Effect of vosoritide on spine morphology in children with achondroplasia: 1-year results from a randomized phase 2 study.

Achondroplasia is a skeletal dysplasia condition caused by reduced endochondral ossification resulting in disproportionate short stature and skeletal deformities, including thoracolumbar kyphosis (TLK) and spinal stenosis. Vosoritide, the first and only approved targeted therapy for achondroplasia, increases bone growth, but its impact on spinal morphology has not been assessed. The randomized, double-blind, placebo-controlled phase 2 CANOPY ACH-2I study (111-206; NCT03583697) evaluated the safety and efficacy of vosoritide in 75 children aged 0 to <5 years. Interpedicular distance (IPD), sagi

Practical guidance for clinicians to optimise orthopaedic outcomes in achondroplasia: International Achondroplasia Forum guiding principles.

BACKGROUND: Achondroplasia is associated with multisystem impairments that may require surgical interventions associated with high complication rates. The orthopaedic treatment landscape in achondroplasia varies globally, with some countries recommending surgical intervention more commonly. Achondroplasia-specific concerns related to preparation and conduct of surgical procedures necessitate careful consideration of treatment options in consultation with a multidisciplinary care team. The International Achondroplasia Forum convened a workshop in October 2024 to develop guiding principles for o

Multilevel Laminectomy for Lumbar Spinal Stenosis With Low Back Pain in Achondroplasia: A Case Report.

BACKGROUND Achondroplasia is an autosomal dominant skeletal dysplasia and the most common genetic cause of dwarfism, characterized by shortened pedicles, thickened laminae, and reduced interpedicular distances that lead to congenital narrowing of the spinal canal. These anatomical abnormalities frequently result in lumbar spinal stenosis (LSS) requiring surgical management. Surgical treatment typically involves decompression of neural elements, with or without fusion, to restore canal patency and prevent postoperative deformity. However, adjusting the surgical approach to the unique anatomical

Dkk1 inhibition restores mandibular growth in an achondroplasia mouse model.

Meckel's and condylar cartilages are key to mandible development, with Meckel's cartilage acting as a template and condylar cartilage as a growth center. In achondroplasia, the most common form of genetic dwarfism, abnormalities of these cartilages lead to micrognathia, with significant functional repercussions for affected individuals. How FGFR3 overactivation in achondroplasia disrupts Meckel's and condylar cartilages is largely unknown. Our aim was to identify the pathways driving these disruptions by analyzing the genes expressed in these cartilages in a mouse model mimicking achondroplasi

C2-C3 anterolisthesis with myelopathy in a toddler with Charcot-Marie-Tooth disease: a rare case and review of syndromic pediatric C2-C3 instability.

INTRODUCTION AND IMPORTANCE: Cervical instability and myelopathy are rare in young children and are frequently overlooked, particularly in those with syndromic or neuromuscular disorders, in which motor delay may be misattributed to the underlying condition. Reported cases of C2-C3 instability are extremely uncommon and have primarily been associated with syndromic diagnoses such as Down syndrome and achondroplasia. Recognizing treatable cervical pathology early is essential to prevent progressive neurological injury. CASE PRESENTATION: We describe the first reported case of C2-C3 anterolisth

Efficacy and safety of vosoritide in children with achondroplasia: a systematic review and meta-analysis.

UNLABELLED: Achondroplasia is caused by a gain-of-function mutation in the FGFR3 gene. Vosoritide activates the NPR-B receptor to inhibit the overactive FGFR3 signaling pathway. We aim to pool the efficacy and safety outcomes of vosoritide in children with genetically confirmed achondroplasia who are receiving the approved dose of 15 μg/kg/day. We searched five databases up to February 10, 2026. A systematic review and single-arm meta-analysis were conducted in accordance with the PRISMA guidelines. The primary outcomes were annualized growth velocity (AGV), height gain, and change in height Z

Foramen magnum decompression in infants and children with achondroplasia: clinical outcomes from a 15-patient surgical series.

BACKGROUND: Achondroplasia (ACH) is the most common skeletal dysplasia characterized by disproportionate short stature due to impaired endochondral ossification. One of the most critical and potentially fatal complications of ACH is foramen magnum and upper cervical canal stenosis. Compression at the cervicomedullary junction may lead to myelopathy, hypotonia, developmental delay, and central sleep apnea. Early detection and timely surgical intervention are essential to prevent permanent neurological injury. METHODS: This retrospective study evaluated 15 pediatric patients with ACH (9 girls,

Linking shox/shox2 deficiency with fgfr3 gain-of-function and natriuretic peptides.

Short stature homeobox (SHOX) and its close paralogue SHOX2 are important transcriptional regulators of developmental processes. To understand how they function both individually and as an ensemble, we examined their mutual interdependence using zebrafish as a model. As both genes maintain important roles in various tissues, we assessed their expression patterns in the whole embryo as well as separately in fins, heart and brain. Fibroblast growth factor receptor 3 (FGFR3) is a direct target of SHOX, and FGFR3 mutations cause achondroplasia, a condition treatable with C-type natriuretic peptide

Premature Consolidation of Regenerated Bone During Limb Lengthening with External Fixation in Achondroplastic Patients: Case Series and Literature Review.

Background: Premature consolidation of the regenerated bone is a relevant and underreported complication of limb lengthening in achondroplasia, with a reported incidence of 7-15% in the international literature. The gain-of-function fibroblast growth factor receptor 3 (FGFR3) mutation paradoxically enhances bone-healing capacity, predisposing patients to accelerated callus maturation during distraction osteogenesis and exposing patients to a risk of invasive procedures to treat this issue. This study reports the incidence and clinical features of premature consolidation in a cohort of achondro

Numerical exploration of low lunar, long-lifetime orbits.

Head and neck squamous cell carcinoma (HNSCC) arises from the epithelial lining of the oral cavity, hypopharynx, oropharynx, and larynx. There are several potential risk factors that cause the generation of HNSCC, including cigarette smoking, alcohol consumption, betel quid chewing, inadequate nutrition, poor oral hygiene, HPV and Epstein-Barr virus, and Candida albicans infections. HNSCC has causative links to both environmental factors and genetic mutations, with the latter playing a more critical role in cancer progression. These molecular changes to epithelial cells include the inactivatio

Update and review of treatment in achondroplasia.

INTRODUCTION: Achondroplasia is the most common skeletal dysplasia associated with disproportionate short stature, with an estimated prevalence of 4.6 per 100 000 births. It is caused by a pathogenic variant in the gene encoding fibroblast growth factor receptor 3 (FGFR3), which disrupts endochondral ossification of the growing skeleton. OBJECTIVE: To provide an updated overview of the therapeutic approach to achondroplasia, including currently approved treatments and those under investigation. CONTENT: The approval of vosoritide in 2021 has brought a major shift in the management of these p
