Ulnar hemimelia (UH), a rare congenital skeletal condition, is frequently linked to other upper limb abnormalities and is characterized by the partial or total absence of the ulna. We describe a rare instance of a 12-year-old patient with unilateral UH, humeroradial synostosis, and oligodactyly. The patient presented with obvious left upper limb shortening and functional limitations. The absence of the ulna, fusion of the humerus and radius, and the underdeveloped digits were all verified by radiographic examination. This case emphasizes the phenotypic variability linked to UH and contributes

OBJECTIVE: Congenital absence of the tibia (congenital tibial aplasia) causes significant disability when walking. The usual treatment, which involves lengthening the lower leg or early amputation followed by prosthetic fitting, requires technical resources that are often lacking in developing countries. The proposed alternative consists of tibialization of the fibula with limb lengthening and by preservation of the foot equinismus.
PATIENTS AND METHOD: The records of 25 children with congenital tibial aplasia were reviewed. Ten of these children underwent tibialization of the fibula (operate

INTRODUCTION: Fibular hemimelia is a rare congenital condition characterized by hypoplasia of a bone segment associated with malformations. Its clinical presentation is highly variable, not only in terms of the types of malformations but also their severity. If untreated, it can lead to significant functional impairments. Managing patients with this condition often spans several decades. There is no standardized treatment protocol, and to our knowledge, no recent French study has analyzed the surgical strategies implemented and the long-term functional outcomes in these patients. The last Fren

We present a dimethyl sulfoxide (DMSO)-enhanced one-pot HDA-CRISPR/Cas12a biosensor for the ultrasensitive detection of the monkeypox virus (MPXV). The MPXV B6R gene was initially amplified using DMSO-enhanced helicase-dependent amplification (HDA) in the bottom of the reaction tubes. DMSO was employed to enhance the amplification efficiency of HDA. CRISPR/Cas12a reagents, pre-added to the caps of the reaction tubes, were subsequently combined with HDA products to generate fluorescence signals. This DMSO-enhanced HDA-CRISPR/Cas12a biosensor enables the detection of synthetic B6R DNA within 1 h

Monkeypox (mpox) is a zoonotic viral disease transmitted by the monkeypox virus (MPXV). Viral protein-based nanovaccines hold promise in preventing the infection of MPXV. MPXV-derived antigens (A29L, A35R, and M1R) were capable of eliciting protective immunity and suffered from low immunogenicity. Adjuvants and delivery systems were critical to improving the immunogenicity of antigens. In this study, a pathogen-mimicking nanovaccine was developed by conjugation of poly(amidoamine) (PAMAM) dendrimers with deoxycholic acid to form cationic nanoparticles as a delivery platform. The three antigens

Monkeypox (Mpox), a zoonotic viral disease characterized by strong pathogenicity and transmissibility, necessitates the development of safe and effective vaccine. A protein-based vaccine holds great promise due to its high safety and strong specificity. A29L and M1R from intracellular mature virion and A35R from extracellular enveloped virion are immunodominant antigens capable of providing adequate antigen coverage. However, these proteins necessitate the adjuvant and the delivery system to optimize the immunogenicity. Here, STG-982 (a STING agonist) was conjugated with the antigens to functi

Monkeypox (mpox), caused by the monkeypox virus (MPXV), is a zoonotic disease of global public health concern. Current live-attenuated vaccines suffer from safety concerns and unsatisfactory protection effectiveness. To address these limitations, a protein-based nanovaccine (Ag-M-Spy) was developed to deal with mpox for its high effectiveness and favorable safety profile, incorporating three critical MPXV-derived antigens (A29L, A35R, M1R). Mannose acted as the adjuvant via mannose receptor-mediated endocytosis. Ag-M-Spy was constructed by conjugation of mannose-functionalized antigens with Sp

The 2022 global mpox virus (MPXV) outbreak highlights the urgent need for safer, next-generation vaccines. We compared the immunogenicity and protective efficacy of individual and multicomponent membrane proteins of MPXV virions in mice to inform the development of a recombinant subunit vaccine against mpox. BALB/c mice were immunized with eukaryotically expressed A35R, A29L, B6R, and M1R proteins, administered individually or in multicomponent combinations with an Al(OH)3 + CpG oligodeoxynucleotide adjuvant. Three multicomponent protein vaccines (A29/B6, A29/B6/M1, and A29/B6/M1/A35) provided

The modified vaccinia virus Ankara (MVA) is approved for use as a smallpox and monkeypox virus vaccine and was also designed as a popular recombinant viral vector for vaccine development and gene therapy. However, the extensive genomes of poxviruses present a significant challenge for the development of recombinant viral vaccines; therefore, it is essential to establish a user-friendly in vitro reverse genetic system. We systematically assembled the 180-kb MVA genome into a five-plasmid system, facilitating one-step packaging of the MVA virus. The MVA rescued using this system exhibited simila

