OS odontoideum in an achondroplastic adult - A case report and review.

BACKGROUND: Achondroplasia is a rare genetic disorder affecting bone development, resulting in disproportionate short stature. The coexistence of os odontoideum (OO) in individuals with achondroplasia is exceptionally uncommon. CASE DESCRIPTION: We present the case of a 29-year-old female with achondroplasia who experienced progressive weakness in the extremities, ultimately leading to difficulty in ambulation. Radiographic evaluation revealed an atlantoaxial dislocation secondary to an unstable OO. Surgical stabilization of C1-C2 resulted in gradual yet significant neurological recovery. CO

Navepegritide: First Approval.

Navepegritide (YUVIWEL®) is a prodrug of C-type natriuretic peptide (CNP) developed by Ascendis Pharma for the treatment of achondroplasia and is being investigated for hypochondroplasia. Navepegritide is administered once weekly and designed to provide sustained release and continuous exposure of active CNP, which is needed to counteract the overactive fibroblast growth factor receptor 3 signaling in achondroplasia, resulting in improved growth velocity. Navepegritide recently received accelerated approval in the USA for increasing linear growth in paediatric patients aged ≥ 2 years with acho

Arylsulfatase L is a Golgi chondroitin sulfatase regulating skeletal development.

Sulfatases are a family of enzymes that hydrolyze sulfate esters from various substrates. Defects, in sulfatase activity, are associated with various human diseases due to the accumulation of sulfated substrates. Deficiency in ARSL, a Golgi sulfatase, is associated with X-linked recessive chondrodysplasia punctata (CDPX), a disorder characterized by defects in cartilage and bone development. However, until now, ARSL function has remained unknown. In this study, we demonstrate that ARSL promotes 4-O-desulfation of chondroitin sulfate (CS) during proteoglycan biosynthesis. Numerical exploration of low lunar, long-lifetime orbits.

Head and neck squamous cell carcinoma (HNSCC) arises from the epithelial lining of the oral cavity, hypopharynx, oropharynx, and larynx. There are several potential risk factors that cause the generation of HNSCC, including cigarette smoking, alcohol consumption, betel quid chewing, inadequate nutrition, poor oral hygiene, HPV and Epstein-Barr virus, and Candida albicans infections. HNSCC has causative links to both environmental factors and genetic mutations, with the latter playing a more critical role in cancer progression. These molecular changes to epithelial cells include the inactivatio

Beyond the surface: plasmalogens are dispensable for retinal integrity and fertility in the mouse.

Ether lipids and their subclass, the plasmalogens, are critical regulators of membrane organization, signaling, and stress responses in multiple tissues. Inborn errors in their anabolism cause severe multi-organ diseases such as Rhizomelic Chondrodysplasia Punctata and related peroxisomal disorders. The Gnpat knockout mouse model, characterized by total ether lipid deficiency, recapitulates key features of this disorder, including dense bilateral cataracts, microphthalmia, and infertility, but the specific contribution of different subclasses, particularly plasmalogens, remains elusive. Update and review of treatment in achondroplasia.

INTRODUCTION: Achondroplasia is the most common skeletal dysplasia associated with disproportionate short stature, with an estimated prevalence of 4.6 per 100 000 births. It is caused by a pathogenic variant in the gene encoding fibroblast growth factor receptor 3 (FGFR3), which disrupts endochondral ossification of the growing skeleton. OBJECTIVE: To provide an updated overview of the therapeutic approach to achondroplasia, including currently approved treatments and those under investigation. CONTENT: The approval of vosoritide in 2021 has brought a major shift in the management of these p

X-linked spondyloepiphyseal dysplasia tarda misdiagnosed as growth hormone deficiency: identification of a novel intronic TRAPPC2 variant by whole-genome sequencing.

BACKGROUND: X-linked spondyloepiphyseal dysplasia tarda (SEDT) is a rare skeletal dysplasia caused by pathogenic variants in TRAPPC2 and typically presents in late childhood or adolescence with short-trunk disproportion and vertebral dysplasia. CASE PRESENTATION: We describe a family series centered on an adolescent male initially diagnosed with GHD due to reduced height velocity and subnormal GH stimulation results, who received recombinant human GH (rhGH) therapy for three years with negligible improvement. During puberty, he developed progressive short-trunk disproportion and characteristi

Natural history of radiological coronal alignment of lower limbs in children and adolescents with achondroplasia.

AIMS: Achondroplasia (ACH) is the most common skeletal dysplasia worldwide, caused by mutations in the fibroblast growth factor receptor 3 gene and characterized by a disproportionately short stature, and many orthopaedic complications. Genu varum affects between 80% and 90% of children with ACH, but the longitudinal natural history of lower limb alignment remains unclear. The aim of this study was to describe age-related changes in lower limb alignment in children with ACH by analyzing longitudinal radiological parameters from infancy to skeletal maturity. METHODS: This retrospective cohort

X-linked ARSL chondrodysplasia punctata: expanding the spectrum with a severe multisystem phenotype.

OBJECTIVES: X-linked chondrodysplasia punctata type 1 (CDPX1) is a rare skeletal dysplasia caused by pathogenic variants in ARSL (Arylsulfatase L), formerly known as ARSE (Arylsulfatase E), resulting in defective arylsulfatase E activity and abnormal cartilage matrix formation. It is typically defined by stippled epiphyses, nasomaxillary hypoplasia, and brachytelephalangy, but the condition has phenotypic variability. We report an infant with a pathogenic ARSL variant (NM_000047.3:c.1442C>T; p.Thr481Met) to highlight an exceptionally severe and complex phenotype. CASE PRESENTATION: A male inf

Assessing signs and impacts of achondroplasia: psychometric evaluation of the Achondroplasia Child Experience Measures.

BACKGROUND: Achondroplasia, although rare, is the most common genetic form of skeletal dysplasia and is associated with a wide range of clinical manifestations and complications throughout the body. This study evaluated the psychometric measurement properties of the Achondroplasia Child Experience Measures (ACEMs), which were previously conceptually developed as condition-specific, observer-reported outcome measures to assess the signs (ACEM-OSM) and impacts (ACEM-Impact) of achondroplasia in children aged 2 to < 12 years. METHODOLOGY: Data for the psychometric evaluation were from a non-inte

Early initiation of therapy with vosoritide: case report. Description of the first Italian patient affected by achondroplasia treated with vosoritide before 2 years of age.

Achondroplasia, the most common skeletal dysplasia, is mainly characterized by disproportionate short stature. Affected patients present a reduced linear growth, and the greatest decrease in growth velocity is evident in infancy. Vosoritide is the first approved drug to improve linear growth in children affected by achondroplasia. Since 2023, it is approved in children from birth by the FDA, and from 4 months of age by the EMA. However, until October 2025, the prescription and reimbursement of vosoritide in Italy was only possible starting from 2 years of age. We present the case of the first
