Craniosynostosis is a congenital bone developmental condition characterized by the premature ossification of calvarial sutures, leading to restricted skull expansion and potential neurological complications. Although little is known about the signaling that governs this accelerated fusion, our research group has previously identified a stiffness-dependent upregulation of osteogenic genes in cells derived from fused sutures, highlighting the role of mechanotransduction in disease progression. Building on these findings, the present study describes the development of a unique patient-derived thr

3M syndrome is a rarely inherited autosomal recessive disorder caused by mutations in cullin-7 (CUL7), obscurin-like 1 (OBSL1), and coiled-coil domain containing protein 8 (CCDC8). It is associated with multiple dysmorphic features, including characteristic facial dysmorphism (a face that is triangular, full lips, frontal bossing, a nasal tip that is fleshy, long philtrum, protruding ears and macrocephaly), severe growth retardation prenatally and postnatally and normal intelligence. Although there are multiple skeletal manifestations of the syndrome, such as joint laxity, there is no mention

BACKGROUND: Nonsyndromic craniosynostosis (NSCS) has been associated with higher rates of cognitive, developmental, and motor delays. This study compares neurodevelopmental outcomes among different NSCS subtypes.
METHODS: A retrospective cohort study was performed using a large multi-institutional database. Patients with sagittal (SCS), metopic (MCS), and coronal (CCS) craniosynostosis were propensity-score matched 1:1 with age-matched unaffected controls. Outcomes included psychiatric and behavioral disorders, intellectual disability, speech and language disorders, neurodevelopmental disorde

OBJECTIVES: SLC4A4 encodes electrogenic sodium bicarbonate cotransporter NBCe1, prominently expressed in kidney and brain. Recessive loss-of-function variants in SLC4A4 cause proximal renal tubular acidosis, no brain edema. In the brain, NBCe1 is expressed by astrocytes, where it regulates pH and mediates astrocyte volume changes. Here we describe a novel dominant variant in SLC4A4 in patients with brain edema and investigate how it affects NBCe1 function.
METHODS: Genetic studies identified a novel gene variant in three unrelated pediatric patients with the same MRI pattern of cerebral subco

Cranial suture formation is a dynamic process that requires precise cellular and molecular coordination to regulate bone growth and maintain suture homeostasis. The Fibroblast Growth Factor Receptor 3 (FGFR3) signaling pathway is among the major pathways disrupted in craniosynostosis; however, its precise role during cranial suture formation is still unknown. Using a relevant fgfr3 LoF zebrafish model exhibiting abnormal suture morphology, we demonstrated for the first time that Fgfr3 plays a pleiotropic role in both the formation and maintenance of cranial sutures. Mutations in the chromatin remodeler CHD8 are associated with autism and macrocephaly. While mouse models of Chd8 haploinsufficiency recapitulate brain overgrowth, the specific cellular mechanisms and developmental timing that lead to these anatomical abnormalities remain poorly understood. Here, we conducted 3D imaging of Chd8 V986*/+ mouse brains using magnetic resonance imaging followed by tissue clearing and cellular resolution light-sheet microscopy across embryonic and postnatal developmental stages. We found that brain overgrowth occurs postnatally, driven by an expansion of oligodendro

Metopic craniosynostosis occurs due to premature fusion of the metopic suture, preventing normal lateral expansion of the frontal bones and resulting in trigonocephaly and hypotelorism. Metopic craniosynostosis with trigonocephaly is commonly treated with open fronto-orbital advancement and cranial vault remodeling (FOA/CVR). Although this procedure improves cranial contour, it does not directly widen the medial orbital walls, and its effect on hypotelorism remains uncertain. This study evaluated changes in bony interorbital distance (BIOD) and related craniometric parameters after standard FO

