Genetic Diversity of Theileria parva and Anaplasma spp. Isolated From Ticks Collected From Kiambu County, Kenya.

Environmental changes and human activities such as deforestation and expansion of agricultural land are increasing tick-borne diseases including Anaplasmosis, Babesiosis, Ehrlichiosis, and Theileriosis. These diseases, which affect animals, can be transmitted to humans through tick bites. Kiambu County's warm, wet climate provides an environment conducive to tick breeding and development. This study investigated the circulation and genetic diversity of Theileria parva and Anaplasma spp. in tick samples collected from Kiambu County, Kenya. Ticks were collected from cattle, goats, and sheep usin

Epidemiological and clinicopathological factors associated with infection by multiple pathogens transmitted by Rhipicephalus sanguineus sensu lato in naturally infected dogs in the Semiarid area of Northeastern Brazil.

Domestic dogs are frequently exposed to tick-borne pathogens such as Babesia vogeli, Hepatozoon canis, Anaplasma platys and Ehrlichia canis, which can cause a wide range of clinical manifestations, although little is known about the epidemiological and clinicopathological profiles of mono- and co-infections. This study aimed to characterize infection patterns and identify clinical and epidemiological factors associated with mono-infections, and co-infections with two or more pathogens in naturally infected dogs. We analyzed 181 dogs from a hospital population with suspected hemoparasitic infec

Haemaphysalis longicornis (Acari: Ixodidae) failed to acquire and consequently transmit Babesia bigemina to naïve cattle.

Since its introduction into the United States, the Asian longhorned tick (Haemaphysalis longicornis) has spread to several states and poses a growing threat to livestock due to its broad host range and rapid reproductive capacity. Since H. longicornis can transmit livestock pathogens such as Theileria orientalis Ikeda, it is a high priority to determine the vector competence of a United States population of H. longicornis for other livestock pathogens that circulate or may re-emerge domestically, such as Babesia bigemina, the causative agent of bovine babesiosis. This study evaluated the vecto

Challenges in Diagnosing Acanthamoebae Infections Using Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry.

Acanthamoebae are amphizoic protozoa that can be human pathogens, causing diseases such as granulomatous amoebic encephalitis or Acanthamoeba-keratitis; Therefore, it is critical to have a rapid and reliable diagnostic approach; Following two studies with divergent results, we have aimed to assess the development of a time-saving and cost-effective method for medical parasitological diagnostics to identify Acanthamoebae by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry based on their characteristic protein profiles. Several xenically cultured Acanthamoeba strains

A case-control study of blood group antigen expression in Babesia microti infected patients in the United States.

BACKGROUND: Babesia and Plasmodium are intraerythrocytic parasites that cause babesiosis and malaria, respectively. Significant associations between human blood group antigens and malaria are known; however, few studies have examined the relationship between blood group antigens and babesiosis. STUDY DESIGN AND METHODS: From June to August 2025, 100 Babesia polymerase chain reaction (PCR)- and 100 B. microti PCR+ human patient samples tested at a national reference laboratory underwent ABO and D phenotyping and red blood cell (RBC) genotyping of 37 antigens. RESULTS: The predicted phenotype

Screening and evaluation of potential histone deacetylase inhibitors against Babesia infections.

Babesia spp., a group of blood protozoa transmitted via the bites of tick vectors, cause enormous economic losses for livestock industry and pose significant risk to public health. Currently, the drugs available for treating babesiosis are limited and possess certain toxicity, making the discovery of new and safer drug candidates an urgent priority. Inhibitors targeting histone deacetylase (HDACs) have attracted increasing attention for its antiparasitic effects as a drug targeting epigenetic regulatory enzymes. However, little is known about the effects of HDAC inhibitors on Babesia parasites

Immunity beyond borders: non-lethal Plasmodium confers cross-protection against lethal Babesia via macrophage activation.

