Biomechanical strain patterns in Meckel's cartilage underlie mandibular variation in FGFR2 mouse models.

Sequence variations in fibroblast growth factor receptor 2 (FGFR2) are associated with craniosynostosis syndromes, disrupting craniofacial development, yet early mandibular alterations remain poorly characterized. This study examined Meckel's cartilage (MC) and mandibular morphology in mouse models of Apert (Fgfr2+/P253R) and Crouzon (Fgfr2cC342Y/+) syndromes using Euclidean distance matrix analysis and finite element modeling at embryonic days 14.5 and 16.5. Apert mice exhibited significant MC shortening and elevated, widespread strain in the growth center area correlating with mandibular sho

Intraoperative and radiographic indicators of extraocular muscle aberrancy in syndromic craniosynostosis.

BACKGROUND: Strabismus is common in syndromic craniosynostosis, often attributed to abnormal extraocular muscle (EOM) trajectory, and orbital anatomy. Absent EOMs have been reported, but the spectrum of primary dysgenesis remains poorly understood. The purpose of this study was to characterize the range of EOM dysgenesis in a large cohort that had undergone strabismus surgery. METHODS: The medical records of patients with syndromic craniosynostosis treated at Boston Children's Hospital (2010-2023) were reviewed retrospectively to identify all cases with neuroimaging and history of strabismus

Blood Transfusion Rates in Non-syndromic Craniosynostosis: A Proportion Meta-analysis of Open Versus Minimally Invasive Techniques.

OBJECTIVES: Premature suture fusion in non-syndromic craniosynostosis often requires surgery to permit normal brain growth. To minimize blood transfusion requirements, minimally invasive surgery (MIS) has been proposed. This study will pool the prevalence of blood transfusion in non-syndromic craniosynostosis surgeries, comparing open surgery (OS) with MISs. METHODS: A systematic review and proportion meta-analysis was conducted in accordance with Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines and registered in PROSPERO (CRD42024604632). Parent-Reported Health-Related Quality of Life at 3 Years of Age in Children Treated for Sagittal or Metopic Synostosis.

Early surgical intervention is recommended for sagittal (SS) and metopic (MS) synostosis to correct head shape and reduce the risk of later social stigmatization. However, the impact of craniosynostosis (CS) and its surgical treatment on health-related quality of life (HRQoL) in early childhood remains insufficiently understood. To evaluate parent-reported HRQoL at 3 years of age in children treated for SS or MS. At the 3-year follow-up, parents of children treated for SS or MS completed the Pediatric Quality of Life Inventory (PedsQL) version 2 to 4 years. Age- and nationality-matched healthy

Implementation of Spring Assisted Cranioplasty at a Single Institution.

Spring-assisted cranioplasty (SAC) is a minimally invasive surgical technique for the treatment of craniosynostosis that may reduce operative morbidity compared with traditional open cranial vault remodeling procedures. A retrospective review was conducted of all patients who underwent SAC between May 2019 and August 2024 at a single academic institution serving a socioeconomically vulnerable population. Demographic, clinical, and operative variables were collected, including suture involvement, spring configuration, perioperative outcomes, complications, and need for secondary procedures. A t

Feasibility of Non-surgical Orthodontic Management in Syndromic Skeletal Class III: A Case Report.

Skeletal Class III malocclusions are frequently associated with craniosynostosis syndromes, resulting in distinctive facial and dental anomalies. Among these, Crouzon syndrome is characterised by mutations in the FGFR2 gene, leading to premature fusion of the cranial vault, cranial base, orbital and maxillary sutures. The phenotypic expression of this congenital condition varies according to the timing and pattern of sutural fusion. This report details the clinical features and management of a 16-year-old male patient presenting with moderate skeletal Class III malocclusion as a manifestation

Definitive reconstruction of frontal asymmetry in an adolescent with anterior plagiocephaly due to unilateral coronal craniosynostosis using a hybrid osteo-titanium technology based on an individualized approach: A case report.

BACKGROUND: Unilateral coronal craniosynostosis causes three-dimensional craniofacial asymmetry. Although correction is usually performed during infancy, some patients present in adolescence with an untreated or persistent deformity. We describe an individualized hybrid reconstruction combining a patient-specific titanium implant with autologous bone. CASE DESCRIPTION: A 15-year-old girl with untreated left unilateral coronal craniosynostosis presented with marked frontal and fronto-orbital asymmetry. Three-dimensional computed tomography demonstrated left frontal retrusion, orbital asymmetry

Cortical development in patients with metopic craniosynostosis.

OBJECTIVE: Patients with metopic synostosis have an increased risk of neurodevelopmental disorders, despite a minimal risk of intracranial hypertension. While mechanical constraint due to premature suture fusion has been proposed as a contributing factor, evidence suggests intrinsic brain malformations may play a more significant role. Cortical thickness, volume, and surface area alterations have been implicated in neurodevelopmental disorders, yet their role in metopic synostosis remains unclear. In this retrospective study, authors investigated whether these cortical parameters are altered i

Shunt-induced craniosynostosis with craniocerebral disproportion: a new way of thinking about an old problem.

OBJECTIVE: Shunting during infancy can precipitate premature closure of one or more cranial suture. Shunt-induced craniosynostosis (SIC) may in turn result in craniocerebral disproportion (CCD). The authors present a single-institution series and offer a pragmatic diagnostic and treatment framework. METHODS: This study was a retrospective case series of patients with SIC-CCD who underwent surgical correction between 2018 and 2025 at a single institution. RESULTS: Eighteen patients were identified. The mean (range) age at initial shunt placement was 2 (0-9) months, and the mean age at CCD cor

Hidden Hollows of the Fetal Cranium-Lacunar Skull in Isolation.

INTRODUCTION: Lacunar skull, also known as Lückenschädel or craniolacunae, is a rare congenital calvarial ossification abnormality characterized by multiple lucent defects of the skull. It is most associated with neural tube defects, Chiari II malformation, craniosynostosis, and other congenital anomalies. Isolated lacunar skull, without associated structural abnormalities, has been infrequently reported, and its natural history, prognosis, and optimal clinical management are poorly defined. CASE PRESENTATION: We report the case of a female infant born at 38 weeks and 5 days' gestation to a 3

Inactivating GNAS complex locus variants impair G protein-coupled receptor signaling and cause multiple suture craniosynostosis in humans and zebrafish.

G protein α-subunit (Gαs), encoded by GNAS, mediates G protein-coupled receptor (GPCR) signaling through the cAMP second messenger pathways, and plays a pivotal role in craniofacial morphogenesis and osteoblast differentiation. Craniosynostosis, one of the most prevalent craniofacial developmental anomalies, is characterized by the premature fusion of cranial sutures. Here, we identify germline heterozygous variants in GNAS as a novel genetic cause of craniosynostosis. Affected individuals presented with multiple-suture synostosis, recognizable dysmorphic features, brachydactyly, short stature

Unilateral Lambdoid Craniosynostosis: A Single Institution's Experience With a Rare Diagnosis.

Unilateral lambdoid craniosynostosis is the least common variant of craniosynostosis. Due to its rarity, the development of new techniques in treatment is limited. Less invasive surgical approaches, including distraction osteogenesis and strip craniectomy, have recently gained popularity in the treatment of all forms of craniosynostosis. These procedures have been shown to decrease intraoperative blood loss, transfusion requirements, and operative time. However, the literature is lacking regarding these treatments for unilateral lambdoid craniosynostosis. Here, we present our institution's exp
