapeutic regimens for treating toxoplasmosis are not fully effective, and new drugs are highly desirable. In this context, we evaluated 4'-(4-methylphenyl)-2,2':6',2''-terpyridine (L) and its previously reported Pt(II) complex (1) against Toxoplasma gondii. The free ligand L exhibited greater antiparas

Clustering of host N-glycans by the microneme MIC1/4/6 complex licenses Toxoplasma rhoptry discharge.

To initiate the process of invasion, apicomplexan parasites must discharge the contents of specialized organelles called rhoptries into host cells. This process requires prior recognition and binding of the host cell by proteins released from another set of parasite organelles, the micronemes. However, the host-parasite interactions required for rhoptry discharge are largely unknown. Here, we performed a host cell-directed genome-wide screen for factors that mediate rhoptry discharge from Toxoplasma gondii, the causative agent of toxoplasmosis. The screen identified host N-glycosylation and ch

Untargeted Lipidomics Reveals Lipid Alterations in Colonic Contents of C57BL/6J Mice with Acute Toxoplasma gondii Infection.

BACKGROUND: Although Toxoplasma gondii has been recognized as an obligate intracellular parasite, the lipid metabolic alterations it induces in colonic contents during acute infection remain poorly characterized. METHODS: An acute infection model was established using C57BL/6J mice by oral inoculation with the T. gondii ME49 strain. Untargeted lipidomics analysis was performed on the colonic contents collected at day 10 post-infection. RESULTS: Acute infection significantly elevated glycerophospholipids (GPs) and reduced glycerolipids (GLs), and prenol lipids (PRs), leading to 136 differenti

Role of Toxoplasma gondii calcium-binding protein (TgpCaBP) and its implications for the development of attenuated live vaccines.

UNLABELLED: Toxoplasma gondii is an obligate intracellular protozoan parasite and the causative agent of toxoplasmosis in humans and animals. To date, an ideal vaccine against toxoplasmosis has not yet been developed. Our previous study demonstrated that deletion of the TgpCaBP gene, which encodes a calcium-binding EF-hand protein, impaired calcium flux, invasion, and egress, thereby reducing T. gondii virulence. However, the mechanisms underlying this loss of virulence and the potential of RHΔTgpCaBP as a live attenuated vaccine remain unclear. Here, we showed that TgpCaBP deletion significan
