Real-world patterns of growth hormone treatment in Japanese children with short stature associated with small for gestational age, Turner syndrome, Noonan syndrome, achondroplasia, and hypochondroplasia: a retrospective database study.

In Japan, GH treatment (GHT) is used for short stature associated with small for gestational age (SGA), Noonan syndrome (NS), Turner syndrome (TS), achondroplasia (ACH), and hypochondroplasia (HCH). However, real-world data on these diagnoses and GHT use for these conditions in Japan remain limited. In this retrospective cohort study, we used the JMDC claims database (2005-2021) to extract data of children with SGA-related short stature or diagnosed with short stature diseases, namely NS, TS, ACH, or HCH, particularly in boys aged <17 years and girls aged <15 years. We reported the prevalence,

Diagnostic guide to hypochondroplasia.

Hypochondroplasia is a skeletal dysplasia characterized by disproportional short stature with rhizomelic limb shortening, caused by pathogenic variants of FGFR3, most frequently the p.Asn540Lys variant. However, affected individuals harbor a wide variety of pathogenic variants, accounting for the broad phenotypic spectrum of the disorder. Despite being closely related to achondroplasia, hypochondroplasia is a milder condition that was previously believed to be recognizable only in childhood, but not in infancy. However, the widespread use of prenatal ultrasonography and frequent diagnosis of f

Growth plate senescence, chondrocyte biology, and the growth hormone-insulin-like growth factor 1 axis in childhood growth.

Longitudinal bone growth in children depends on endochondral ossification within the epiphyseal growth plate, a process long viewed as a passive readout of systemic hormones but now recognized as an actively regulated, self-limiting process. This mini-review synthesizes recent evidence on chondrocyte biology, growth plate senescence, and the growth hormone (GH)-insulin-like growth factor 1 (IGF-1) axis. Local paracrine networks establish precise antagonistic and synergistic interactions: fibroblast growth factor receptor 3 (FGFR3) suppresses chondrocyte proliferation via MAPK/STAT signaling, w

Consensus statement on the management of Achondroplasia in the Middle East.

Achondroplasia is the most common skeletal dysplasia and presents as a lifelong, multi-system condition characterized by disproportionate short stature, rhizomelic limb shortening, macrocephaly with frontal bossing, and a spectrum of neurological, respiratory, orthopedic, and psychosocial complications. Effective management requires anticipatory surveillance, coordinated multidisciplinary care, and early recognition of life-threatening complications. Therapeutic options have expanded in recent years with the introduction of vosoritide, a C-type natriuretic peptide analog that targets the under

Cardiovascular risk in achondroplasia: a systematic review.

BACKGROUND: Achondroplasia is the most common form of disproportionate short stature and is associated with reduced life expectancy. It is not clear to what extent cardiovascular disease (CVD) is responsible for this. The primary aim of this systematic review was to identify the prevalence of CVD in individuals with achondroplasia. METHODS: A systematic review of the literature was conducted in accordance with PRISMA guidelines by two independent reviewers using two databases. There were no language or date restrictions. The search strategy consisted of the terms: "achondroplasia" AND "vascul

Functional analysis of NPR2 variants supports the therapeutic rationale for CNP in short stature.

Human height is a complex trait regulated by genetic and environmental factors. Several components of chondrocyte proliferation and differentiation regulatory pathways are implicated in short-stature disorders; one such component is natriuretic peptide receptor 2 (NPR2), the receptor for C-type natriuretic peptide. We developed a high-throughput GFP reporter assay to evaluate the functional impact of NPR2 missense variants on receptor activity, which was utilized alongside a commercially available CatchPoint assay, and previously published data. Measurements from these sources were integrated

Homozygous Achondroplasia With Long-Term Survival: Growth Patterns, Medical Interventions, and Practice Implications.

Homozygous achondroplasia is widely considered perinatal lethal by the medical community. In this case series, we report two children from a single family with longer-term survival. One child lived for 17 months and the other was 60 months at the time of publication. We describe two siblings born to parents with achondroplasia with homozygous achondroplasia and long-term survival.

An overview of the International Consensus Statement on achondroplasia.

