BACKGROUND: The introduction of second-generation antipsychotics (SGAs) over the past 2 decades generated considerable optimism that better antipsychotic treatments for schizophrenia and bipolar disorder were possible. SGAs offer several tolerability benefits over first-generation antipsychotics (FGAs), particularly with respect to extrapyramidal symptoms. However, SGAs can induce serious metabolic dysregulations, especially in drug-naive, first-episode, and child and adolescent populations, with olanzapine and clozapine having the highest propensity to cause these abnormalities. In this conte

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

Chromosome cohesion, mediated by the cohesin complex, is essential for the process of chromosome segregation. Mutations in cohesin and its regulators are associated with a group of human diseases known as the cohesinopathies. These diseases are characterized by defects in head, face, limb, and heart development, mental retardation, and poor growth. The developmental features of the diseases are not well explained by defects in chromosome segregation, but instead are consistent with changes in gene expression during embryogenesis. Thus a central question to understanding the cohesinopathies is

BACKGROUND: Tibial hemimelia is an uncommon congenital abnormality ranging from a solitary skeletal deformity to associations with other syndromes. Despite Paley et al.'s detailed classification of tibial hemimelia, some unique sporadic cases still do not fit within this system. In 2021, Chong et al. proposed the most recent surgical approach based on the Paley classification, which inspired cases such as ours.
CASE PRESENTATION: This case study presents a novel case of tibial hemimelia characterized by a bilobed distal tibia, a shortened first ray, and an equinovarus deformity in a 5-year-ol

Bilateral anterior amelia (BAA) is the congenital absence of thoracic limbs and has been reported in the Chihuahua as an autosomal recessive disorder. In some cases, the digits of the pelvic limbs can be variably affected, but otherwise, the pelvic limbs are generally spared. A GWAS performed with nine BAA affected Chihuahuas identified a significant association on chromosome 13, and homozygosity mapping delineated a 2.1 Mb chromosomal region containing the RSPO2 gene. Loss of function variants of RSPO2 in humans and cattle has been associated with the absence of all limbs. Six affected Chihua

In 2022, a global outbreak of mpox was anticipated, with several cases reported in non-endemic countries in early May. Given the challenge of distinguishing the mpox virus (MPXV) from other pathogens based solely on symptoms, there is an urgent need for prompt and reliable MPXV detection methods. In this study, we developed assays using recombinase-aided amplification (RAA) to identify MPXV and evaluated their applicability with clinical samples. The assays were designed to detect the N4R gene of MPXV. All assays demonstrated detection limits of 1 copy/µL within the reaction system and exhibit

Hereditary diseases in cattle lead to economic losses across various breeds worldwide, with arachnomelia syndrome being one such disorder found in Simmental cattle. Arachnomelia results from a recessively inherited loss-of-function variant in the MOCS1 gene detected only in Simmentals. In Türkiye, prior research on hereditary cattle diseases has focused exclusively on dairy breeds, particularly Holstein. This study marks the first investigation in Türkiye of a genetic disease in a dual-purpose breed that is also valued for its beef traits. For this purpose, blood samples were collected from 38

Ectromelia virus (ECTV), a member of the Orthopoxvirus genus, serves as both a causative agent of mousepox and a pivotal surrogate model for studying highly pathogenic orthopoxviruses. Although genomic data on ECTV remains limited, we report the isolation and characterization of a novel strain, ECTV-C-Tan-GD01, obtained from rodents in Guangdong Province, China. Nanopore sequencing yielded a complete genome (199 annotated genes, including one gene truncated at the C-terminus) with inverted terminal repeats (ITRs) harboring a conserved hairpin structure. Notably, a frameshift-inducing "G" delet

The cohesin protein complex plays a very important role in chromosome segregation, transcription, DNA replication and chromosome condensation. Mutations in cohesin proteins give rise to a disease collectively referred to as Cohesinopathies. The major cause of cohesinopathies arise due to defects associated with gene expression, that give rise to developmental disorders. We have used Saccharomyces cerevisiae to mimic the cohesinopathy disorder Roberts syndrome with mutations (eco1W216G) homologous to that of humans (esco2). Our data suggests that polyol sugars like sorbitol, can repair misfolde

