Cutaneous acanthamoebiasis in AIDS.

Acanthamoeba is a recognized pathogen in the immunocompromised patient, commonly presenting as chronic or subacute encephalitis. However, cutaneous disease in the absence of CNS involvement is increasingly recognized, especially in the setting of chronic, nonhealing skin lesions in the patient with AIDS. We describe a patient with AIDS and cutaneous acanthamoebiasis and review our experience with treatment and diagnosis.

Purification and characterization of iron superoxide dismutase and copper-zinc superoxide dismutase from Acanthamoeba castellanii.

Two superoxide dismutases (SOD I and SOD II) were purified from Acanthamoeba castellanii and characterized for several biochemical properties. Analysis of the primary structure and inhibition studies revealed that SOD I is iron SOD (Fe-SOD), with a molecular mass of 50 kDa, and SOD II is copper-zinc SOD (Cu,Zn-SOD), with a molecular mass of 38 kDa. Both enzymes have a homodimeric structure consisting of 2 identical subunits, each with a molecular mass of 26 and 19 kDa for SOD I and SOD II, respectively. The isoelectric points of SOD I and SOD II were 6.4 and 3.5, respectively, and there were n

Successful treatment of widely disseminated acanthamoebiasis.

Disseminated Acanthamoeba infections are almost invariably fatal, with no universally accepted standard for treatment. Reports of acanthamoebiasis in non-human-immunodeficiency-virus infected hosts are rare. We successfully treated a lung transplant patient who had disseminated acanthamoebiasis using a combination of pentamidine, 5-fluorocytosine, itraconazole, and topical chlorhexidine gluconate/ketoconazole cream.

Acanthamoebiasis in Korea: two new cases with clinical cases review.

The first case was 7-month-old immunodeficiency girl in whom the diagnosis of Acanthamoeba pneumonia was established by culture of a bronchial washing. The patient had been ill for a month when she was admitted due to neonatal thrombocytopenia with respiratory difficulty and treated with gammaglobulin and steroid. Her chest X-ray showed diffuse alveolar consolidation on the left lung with interstitial hazziness and a partial sign of hyperinflation on the right lung. Laboratory tests showed that the Candida antigen was negative and Pneumocystis carinii was not detected. Mycoplasma antigen was n

Partial characterization of the proteolytic secretions of Acanthamoeba polyphaga.

The ocular pathology associated with acanthamoebiasis may result, at least in part, from the excretory and secretory (E&S) products of the active Acanthamoeba trophozoites. To test this hypothesis, the ability of A. polyphaga (ATCC Strain 30461) trophozoite E&S products to digest collagen, the major constituent of the corneal stroma, was evaluated. The secreted proteinases of A. polyphaga were identified using in vitro azocoll degradation, activity-PAGE, radiolabeled extracellular matrix (ECM) degradation, and collagen degradation assays. Inhibitors of serine (phenylmethylsulfonyl fluoride, di

[Review article: occurrence, parasitism and pathogenetic potency of free-living amoeba].

In areals of human life free living amoebae (FLA) are widely distributed and various possibilities to infect men as well as animals do exist under specific circumstances. While Naegleria species, respectively, Naegleria fowleri give rise to fulminant fatal processes of CNS Acanthamoeba infections mostly have a chronical course and immunodeficiency could be a favouring factor for acanthamoebiasis. Naegleria amebae may invade nasal mucosa after having contacts to warm and contaminated water, on the other hand Acanthamoeba infections develop when damaged skin come into contact with soil or slug o

Diagnosis of Acanthamoeba infection by cutaneous manifestations in a man seropositive to HIV.

A case of Acanthamoeba infection with cutaneous manifestations in a man seropositive for human immunodeficiency virus is reported. The diagnosis was made by identification of amoebic trophozoites and cysts in tissue specimens. Two other cases of disseminated Acanthamoeba infection with similar skin findings in patients with acquired immunodeficiency syndrome have been previously reported. Recognition of the cutaneous manifestations of acanthamoebiasis in patients seropositive for human immunodeficiency virus may lead to earlier diagnosis and improved therapy.

[Human diseases caused by free-living amoebae. I. Acanthamoebiasis].

This summary offers an updated panorama of the problems caused in many countries, including our own, by free protozoan, Acanthamoeba class amoebas that in certain circumstances depend on either pathogenic potential or the immunological condition of the host and which produce significant disease syndromes. Presented here are the main morphological and ecological characteristics of these amoeba, a summarized study of their pathological characteristic, diagnostic and therapeutic data, as well as a synopsis of the majority of clinical cases diagnosed in the world to date.

Acanthamoebiasis and immunosuppression. Case report.

Immunosuppression and debilitating illnesses are occasionally associated with multifocal brain lesions of Acanthamoebiasis, an encephalitis distinct from the acute, water-sport related meningoencephalitis caused by Naegleria fowler. A 38-year-old man with a renal transplant two and one-half years before his final illness developed pneumonia due to Legionella micdadei. Candida albicans was isolated from sputum and cytomegalovirus was found in lung and liver biopsies. He had continuous corticosteroids, broad spectrum antibiotics, and immunosuppressive therapies. Coma developed and computerized t

Amebic meningoencephalitis.

As the result of research by many workers, information as to the accurate diagnosis and possibly effective treatment has accumulated. While much more definitive academic investigation will aid in improving the results of medical management, the next objective should be the serious study of patients with suggestive histories and with acute and chronic respiratory ailments for the possible role of free-living amebas therein. The state of the art has reached the point where the two free-living amebic infections can probably be diagnosed by direct examination of exudates from the respiratory tract