Achondroplasia is the most common form of short-limbed short stature of genetic origin. Most people with achondroplasia live fully independent, productive, and socially engaged lives. However, the condition is associated with several potential medical complications. Individuals with achondroplasia may experience medical, functional, and psychosocial challenges at different times in their life. The goal for lifelong care of individuals with achondroplasia is to optimize their physical and mental health through provision of individualized care and to promote participation and inclusion in societ

Foramen magnum stenosis, cervicomedullary decompression, and growth in children with achondroplasia: a retrospective cohort study.

BACKGROUND: Foramen magnum stenosis (FMS) is a serious complication in children with achondroplasia that may necessitate cervicomedullary decompression (CMD). It is unclear how FMS and CMD influence growth in these children. This study aimed to assess the effects of FMS and CMD on the growth of children with achondroplasia. METHODS: Eighty-seven children (45 males, 42 females) with achondroplasia, aged 4 to 6 years, were evaluated. Height, weight, head circumference, and body mass index were expressed as standard deviation scores (SDS) according to Merker et al., while sitting height SDS was d

The CrescNet Registry Achondroplasia Module: Real-World Demographic Data and Clinical Outcomes in Untreated and Vosoritide-Treated Individuals.

INTRODUCTION: Achondroplasia is a rare skeletal dysplasia characterized by severe disproportionate short stature. Vosoritide is currently the only approved therapy. The CrescNet registry is a network of primary and specialized pediatric tertiary centers that aims to improve early detection of growth disorders in Europe. In 2021, an achondroplasia-specific data collection module was set up within CrescNet to enhance data collection among children with achondroplasia and assess the impact of interventions. Here, we describe the module setup and report preliminary real-world outcomes of vosoritid

Foramen Magnum Stenosis in Achondroplasia: Imaging-Based Surgical Indications, Synchondrosis Fusion, and the Role of Ventriculomegaly.

INTRODUCTION: Foramen magnum stenosis (FMS) and ventriculomegaly are frequent in achondroplasia due to cranial base hypoplasia. While FMS can cause spinal cord compression, the optimal treatment strategy remains debated. We reviewed our single-center experience and explored factors influencing surgical decisions. METHODS: We retrospectively analyzed 18 children with achondroplasia evaluated between 2008 and 2024. MRI was used to assess FMS by anterior-posterior diameter and cerebrospinal fluid (CSF) area at the foramen magnum level. Ventriculomegaly was defined as an Evans Index ≥0.30. CT was

METTL16 Modulates GPX4 Expression to Regulate Chondrocyte Ferroptosis.

BACKGROUND: Achondroplasia (ACH), the predominant inherited form of disproportionate short stature, results from specific genetic alterations in fibroblast growth factor receptor 3 (FGFR3). N6-methyladenosine (m6A) modification is reported to modulate mRNA stability and translation. The present investigation systematically explored the epigenetic regulatory function of METTL16, an m6A RNA methyltransferase, within the pathophysiological framework of ACH. METHODS: We generated an ACH mouse model via Fgfr3380R (Fgfr3ach) gene mutation. Primary chondrocytes were isolated from newborn mice and st

Effect of vosoritide on genu varum in children with achondroplasia after 1 year in randomized placebo-controlled trials.

Achondroplasia is a skeletal dysplasia associated with multisystem complications including genu varum, which causes pain and limits mobility. Vosoritide, a targeted treatment for achondroplasia, improves growth in children and has an established safety profile, but its effect on genu varum is unclear. Data were collected from 183 participants from randomized, double-blind, placebo-controlled phase 2 (CANOPY ACH-2I [111-206; NCT03583697]) and phase 3 (CANOPY ACH-3 [111-301; NCT03197766]) studies evaluating vosoritide (weight-based dose of 15 or 30 µg/kg/day) in children aged 0-5 and >5 years, r

Maternal decision-making through temporal uncertainties: The anticipatory biopolitics of Vosoritide in dwarfism communities.

Vosoritide, a biotechnological therapy designed to increase growth in children with achondroplasia, has introduced new pressures and bodily possibilities for families navigating this rare genetic condition. While debates around its use often centre on its efficacy as a non-surgical growth treatment for the most common form of non-lethal human dwarfism, far less attention has been paid to how the medication (re)shapes the temporal landscape of maternal decision-making, children's bodily autonomy, and community dynamics. Drawing on qualitative interviews with mothers from UK dwarfism communities
