Screening and evaluation of potential histone deacetylase inhibitors against Babesia infections.

Babesia spp., a group of blood protozoa transmitted via the bites of tick vectors, cause enormous economic losses for livestock industry and pose significant risk to public health. Currently, the drugs available for treating babesiosis are limited and possess certain toxicity, making the discovery of new and safer drug candidates an urgent priority. Inhibitors targeting histone deacetylase (HDACs) have attracted increasing attention for its antiparasitic effects as a drug targeting epigenetic regulatory enzymes. However, little is known about the effects of HDAC inhibitors on Babesia parasites

Immunity beyond borders: non-lethal Plasmodium confers cross-protection against lethal Babesia via macrophage activation.

INTRODUCTION: In co-endemic regions, hosts are concurrently infected by related apicomplexan hemoparasites, Plasmodium and the zoonotic pathogen Babesia. This study investigates how Plasmodium berghei XAT-Babesia rodhaini co-infection modulates host immunity, dissecting the contributions of innate and adaptive immune cells. METHODS: Disease progression was assessed by survival, parasitemia, body weight, and hematological parameters. Splenic histopathology and qPCR were used to evaluate tissue damage and parasite burden. Immune responses were analyzed by cytokine, antibody measurement, and flo

Combination of essential oils from Ocimum americanum L., Ocimum gratissimum L., and Lippia multiflora Mold.: a potential solution for controlling the cattle tick Rhipicephalus microplus (Acari: Ixodidae).

The cattle tick Rhipicephalus microplus is one of the most significant ectoparasites affecting livestock in tropical and subtropical regions worldwide. This tick causes substantial economic losses in cattle production by reducing weight gain and milk yield and by transmitting pathogens responsible for diseases such as babesiosis and anaplasmosis. Conventional control strategies rely largely on synthetic acaricides; however, their use poses risks to human health and the environment and has contributed to the rapid emergence of resistant tick populations. Under these circumstances, the search fo

Characterization of vesicle-associated exported immunodominant antigens of the human pathogen Babesia duncani.

Human babesiosis is an emerging disease caused by intraerythrocytic Babesia parasites, primarily transmitted through tick bites. Among the species of Babesia that infect humans, Babesia duncani causes severe disease in murine models, associated with excessive cytokine responses that contribute to immunopathology. The parasite factors that mediate host immune activation remain poorly defined. Here, we show that B. duncani, similar to B. microti, exports antigens through vesicle-mediated pathways. Screening a panel of 105 known and predicted secreted proteins using sera from B. duncani-infected

Babesia microti: Breaking the culture barrier.

Babesia microti poses a significant threat to human health, underscoring the need for an improved in vitro culture system to reduce reliance on animal models and support drug and vaccine screening. Key challenges include the tropism of preferential host red blood cells (RBCs), parasite metabolic needs, culture medium formulation, and optimization of the microaerophilic environment (oxygenation).

Development and evaluation of a bead-based xMAP immunoassay for differential serodiagnosis of Babesia bovis and B. bigemina in cattle.

Bovine babesiosis is caused by Babesia bovis and Babesia bigemina and requires species-resolved serosurveillance to guide control programs. With this goal in mind, we developed an indirect bead-based liquid microarray (xMAP/Luminex) immunoassay using recombinant SBP4 (B. bovis) and RAP-1 (B. bigemina). Antigens immunoreactivity was confirmed by dot-blot and western blot. and antigens were covalently coupled to magnetic microspheres. Assay conditions were optimized with anti-bovine IgG-phycoerythrin detection, and diagnostic performance was assessed in 89 bovine sera previously classified by an

Comparative therapeutic efficacy of Artesunate-Atovaquone-Azithromycin and Doxycycline-Clindamycin-Metronidazole regimens in dogs naturally infected with Babesia gibsoni.

Canine babesiosis, caused by Babesia gibsoni infection, is a potentially fatal tick-borne disease in dogs and depicts variable responses to combination therapies. This study was designed to evaluate a novel Artesunate-Atovaquone-Azithromycin (AAA) regimen against the standard Doxycycline-Clindamycin-Metronidazole (DCM) protocol in naturally infected dogs. The study included two treatment groups and a healthy control group, each comprising 10 dogs. Treatment efficacy was assessed through haemato-biochemical analysis and estimation of parasite load by relative quantification (RQ) real-time PCR o

Advances and Challenges in the Diagnosis of Vector-Borne Protozoal Infections in Veterinary Medicine.

Vector-borne protozoal infections-including babesiosis, theileriosis, hepatozoonosis, trypanosomosis, and leishmaniosis-impose a substantial burden on livestock and companion animal health worldwide and carry important zoonotic and public health implications. Accurate diagnosis is essential yet challenging, given the diversity of parasite genera, their markedly different tissue tropisms, and the uneven distribution of diagnostic resources across veterinary settings. This review provides an integrated overview of the principal diagnostic approaches available, structured around the biological lo

Characterization of a Novel OTU-like Deubiquitinase from Babesia microti: Implications for Babesiosis Treatment.

Babesia microti is the primary agent of human babesiosis, an emerging tick-borne disease with limited treatment options and growing evidence of drug resistance. Deubiquitinases (DUBs) play critical roles in protein homeostasis and host-pathogen interactions, yet none have been characterized in B. microti. Here, we report the first molecular cloning, expression, and functional characterization of an OTU-like cysteine protease from B. microti (bm-OTU). The recombinant bm-OTU protein (~22 kDa) was expressed in E. coli, purified to high homogeneity, and exhibited ~93% solubility under native condi

Not all trophozoites are malaria: A case of babesiosis in a traveler from the USA.

Babesiosis, a tick-borne disease caused by protozoa of the genus Babesia, is endemic in North America, but rarely reported in Europe. We report a case involving a splenectomised 72-year-old man from New Hampshire, USA, who was admitted at Careggi University Hospital (Florence, Italy), complaining of a febrile syndrome appeared during a trip to central Italy. After the laboratory flagged the presence of intra-erythrocytic ring-shaped elements, malaria was initially suspected but subsequently excluded by molecular testing. Based on epidemiological and clinical history, Babesia microti infection

Diversity and population genetic analysis of the Babesia bovis merozoite surface Antigen 1 (MSA-1) gene in South African cattle isolates from field and clinical samples.

Babesia bovis causes significant losses in South African cattle, and antigenic variation in the merozoite surface antigen 1 (msa-1) complicates vaccine and diagnostic development. This study provides the first detailed molecular characterisation of msa-1 genetic diversity in South African cattle populations. Analysis of 18 successfully sequenced isolates (∼960 bp) revealed extensive polymorphism, with pairwise nucleotide sequence identities ranging from 30.1% to 100%, and amino acid similarities ranging from 37.6% to 100%. Population genetic metrics of the highly variable MSA-1 confirmed high
