Challenges in Diagnosing Acanthamoebae Infections Using Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry.

Acanthamoebae are amphizoic protozoa that can be human pathogens, causing diseases such as granulomatous amoebic encephalitis or Acanthamoeba-keratitis; Therefore, it is critical to have a rapid and reliable diagnostic approach; Following two studies with divergent results, we have aimed to assess the development of a time-saving and cost-effective method for medical parasitological diagnostics to identify Acanthamoebae by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry based on their characteristic protein profiles. Several xenically cultured Acanthamoeba strains

Cutaneous Acanthamoebiasis: Two Cases Highlighting Diverse Histopathologic Findings.

Acanthamoeba is a free-living ameba, known to most commonly cause amebic keratitis in contact lens users and granulomatous amebic encephalitis in immunocompromised patients. Cutaneous Acanthamoebiasis as a single-organ manifestation is less common. We report two cases seen in our department. Case one is a 23-year-old hospitalized patient with systemic lupus erythematosus and pancytopenia who acutely developed nodules on the extremities. Biopsy showed lymphohistiocytic inflammation and focal coagulative necrosis in the subcutaneous fat. Acanthamoeba trophozoites and cysts were apparent in H&E-s

Successful Treatment of Cutaneous Acanthamoeba polyphaga With Marine Algae, Padina pavonica, Extract: Targeting TLR2/TLR4 Signal and Cytokeratin 18 in Immunocompromised Animals.

Cutaneous amoebiasis is a severe opportunistic infection, particularly prevalent among immunocompromised individuals, with high mortality due to the insidiousness of the disease, its ability to evade early diagnosis, and lack of specific symptoms. Herein, we aimed to evaluate the therapeutic potential of the marine alga, Padina pavonica, extract as a topical treatment for Acanthamoeba polyphaga skin infections. Immunocompromised mice with induced cutaneous lesions were topical infected with A. polyphaga and examined 3, 9, and 15 days post infection through histological and molecular analysis.

Isolation, characterization, and pathogenicity assay of Acanthamoeba and its endosymbionts in respiratory disorders and COVID-19 hospitalized patients, northern Iran.

Acanthamoeba spp., are common free-living amoebae found in nature that can serve as reservoirs for certain microorganisms. The SARS-CoV-2 virus is a newly emerged respiratory infection, and the investigation of parasitic infections remains an area of limited research. Given that Acanthamoeba can act as a host for various endosymbiotic microbial pathogens and its pathogenicity assay is not fully understood, this study aimed to identify Acanthamoeba and its bacterial and fungal endosymbionts in patients with chronic respiratory disorders and hospitalized COVID-19 patients in northern Iran. Hypoxia and collagen deposition in the kidneys infected with Acanthamoeba sp.

Acanthamoeba spp. are facultative, opportunistic pathogens that are found in diverse environments. In the hosts, they lead to multi-organ disease. Recent studies reported that they may induce changes in the kidneys of hosts. The aim of the study was to determine the influence of Acanthamoeba sp. on hypoxia and collagen deposition in the kidneys of immunocompetent and immunosuppressed mice infected with Acanthamoeba sp. The results strongly suggest that Acanthamoeba sp. induces hypoxia in mice with normal and reduced immune response by increasing gene and/or protein expression of HIF1α as well

Metagenomic next-generation sequencing-assisted diagnosis of a rare case of primary cutaneous acanthamoebiasis in an HIV patient: a case report.

Pathogenic and free-living Acanthamoeba are widely distributed in the environment and have been reported to cause keratitis and universally fatal encephalitis. Primary cutaneous acanthamoebiasis caused by Acanthamoeba is exceedingly rare and presents as isolated necrotic cutaneous lesions without involvement of the cornea or central nervous system. Cutaneous acanthamoebiasis often occurs in immunocompromised patients and is likely overlooked or even misdiagnosed only by cutaneous biopsy tissue histopathological analysis. Here, we report a HIV-infected 63-year-old female with oral leukoplakia f

Successful Treatment of Primary Acanthamoeba Cutis in a Patient With Chronic Lymphocytic Leukemia on Acalabrutinib.

Primary cutaneous amoebiasis is rare, and typically affects immunocompromised patients and presents with unique clinical and histopathologic changes. Untreated, the infection could progress to involve the central nervous system, which is almost universally fatal. We present a case of primary cutaneous acanthamoebiasis in a patient with chronic lymphocytic leukemia on acalabrutinib. Timely diagnosis and treatment resulted in complete resolution of lesions and a disease-free status at the 14-month follow-up. A 76-year-old man presented with a 2-month history of multiple, nonhealing, ulcerated, e

The Immunological Changes in the Skin of BALC/c Mice with Disseminated Acanthamoebiasis.

Toll-like receptors (TLR) are involved in the recognition of numerous pathogens, including Acanthamoeba spp. Thanks to this, it is possible for immune cells to recognize microorganisms and trigger the body's innate immune response. The stimulation of TLRs also leads to the activation of specific immunity. The aim of the study was to determine the TLR2 and TLR4 gene expression in the skin of BALC/c mice infected with Acanthamoeba with AM22 strain isolated from a patient. Receptor expression was assessed by real-time polymerase chain reaction (qPCR) in the amoeba-infected host with normal (A) an

The role of apoptosis and oxidative stress in the pathophysiology of Acanthamoeba spp. infection in the kidneys of hosts with different immunological status.

BACKGROUND: Acanthamoeba spp. are opportunistic pathogens that cause inflammation, mostly in the brain, lungs and cornea. Recent reports indicate kidney dysfunction in hosts with systemic acanthamoebiasis. The aim of the study was to analyze the gene expression and protein concentration of NADPH oxidase 2 and 4 (NOX2 and NOX4, respectively) and nuclear erythroid 2-related factor (Nrf2) in the kidneys of hosts with systemic acanthamoebiasis. We also aimed to determine the protein and gene expressions of Bcl2, Bax, caspases 3 and 9. METHODS: Mice were divided into four groups based on their imm

The First Case of Cutaneous Acanthamoebiasis Caused by Acanthamoeba triangularis in Korea.

A 62-year-old man with multiple myeloma visited our clinic with multiple painful erythematous to purpuric nodules on his whole body. He received a skin biopsy which showed septal and lobular inflammation with vasculitis, and multiple amoebic organisms were found. Polymerase chain reaction and culture were performed and an Acanthamoeba triangularis infection was diagnosed. This is the first report on cutaneous acanthamoebiasis caused by A. triangularis, suggesting that A. triangularis should be regarded as a clinical pathogen that can cause ocular as well as disseminated infection.

Clinical Improvement of Disseminated Acanthamoeba Infection in a Patient with Advanced HIV Using a Non-Miltefosine-Based Treatment Regimen in a Low-Resource Setting.

Disseminated Acanthamoeba species infection is likely an underrecognized and underdiagnosed opportunistic infection in patients with advanced human immunodeficiency virus (HIV) disease in South Africa. It presents a unique clinical challenge in that the diagnosis can be difficult to establish and management options are limited in low-resource settings. To our knowledge, there is a paucity of literature to date on the successful use of combination treatment options for patients in low-resource settings without access to miltefosine. We present a case describing the clinical improvement of disse
