Craniofrontonasal syndrome (CFNS; MIM #304110) is a rare craniofacial disorder characterized by hypertelorism, a broad nasal root with a bifid nasal tip, orofacial clefting, and genital malformations caused by pathogenic variants in the X-linked gene EFNB1 (MIM *300035). CFNS exhibits sex-specific heterogeneity, with increased severity in females likely secondary to cellular interference related to random X-inactivation, resulting in mosaic EFNB1 expression. Previous studies have identified over 140 variants in EFNB1, but approximately 20% of CFNS have negative molecular testing, either due to

Recurrent somatic mosaic pathogenic variants of RHOA have been observed in a newly identified neuroectodermal syndrome, Ectodermal Dysplasia with Facial Dysmorphism and Acral, Ocular, and Brain Anomalies, Somatic Mosaic [EDFAOB]. All 12 previously reported patients had somatic mosaicism for RHOA variants. Conversely, no patients with non-mosaic germline variants of RHOA have been reported. The absence of non-mosaic patients has been explained by the presumed lethal effect of all RHOA variants in non-mosaic status. Here we report an 11-month-old female with EDFAOB-like features but without Blas

BACKGROUND: Craniofrontonasal dysplasia (CFND) is a rare X-linked disorder caused by mutations in the EFNB1 gene, typically characterized by hypertelorism, craniosynostosis, and facial asymmetry. Although other congenital anomalies have been reported, neural tube defects-particularly myelomeningocele (MMC)-have not previously been associated with CFND in humans.
CASE DESCRIPTION: We present the case of a full-term female neonate with prenatally diagnosed MMC and Chiari II malformation. She also exhibited a unique constellation of craniofacial features, including a wide frontal bone defect wit

Allelic variations of Sperm antigen with calponin homology and Coiled-Coil domains 1 Like (SPECC1L) have been associated with a spectrum of cranial-facial pathologies including Teebi hypertelorism and Opitz G/BBB syndrome which manifest as clefting of the palate, wide-eyes, and incomplete closure of the esophagus among others. These pathologies may be indicative of improper cranial neural crest cell (CNCC) delamination and migration. SPECC1L is hypothesized to be an actin-microtubule cross-linking protein as it co-localizes with both microtubules and actin in tissue culture cells. Further, it

Missense and loss-of-function variants in SPECC1L are associated with Teebi Hypertelorism Syndrome 1 (TBHS1). Here, we report a novel SPECC1L intragenic deletion encompassing exon 3, which contains the canonical translation start site, in two related individuals with craniofacial features consistent with TBHS1. We use functional overexpression assays to demonstrate that this deletion leads to alternative start codon usage and protein truncation. This is the first report of a SPECC1L intragenic deletion associated with TBHS1. Our findings suggest that deletion of the canonical N-terminal disord

SPECC1L encodes a cytoskeletal scaffolding protein that interacts with filamentous actin, microtubules, and cell junctional components. In humans, autosomal dominant mutations in SPECC1L cause a syndrome characterized by craniofrontonasal anomalies including broad nasal bridge, ocular hypertelorism, prominent forehead, and cleft lip/palate. Complete loss of SPECC1L in mice on a homogenous genetic background results in perinatal lethality, accompanied by subtle cranial differences and incompletely penetrant cleft palate. This lethality limits postnatal analysis of craniofacial development. Beca

Rothumnd-Thomson syndrome (RTS) is a rare genetic condition characterized by poikiloderma, sparse hair, short stature, skeletal abnormalities, cataracts, and increased risk for malignancies. The presenting symptom is often a classic rash with erythema on the cheeks and face with spread to extensor surfaces of extremities. Gradually over months to years, this rash develops into poikiloderma (reticulated hyper- and hypopigmentation, telangiectasias, and areas of punctate atrophy). Identification of this characteristic rash may facilitate early diagnosis and management in patients with RTS; howev

Orbital hypertelorism (OHT) can be corrected surgically utilizing a 4-wall box osteotomy, facial bipartition, or medial orbital composite unit translocation (MOCUT). However, the impact on periorbital symmetry and patient-reported outcomes (PROs) is less well known. This study assesses periorbital symmetry and PROs after OHT correction at long-term follow-up. Patients who underwent OHT correction at a single institution from 2000 to 2024 were identified. Hypertelorism index (HI) and periorbital symmetry was compared preoperatively, 1-year postoperatively, and at long-term follow-up (average 9

Sotos syndrome previously known as cerebral gigantism is a genetic disease characterized by excessive growth from the prenatal stage to childhood, accompanied with distinctive facial dysmorphism (broad and prominent forehead, hypertelorism, downward-slanting palpebral fissures, and a pointed chin), advanced bone age, macrocephaly, congenital malformations (heart defects, seizures, scoliosis, increased tumor susceptibility) and developmental delay (speech delay, learning disabilities, infantile hypotonia). Most cases of Sotos syndrome (90%) are linked to heterozygous mutations of the NSD1 gene.

A male infant born at full term presented with bilateral upper eyelid colobomas, absent upper puncta, and fusion of the eyelids to the globes. He was admitted for management of bilateral exposure keratopathy and a central corneal ulcer in the right eye. Further examination revealed multiple congenital abnormalities, such as penile torsion, unusual hair growth pattern on the forehead, bifid nasal tip, and hypertelorism. Genetic testing confirmed a diagnosis of Manitoba oculo-tricho-anal syndrome. Because conservative management of the ocular surface was insufficient, bilateral upper eyelid reco