BACKGROUND: Pediatric complex cranial vault reconstruction (CCVR) for repair of craniosynostosis is associated with significant analgesic requirements. Intravenous acetaminophen (IVAPAP) has been shown to be effective in reducing opioid requirements for other types of pediatric surgery. However, the efficacy of IVAPAP in CCVR has not been established. The aim of this study was to determine the impact of IVAPAP on perioperative opioid requirements for CCVR.
METHODS: Children 12 years of age or younger who underwent CCVR between June 2017 and August 2020 were retrospectively evaluated. The coho

INTRODUCTION: Dysmorphic syndromes are characterised by congenital anomalies affecting craniofacial features, body proportions, and organ structure. They are frequently caused by complex genetic alterations, including deletions, duplications, and microdeletions, which impede optimal growth regulation.
AIM OF THE STUDY: The objective of this paper is to present a synopsis of selected dysmorphic syndromes associated with excessive growth, with particular emphasis on syndromes with confirmed or strongly suspected chromosomal aetiology or associated with DNA methylation disorders (epigenetic diso

Accurate measurement of fetal head circumference (HC) is critical for monitoring fetal growth and the diagnosis of microcephaly and macrocephaly. However, conventional manual 2D ultrasound measurements tend to vary because of factors such as the presence of amniotic fluid, acoustic shadowing, occlusions, and variations in fetal position. To address these problems, this study proposes a regression-based deep learning model using EfficientNet-B7 and the Adam optimizer to enhance the accuracy and robustness of HC measurement. The proposed model utilizes EfficientNet’s powerful feature extraction

Craniosynostosis occurs when one or more cranial sutures fuse prematurely and can present complex medical, psychosocial, and caregiving challenges for affected families. Prior research documents parents' experiences of navigating diagnosis and treatment, highlighting value in connecting with other families to share insights and support. However, little is known about specific practical and emotional guidance that parents themselves would offer to others facing similar circumstances. To address this gap, we used reflexive thematic analysis to examine pre-existing qualitative interview data from

BACKGROUND/OBJECTIVES: Malan syndrome (MALNS) is a rare overgrowth disorder caused by pathogenic Nuclear Factor I × (NFIX) gene variants, and characterized by postnatal overgrowth, macrocephaly, developmental delay, intellectual disability and distinctive facial features. Chiari type I malformation (CMI), a condition where the cerebellar tonsils extend below the foramen magnum, has been observed in some patients with MALNS, although the exact relationship between these disorders remains unclear. The objective of this case report is to describe a novel NFIX variant in a patient with MALNS and a

BACKGROUND: Sagittal craniosynostosis produces a characteristic cranial deformity resulting from restricted transverse growth and compensatory anteroposterior elongation. Multiple open cranial vault remodeling techniques are used. However, achieving controlled morphologic correction and intracranial volume expansion while preserving uninvolved structures remains challenging. The Boston Method is a geometry-driven approach that emphasizes posterior vault remodeling to address biparietal narrowing and occipital deformity.
METHODS: A retrospective, single-center cohort study was performed, inclu

Background: Glutaric acidemia type 1 (GA1) is an autosomal recessive neurometabolic disorder caused by pathogenic variants in glutaryl-CoA dehydrogenase (GCDH), with variable clinical severity despite early biochemical detectability. Population-specific mutational spectra and genotype-phenotype correlations remain insufficiently defined in infantile-onset disease. Therefore, this study aimed to define the GCDH variant spectrum in GA1 patients with mild hepatopathy and assess genotype-phenotype correlations. Methods: We performed integrated clinical, biochemical, and molecular characterization

INTRODUCTION: Craniosynostosis is a common congenital disorder characterized by premature fusion of one or more cranial sutures, categorized into non-syndromic (NSCS) and syndromic craniosynostosis (SCS). SCS accounts for ∼30% of cases, often accompanied by severe clinical complications, with 20%-25% of patients lacking a clear molecular etiology.
METHODS: We enrolled 18 Chinese SCS patients and analyzed their clinical and genetic data via whole-exome sequencing (WES), copy number variation (CNV) analysis, and Sanger sequencing validation.
RESULTS: We identified 15 single nucleotide variants
