Development of an ERA-CRISPR/Cas12a-based visual diagnostic assay for bovine babesiosis.

Bovine babesiosis, caused by Babesia bovis, is a tick-borne hemoparasitic disease causing substantial economic losses to cattle industries worldwide. Current diagnostic methods, particularly microscopy, lack sensitivity for subclinical infections, while molecular assays require sophisticated equipment unsuitable for field use. To address this gap, we developed a novel diagnostic assay integrating enzymatic recombinase amplification (ERA) with CRISPR-Cas12a-mediated trans-cleavage, targeting the B. bovis spherical body protein 2 (SBP2) gene. The assay enables visual readout via UV/blue light fl

Synergistic in-vitro activity of Fosinopril alone and in combination with imidocarb dipropionate against Babesia bovis.

Bovine babesiosis, caused by Babesia bovis is a major cattle disease worldwide, particularly in tropical and subtropical regions. A commonly used drug for the control and treatment of babesiosis is imidocarb dipropionate (ID), which has proved effective against B. bovis. Nevertheless, its toxicity to ruminants and other mammals, along with the persistence of drug residues in meat and milk for several months after treatment, has led to restriction on its use. Furthermore, several reports indicate that complete parasite elimination is not obtained in some cases. Interestingly, the angiotensin-co

Development and preliminary evaluation of a chimeric epitope-based antigen (TevISDY) for serological detection of surra.

Trypanosoma evansi remains a neglected yet economically significant pathogen in Latin America, where current serological diagnostic tools suffer from low specificity, cross-reactivity, and limited field applicability. This study presents the design and preliminary evaluation of a novel chimeric antigen derived from epitopes of the TevSTIB805.3.100 protein, a variant surface glycoprotein (VSG) of T. evansi, and explores its potential application as a serological marker for surra. An ISDY-based chimeric antigen was engineered, expressed, purified, and structurally characterized using integrated

Thermostabilization of Vaccines by Cryopreservation.

For thermostabilization of vaccines that incorporate live eukaryotic organisms (e.g., parasites) or cells, cryopreservation is the preferred and usually only method for long-term storage. Parasites include the unicellular protozoa and multicellular helminths (nematodes, trematodes, and cestodes). The PfSPZ live sporozoite vaccines against malaria are cryopreserved. Additionally, many parasites are transmitted by vectors (e.g., mosquitoes, ticks), and these can be targets for vaccination or used in the production of vaccines. Veterinary live vaccines against bovine babesiosis, East Coast Fever,

Identification of Small-Molecule Inhibitors of Babesia bovis by High-Throughput SYBR Green I Screening.

Background: Bovine babesiosis, caused by Babesia bovis, remains a major constraint to cattle production in endemic regions, while the current control relies on only a few drugs with concerns over resistance and residues. Methods: This study screened 9600 chemically diverse compounds from the Drug Discovery Initiative, University of Tokyo, against B. bovis using a 96-well SYBR Green I high-throughput assay at 1 µM. Primary hits were subjected to dose-response testing to determine IC50 values, followed by Madin-Darby bovine kidney cell cytotoxicity assays for selectivity index calculation. Combi

A Compendium Review of the Global Epidemiology of Ticks and Tick-borne Diseases: Regional Insights from Türkiye.

Vector-borne diseases have historically posed significant threats to plants, humans, domestic animals, and wildlife, with their impact being especially pronounced in tropical and subtropical regions. Among these, tick-borne diseases (TBDs) have emerged as an increasingly critical global concern. This growing threat is largely driven by the expanding geographic range of ticks and the wide array of pathogens they transmit, including viruses, bacteria, protozoa, nematodes, fungi, and infectious prion proteins. The global cumulative economic impact of the challenges caused by ticks and TBDs contri

Clinical form of babesiosis caused by Babesia canis in Polish foxes (Vulpes vulpes).

The red fox (Vulpes vulpes) is known to be a reservoir host of various vector-borne protozoan parasites. However, the impact of infections caused by Babesia canis on the health status of the red fox remains unknown, and research on this topic conducted on fox populations in Poland and worldwide has been fragmentary. It is known that these animals can become infected with Babesia canis, but it is unclear whether a clinical form of the disease can develop in them. This study aimed to present for the first time the cases of clinical babesiosis in foxes in Poland. The observations covered four fox

Infectious diseases and blood supply safety in the United States in 2026: A review.

From the infectious diseases perspective, the safety of the US blood supply, already high-level, continues to improve via multiple mechanisms, making transfusion-transmitted infections (TTIs) now rare. Blood supply safety in 2026 rests on multiple, interdigitating cornerstones including: 1) updated donor questionnaires and guidances for blood collection centers from the Food and Drug Administration, 2) the long-term commitment and expertise of national organizations (e.g., the American Red Cross and the Association for the Advancement of Blood and Biotherapies), 3) screening of donor blood pro

Congenital babesiosis in China: first molecularly confirmed case of vertical transmission of Babesia microti.

Congenital babesiosis is rarely reported globally. We report a 74-day-old male infant presented with fever, pallor, and severe, life-threatening haemolytic anaemia (haemoglobin: 45 g/L). The infant had 16% parasitemia with ring forms evident on peripheral blood smear. Babesia microti infection was confirmed in both the mother and infant by PCR and metagenomic next- generation sequencing. Genetic analysis revealed an identical strain in bot. Treatment with intravenous azithromycin and oral atovaquone/proguanil resulted in rapid clearance of parasitemia and resolution of anaemia. This first mole

Tafenoquine succinate inhibits the growth of the equine piroplasmosis hemoparasites Theileria equi and Babesia caballi.

BACKGROUND: Equine piroplasmosis (EP) is a tick-borne disease of equids caused by the intraerythrocytic apicomplexan parasites Theileria equi, Babesia caballi and the recently identified Theileria haneyi. Acute cases can be severe, with anemia, jaundice, abortion or sudden death. Survivors remain lifelong carriers, serving as reservoirs for tick-borne and iatrogenic transmission. No vaccines are currently available, and control strategies rely heavily on accurate diagnostics and chemotherapeutic intervention. Imidocarb dipropionate (ID) is the current standard of care for both acute treatment

Acquired motility of Babesia microti-infected red blood cells.

Babesia microti is an intraerythrocytic protozoan parasite and the main causative agent of human babesiosis in the United States. While extensive research has focused on the prevalence of this vector-borne pathogen in natural populations, increases of human cases and clinical manifestation, and pathogen structure, little is known about the movements of B. microti within vertebrate red blood cells (RBCs). RBCs are nonmotile due to their lack of cellular structures for active movement. Here, we report a phenomenon in which B. microti-infected RBCs exhibit an acquired motility compared to uninfec
