Babesia are tick-transmitted apicomplexan parasites of widespread medical and veterinary importance. Although atovaquone, a first-line therapy for human babesiosis, targets one of the three proteins encoded by mitochondrial DNA, a full characterization of the nuclear-encoded mitochondrial proteome is lacking. Here, we combined organelle immunoprecipitation and density gradient separation to enrich mitochondria from Babesia divergens. Using protein mass spectrometry and protein correlation profiling, we generated B. divergens MitoCarta (BdMitoCarta), a high-confidence inventory of 525 mitochond

In Kazakhstan, despite the diversity of ixodid ticks and extensive livestock movement throughout Central Asia, ovine Babesia infections are poorly characterized. The objective of this study was to determine the occurrence, genetic diversity and geographic distribution of Babesia spp. in sheep from southern and southeastern Kazakhstan. A total of 1458 blood samples from 51 settlements across five regions were screened by nested PCR targeting mitochondrial cytb. Positive samples were characterized by Sanger sequencing, with additional multilocus analysis based on 18S rRNA and mitochondrial cox1

Babesiosis is a tick-borne infection caused by intraerythrocytic Babesia parasites and may result in severe hemolytic anemia and systemic complications. Coinfection with other Ixodes-borne pathogens may further complicate diagnosis and management. We report a 45-year-old immunocompetent man working as a landscaper in Connecticut who presented with syncope following two weeks of fever, fatigue, diaphoresis, arthralgias, jaundice, and dark urine. Laboratory evaluation revealed profound hemolytic anemia with a hemoglobin of 6.8 g/dL, elevated lactate dehydrogenase (LDH), undetectable haptoglobin,

BACKGROUND: While socioeconomic status (SES) indicators such as educational attainment and employment are among the major drivers of health and illness, the health returns of SES indicators may differ across racial groups. Built on Marginalization related Diminished Returns framework (MDRs) that refers to weaker health effects of SES indicators for marginalized and minoritized groups than non-Hispanic White people, we conducted this study with two aims: First to test the association between educational attainment and employment with cardiometabolic diseases (CMDs), and second, to test racial v

Babesiosis is a vector-borne protozoal disease primarily transmitted by the Ixodes scapularis tick, though it can also be transmitted through blood transfusions from infected donors. The illness can be asymptomatic or present with mild flu-like symptoms. Still, in severe cases, it can lead to disseminated intravascular coagulation and severe hemolytic anemia, sometimes necessitating management in the intensive care unit. Traditionally, severe babesiosis has been linked to individuals over 50 years old, asplenia, and those who are immunocompromised. Notably, parasitemia levels greater than 10%

Rhipicephalus microplus is a critical cattle ectoparasite and a vector of pathogens, including bacteria and protozoan parasites, that cause diseases such as bovine anaplasmosis and babesiosis. To better understand the pathogen profile of this tick in China, we collected 592 tick specimens from seven provinces. Using conventional or nested PCR, we detected a range of pathogens. Six Anaplasma species were identified (prevalence: 3 - 21%), with the highest detection rate in Hunan Province (24%). Spotted fever group rickettsiae, including R. raoultii (21%), Candidatus R. jingxinensis (12%), R. slo

BACKGROUND: Human babesiosis is an emerging tick-borne disease caused by Babesia parasites, most notably Babesia microti and Babesia duncani in North America and Babesia divergens in Europe. Infections can be severe, persistent, or relapse despite treatment, and current therapeutic options remain limited, underscoring the urgent need for new and effective treatment options.
METHODS: Acridone derivatives, originally developed as potent antimalarial agents against multiple life cycle stages of the malaria parasites, were evaluated for their antibabesial activity using continuous in vitro cultur

Egress from host cells is fundamental for the spread of infection by apicomplexan parasites, including Babesia species. These tick-borne pathogens represent emerging zoonoses, but treatment options are limited. Here, using microscopy, transcriptomics and chemical genetics, we identified signalling, proteases and gliding motility as key drivers of egress by Babesia divergens. We developed reverse genetic tools in B. divergens to perform a knockdown screen of putative mediators of egress, identifying kinases and proteases involved in distinct steps of egress (aspartyl protease (ASP) 3 and kinase

Red foxes are widespread carnivorans acting as reservoirs of Babesia vulpes , which is highly prevalent in foxes, yet its clinical impact remains poorly understood. The description of Babesia banethi raised knowledge gaps regarding pathogenicity and life cycle. This study investigated the occurrence of Babesia spp. in red foxes from southern Italy. Overall, 54 red foxes were clinically examined, sampled for hemato-biochemical analyses, and screened for Babesia spp. using microscopy and PCR. Necropsies were performed on 20 animals. Ticks were collected, identified, and dissected for cytological