This study addresses the limited understanding of factors affecting the efficiency of water treatment plants in reducing trihalomethane (THM) formation through total organic carbon (TOC) removal, highlighting significant challenges in improving treatment effectiveness. The aim of this study was to examine the influence of water quality on the efficiency of water treatment plants to remove TOC and reduce THM formation. Linear regression and correlation analyses were conducted to examine the relationship between water quality parameters and THM concentrations. The results showed that there was a

Six decades ago the world learned that thalidomide, a seemingly non-toxic sedative and hypnotic, caused severe birth defects including the flipper-like deformity of the arms known as phocomelia. When thalidomide was tested against placebo by the trialist Louis Lasagna in 1960 (while the drug was banned from the U.S. marketplace), he found the 100-mg dosage equivalent to placebo, as well as greatly inferior to the 200-mg dosage, in producing sleep. Even as these findings were made known, a 100-mg dose of thalidomide was in general use as a sleep aid for pregnant women. It appears that unbeknown

Scientific interest in orthopoxvirus infections and search for new highly effective compounds possessing antiviral activity against orthopoxviruses have significantly increased as a result of worldwide mpox outbreak in 2022. The present work deals with the synthesis of new 2-arylimidazoles exhibiting in vitro activity not only against the vaccinia virus, cowpox virus and ectromelia (mousepox) virus but also against the variola virus. Among the imidazole derivatives under consideration (1-hydroxyimidazoles, 1-methoxyimidazoles, 1-benzyloxyimidazoles, and imidazole N-oxides), the most promising

Roberts syndrome (RBS) is an autosomal recessive disorder with profound growth deficiency and limb reduction caused by ESCO2 loss-of-function variants. Here, we elucidate the pathogenesis of limb reduction in an Esco2fl/fl;Prrx1-CreTg/0 mouse model using bulk- and single-cell-RNA-seq and gene co-expression network analyses during embryogenesis. Our results reveal morphological and vascular defects culminating in hemorrhage of mutant limbs at E12.5. Underlying this abnormal developmental progression is a pre-apoptotic, mesenchymal cell population specific to mutant limb buds enriched for p53-re

The role of CD4+ T cells in the induction of protective CD8+ T cells by mRNA lipid nanoparticle (LNP) vaccines is unknown. We used B6 or Tlr9 -/- mice depleted or not of CD4+ T cells and LNP vaccines loaded with mRNAs encoding the ectromelia virus (ECTV) MHC class I H-2 Kb-restricted immunodominant CD8+ T cell epitope TSYKFESV (TSYKFESV mRNA-LNPs) or the ECTV EVM158 protein, which contains TSYKFESV (EVM-158 mRNA-LNPs). Following prime and boost with 10 μg of either vaccine, Kb-TSYKFESV-specific CD8+ T cells fully protected male and female mice from ECTV at 29 (both mRNA-LNPs) or 90 days (EVM15

INTRODUCTION: Amongst low- to middle income countries (LMIC), outdated and poorly documented orthopedic interventions may pose patients at risk for complications, serious adverse events (SAE) and unsatisfactory results, contradicting the World Health Organization's (WHO) Safe Surgery mandates. Use cases of conditions managed inadequately in LMIC environments are necessary to increase awareness of stakeholders and overcome deficiencies in collaboration with high-income countries (HIC).

Poxviruses are dsDNA viruses infecting a wide range of cell types, where they need to contend with multiple host antiviral pathways, including DNA and RNA sensing. Accordingly, poxviruses encode a variety of immune antagonists, most of which are expressed early during infection from within virus cores before uncoating and genome release take place. Amongst these antagonists, the poxvirus immune nuclease (poxin) counteracts the cyclic 2'3'-GMP-AMP (2'3'-cGAMP) synthase (cGAS)/stimulator of interferon genes DNA sensing pathway by degrading the immunomodulatory cyclic dinucleotide 2'3'-cGAMP, the

Sirenomelia is a rare congenital anomaly characterized by fusion of the lower extremities and multiple visceral abnormalities. It usually has a lethal prognosis due to the severity of the associated abnormalities. We present the case of a 26-year-old female patient, in her second pregnancy without associated comorbidities, who was admitted to the Emergency department due to a 26-week pregnancy and anhydramnios. She subsequently underwent an ultrasound in the Maternal Fetal Medicine service, who performed the Sirenomelia diagnosis. It was decided to terminate the pregnancy due to the poor progn