PURPOSE: Phocomelia is an intercalary segmental dysplasia of the upper limb, a distinct entity from longitudinal deficiencies. Nearly 20 years ago, more severely dysplastic limbs initially thought to be phocomelia were able to be reclassified as more severe forms of longitudinal dysplasia. This study sought to evaluate current phocomelia cases to identify if these represented true phocomelia.
METHODS: Using the Congenital Upper Limb Differences Registry, all cases of phocomelia were evaluated to consider the phocomelia diagnosis. Twenty patients were included for assessment, in whom 23 extrem

The study presents the discovery of a novel class of N-arylated 1,2,4-oxadiazol-5(4H)-ones as potent inhibitors of orthopoxviruses, including the variola virus (VARV). Through systematic structural modifications, two lead compounds, 4 (4-CF3/4-NO2) and 10 (4-I/4-NO2), demonstrated in submicromolar concentration antiviral activity against Vaccinia virus (VACV), cowpox virus (CPXV), ectromelia virus (ECTV), and VARV, with selectivity indices (SI) up to 13738. Studies of mechanisms of action, including time-of-addition experiments and molecular modeling, have shown that these compounds can target

OBJECTIVE: To report a new type of sirenomelia in early pregnancy, which was different from the reported types.
CASE REPORT: A 33-year-old Chinese pregnant women (gravida 1 and para 1) attended our hospital in early pregnancy for a routine prenatal ultrasound screening. The Ultrasonography showed fused lower extremities that moved at unison. Two femurs in the thigh were present, while the tibias and fibulas were absent. The right upper limb was not confirmed, with a soft tissue mass located in the right upper limb region. In addition, it revealed an anechoic structure in the posterior cervica

BACKGROUND: Fibular hemimelia (FH) is a rare congenital condition characterized by longitudinal lower limb deficiency with fibular hypoplasia or aplasia commonly associated with femoral and tibial shortening resulting in leg length discrepancy (LLD). Femoral distraction osteogenesis represents a well-established reconstructive approach in patients with LLD exceeding 2 cm.
METHODS: Twenty-three patients with FH were treated with antegrade femoral distraction osteogenesis between 2018 and 2021 using magnetically controlled motorized intramedullary lengthening nails (Precice P2 or Stryde, NuVasi

Fibular hemimelia (FH) is a rare congenital abnormality where the fibula is either totally or partially absent. It can occur alone or alongside other skeletal malformations, and in very few cases, it may occur along with non-skeletal anomalies. A 4-year-old female was diagnosed with unilateral right-sided FH, accompanied by limb shortening, a right-side ankle deformity, valgus foot, and 3 lateral rays that had been totally absent since the first week of birth. And she was incidentally diagnosed with spina bifida occulta at a 3-year-old age. FH is most commonly unilateral and mostly affects the

Historically, thalidomide-induced congenital malformations have served as an important example of the enhanced susceptibility of developing embryos to chemical perturbation. The compound produced a wide variety of congenital malformations in humans, which were initially detected by an association with a relatively rare limb defect labeled phocomelia. Although true phocomelia in the most severe form is a transverse defect with intercalary absence of limb regions, it is proposed that thalidomide produces a longitudinal limb phenotype in humans under usual circumstances that can become transverse

Tibial hemimelia is a rare congenital deficiency that is challenging to treat. Appropriate care varies due to the many subtypes and associated knee and ankle deformities. Amputation is still the most accepted treatment, but it is not generally preferred by families. Consequently, other reconstruction options have been made available as well. In the following two cases, we performed knee joint reconstruction by fusion of the ankle joint instead of amputation. This technique has not been reported before in the literature and has been named as Cabukoglu reconstruction technique or cruris plasty.

Ulnar hemimelia (UH), a rare congenital skeletal condition, is frequently linked to other upper limb abnormalities and is characterized by the partial or total absence of the ulna. We describe a rare instance of a 12-year-old patient with unilateral UH, humeroradial synostosis, and oligodactyly. The patient presented with obvious left upper limb shortening and functional limitations. The absence of the ulna, fusion of the humerus and radius, and the underdeveloped digits were all verified by radiographic examination. This case emphasizes the phenotypic variability linked to UH and contributes