Babesiosis is a tick-borne protozoan infection marked by invasion of red blood cells and causing hemolytic anemia. When symptomatic, patients present with symptoms and signs secondary to hemolysis. We report a case of Babesiosis presenting with a rare manifestation of a splenic infarct, emphasizing the importance of early recognition and prompt treatment. A 41-year-old man with no significant past medical history presented to the hospital with fever, chills and sweats for 2 days. He also had left lower quadrant abdominal pain which started 10 days prior to the admission. Initial laboratory res

The cattle fever ticks Rhipicephalus microplus (Canestrini, 1888) and Rhipicephalus annulatus (Say, 1821) transmit pathogens responsible for bovine babesiosis and anaplasmosis. Although eradicated from the United States, these ticks and the pathogens they transmit are endemic in Mexico and other countries, and pose a serious risk to the U.S. cattle industry. Cattle imported into the U.S. are inspected and treated with an acaricide, coumaphos, to prevent tick reentry into the U.S. Coumaphos is an organophosphate (OP) that kills ticks by inactivating acetylcholinesterase (AChE), an enzyme vital

IntroductionTicks are globally recognised as the second most important vectors of infectious diseases, posing significant threats to human and animal health. Haemaphysalis parva (Acari: Ixodidae) is frequently reported infesting humans and domestic animals and has been experimentally demonstrated to transmit Babesia ovis, with field associations to ovine babesiosis during the colder months. It has also been reported to harbour several zoonotic pathogens, including Coxiella burnetii, Francisella tularensis, and various Rickettsia species. Here, we aim to report the complete mitochondrial genome

The discovery of broad-spectrum antiparasitic agents relies on the ability to evaluate drug efficacy under harmonized in vitro conditions across related species. However, current drug screening pipelines for intraerythrocytic parasites are constrained by the use of species-specific media with distinct nutrient compositions and serum sources, which hinder direct comparison of compound potency. To address this gap, we describe a unified erythrocytic culture system based on DMEM/F12 supplemented with 20% fetal bovine serum (DFS20), which supports robust asexual growth of multiple Plasmodium falci

INTRODUCTION: Babesia rossi infection in dogs is associated with severe hemostatic dysregulation and systemic inflammation. The close interplay between innate immunity and coagulation in pathological states, including acute infections, may lead to thrombus formation, termed thromboinflammation, and has been proposed to underlie disease severity and poor outcomes. The objectives of this study were to investigate the presence of thromboinflammation in dogs with B. rossi infection by characterizing the overall hemostatic state, using thromboelastography (TEG) and plasma-based assays, and the syst

BACKGROUND: Babesia, an intraerythrocytic protozoan, is globally distributed and poses a significant public health burden. In China, human babesiosis is mainly caused by Babesia microti, and the Zhejiang Province is endemic. This study aimed to determine the prevalence of B. microti in the population, reservoir hosts, and vectors; assess population exposure risks; and gain further insight into the genotype structure in Zhejiang.
METHODS: From May 2020 to October 2023, blood samples from local residents and blood donors, as well as blood or liver tissues from rodents and questing or host-attac

Human granulocytic anaplasmosis is an emerging tick-borne illness caused by Anaplasma phagocytophilum and transmitted by Ixodes ticks. Clinical manifestations are often nonspecific, including fever, malaise, and cytopenias, which can delay recognition and treatment. Although thrombocytopenia is common in human granulocytic anaplasmosis (HGA), profound thrombocytopenia is rarely described, and cardiac conduction abnormalities remain infrequently reported. We describe a 73-year-old woman in New England who developed fever, somnolence, acute kidney injury, transaminitis, a platelet nadir of 12 ×

Tick-transmitted apicomplexan parasites of the genus Babesia infect vertebrate red blood cells and cause babesiosis in humans and animals. Detailed studies of their subcellular organization have been constrained by their small size and intracellular development. In this study, we report the identification of a previously unrecognized apical tubulin rosette (ATRo) in Babesia divergens - a unique rosette-like microtubule structure located at the apical pole of the parasite. The ATRo consists of 6-7 radial spines and represents a distinct architecture that appears morphologically different from t

Babesia microti is an emerging tick-transmitted agent causing babesiosis in humans, becoming a major concern to public health due to the lack of a vaccine. In silico methods such as reverse vaccinology and immunoinformatics offer a convenient tool for identifying promising vaccine targets. Using computational immunoinformatics and molecular modelling tools, the current study aims to develop a multi-epitope vaccination against B. microti. For identifying the antigenic proteins, subtractive proteomics, NCBI, and PPGA were used, whereas for their screening, ProtParam (ExPASy), VaxiJen 2.0, AllerT