INTRODUCTION: In co-endemic regions, hosts are concurrently infected by related apicomplexan hemoparasites, Plasmodium and the zoonotic pathogen Babesia. This study investigates how Plasmodium berghei XAT-Babesia rodhaini co-infection modulates host immunity, dissecting the contributions of innate and adaptive immune cells. METHODS: Disease progression was assessed by survival, parasitemia, body weight, and hematological parameters. Splenic histopathology and qPCR were used to evaluate tissue damage and parasite burden. Immune responses were analyzed by cytokine, antibody measurement, and flo

Combination of essential oils from Ocimum americanum L., Ocimum gratissimum L., and Lippia multiflora Mold.: a potential solution for controlling the cattle tick Rhipicephalus microplus (Acari: Ixodidae).

The cattle tick Rhipicephalus microplus is one of the most significant ectoparasites affecting livestock in tropical and subtropical regions worldwide. This tick causes substantial economic losses in cattle production by reducing weight gain and milk yield and by transmitting pathogens responsible for diseases such as babesiosis and anaplasmosis. Conventional control strategies rely largely on synthetic acaricides; however, their use poses risks to human health and the environment and has contributed to the rapid emergence of resistant tick populations. Under these circumstances, the search fo

Characterization of vesicle-associated exported immunodominant antigens of the human pathogen Babesia duncani.

Human babesiosis is an emerging disease caused by intraerythrocytic Babesia parasites, primarily transmitted through tick bites. Among the species of Babesia that infect humans, Babesia duncani causes severe disease in murine models, associated with excessive cytokine responses that contribute to immunopathology. The parasite factors that mediate host immune activation remain poorly defined. Here, we show that B. duncani, similar to B. microti, exports antigens through vesicle-mediated pathways. Screening a panel of 105 known and predicted secreted proteins using sera from B. duncani-infected

Babesia microti: Breaking the culture barrier.

Babesia microti poses a significant threat to human health, underscoring the need for an improved in vitro culture system to reduce reliance on animal models and support drug and vaccine screening. Key challenges include the tropism of preferential host red blood cells (RBCs), parasite metabolic needs, culture medium formulation, and optimization of the microaerophilic environment (oxygenation).

Babesia gibsoni in Brazil: First multilocus molecular characterization in a domestic dog with clinical, hematological and morphological data.

Canine babesiosis is a disease caused by protozoa of the genus Babesia, an important hemoparasite in veterinary clinical practice. In Brazil, Babesia vogeli predominates; however, Babesia gibsoni, common in Asia and the United States, has emerged globally with distinct diagnostic, therapeutic, prophylactic, and prognostic features. In the present study, we report the first multilocus molecular characterization of B. gibsoni in Brazil with supported clinical, laboratory and morphological data. A Dachshund dog from the Federal District presented with nonspecific clinical signs, including fever,

Development and evaluation of a bead-based xMAP immunoassay for differential serodiagnosis of Babesia bovis and B. bigemina in cattle.

Bovine babesiosis is caused by Babesia bovis and Babesia bigemina and requires species-resolved serosurveillance to guide control programs. With this goal in mind, we developed an indirect bead-based liquid microarray (xMAP/Luminex) immunoassay using recombinant SBP4 (B. bovis) and RAP-1 (B. bigemina). Antigens immunoreactivity was confirmed by dot-blot and western blot. and antigens were covalently coupled to magnetic microspheres. Assay conditions were optimized with anti-bovine IgG-phycoerythrin detection, and diagnostic performance was assessed in 89 bovine sera previously classified by an

Characterization of a Novel OTU-like Deubiquitinase from Babesia microti: Implications for Babesiosis Treatment.

Babesia microti is the primary agent of human babesiosis, an emerging tick-borne disease with limited treatment options and growing evidence of drug resistance. Deubiquitinases (DUBs) play critical roles in protein homeostasis and host-pathogen interactions, yet none have been characterized in B. microti. Here, we report the first molecular cloning, expression, and functional characterization of an OTU-like cysteine protease from B. microti (bm-OTU). The recombinant bm-OTU protein (~22 kDa) was expressed in E. coli, purified to high homogeneity, and exhibited ~93% solubility under native condi
