AIMS: Achondroplasia (ACH) is the most common skeletal dysplasia worldwide, caused by mutations in the fibroblast growth factor receptor 3 gene and characterized by a disproportionately short stature, and many orthopaedic complications. Genu varum affects between 80% and 90% of children with ACH, but the longitudinal natural history of lower limb alignment remains unclear. The aim of this study was to describe age-related changes in lower limb alignment in children with ACH by analyzing longitudinal radiological parameters from infancy to skeletal maturity.
METHODS: This retrospective cohort

BACKGROUND: Achondroplasia, although rare, is the most common genetic form of skeletal dysplasia and is associated with a wide range of clinical manifestations and complications throughout the body. This study evaluated the psychometric measurement properties of the Achondroplasia Child Experience Measures (ACEMs), which were previously conceptually developed as condition-specific, observer-reported outcome measures to assess the signs (ACEM-OSM) and impacts (ACEM-Impact) of achondroplasia in children aged 2 to < 12 years.
METHODOLOGY: Data for the psychometric evaluation were from a non-inte

BACKGROUND: Parents of children with achondroplasia face sustained caregiving demands that may affect multiple dimensions of well-being. Despite growing recognition of these challenges, no validated, condition-specific instrument exists to assess the quality of life (QoL) of parents of children with achondroplasia. This study aimed to develop, and pilot test the Quality of Life of Parents of Children with Achondroplasia (QOLA) questionnaire.
METHODS: QOLA was developed using a multi-phase mixed-methods design in accordance with established standards for developing self-reported outcome measur

BACKGROUND: Achondroplasia is a genetic skeletal condition caused by FGFR3 pathogenic variants. Infigratinib, an oral FGFR1-3 tyrosine kinase inhibitor, down-regulates key pathways in the pathogenesis of achondroplasia.
METHODS: In this phase 3, multicenter, double-blind, placebo-controlled trial, we randomly assigned children with achondroplasia (3 to 17 years of age) in a 2:1 ratio to receive infigratinib (at a dose of 0.25 mg per kilogram of body weight) or placebo once daily for 52 weeks. The primary end point was the change from baseline in the annualized height velocity in the infigrati

Very few articles have investigated pregnancy in women with achondroplasia. A scoping review methodology was used to record and summarize the existing research evidence. The review process was conducted in accordance with the PRISMA-ScR guidelines. Original studies and case series mentioning achondroplasia with a minimum of 5 pregnancies were included. A total of 1527 articles were screened by title and abstract. Twenty-one full-text articles were reviewed for eligibility criteria. Seven studies were included. The reference details, study characteristics, topics of interest, and main findings

Hypochondroplasia is a rare genetic condition that causes disproportionate short stature. This retrospective, real-world, matched-cohort study used electronic health records data from the Clinical Practice Research Datalink Aurum primary care database, linked to Hospital Episode Statistics and Office of National Statistics mortality records, to assess the natural history of hypochondroplasia in England from 1998 to 2019. A total of 549 participants with hypochondroplasia and 2196 matched controls were included. Mean follow-up time was 7.50 years for participants with hypochondroplasia and 11.6

Achondroplasia, the most common skeletal dysplasia, is mainly characterized by disproportionate short stature. Affected patients present a reduced linear growth, and the greatest decrease in growth velocity is evident in infancy. Vosoritide is the first approved drug to improve linear growth in children affected by achondroplasia. Since 2023, it is approved in children from birth by the FDA, and from 4 months of age by the EMA. However, until October 2025, the prescription and reimbursement of vosoritide in Italy was only possible starting from 2 years of age. We present the case of the first

INTRODUCTION: Achondroplasia is characterized by disproportionate short stature, yet little is known about how growth patterns and body composition differ from peers during childhood and adolescence. We aimed to compare anthropometric variables and detailed body composition profiles between children and adolescents with achondroplasia and age-matched controls.
METHODS: In this cross-sectional study, anthropometry and body composition were assessed in participants aged 6-18 years using air displacement plethysmography (BOD POD). Height, weight, BMI, fat mass (FM), fat mass index (FMI), fat-fre

Vosoritide, a recombinant C-type natriuretic peptide analog that antagonizes fibroblast growth factor receptor 3 (FGFR3) signaling, is recommended for early initiation in infants with achondroplasia. However, clinical safety data for infants under six months remain limited, and concerns persist regarding its potential to induce symptomatic hypotension. We present a case of a three-month-old female infant with genetically confirmed achondroplasia (FGFR3 p.Gly380Arg) who accidentally received an inadvertent double dose of vosoritide (0.24 mg; 53 μg/kg) during home self-injection therapy. BACKGROUND: Growing-spine surgery aims to promote thoracic growth in skeletally immature patients by achieving adequate T1-T12 length. Surgical decision-making often relies on assumptions regarding spinal length thresholds and their relationships to cardiopulmonary function. This study aimed to characterize thoracic (T1-T12) and total spine (T1-S1) lengths in skeletally mature patients with skeletal dysplasias, with a primary focus on achondroplasia, and to evaluate whether these measurements were associated with cardiopulmonary comorbidities.
METHODS: A retrospective review was conducted of

Background: Achondroplasia, the most common genetic dwarfism caused by the FGFR3 mutation (autosomal dominant, 80% de novo), results in a disproportionately short stature. Thoracolumbar kyphosis (TLK), combined with characteristic spinal canal stenosis, increases the risk of symptomatic compression, yet the literature lacks clear thresholds for symptom onset or progressive deformity angles. Methods: A 16-year-old female with achondroplasia presented with rapidly progressive kyphosis despite conservative management (bracing and therapy). Over six months, she developed neurogenic claudication; b

PURPOSE: To systematically review the efficacy, safety, and outcomes of limb lengthening procedures in patients with achondroplasia, including effects on quality of life.
METHODS: Following PRISMA guidelines, a systematic review and meta-analysis was performed. Eligible studies included patients with achondroplasia who underwent limb lengthening of the upper and/or lower extremities. Data were extracted on length gain, external fixator index, fixation duration, complications, and quality of life. Pooled means and 95% confidence intervals (CIs) were calculated using single-arm meta-analysis.

Achondroplasia is the most common skeletal dysplasia, characterized by disproportionate short stature and associated craniofacial, musculoskeletal, and neurological abnormalities. Respiratory complications are frequent and represent a major contributor to morbidity and mortality. Sleep-disordered breathing (SDB), encompassing obstructive (OSA) and central sleep apnea (CSA), is the most prevalent respiratory complication with pediatric prevalence reported between 22% and 93%. OSA results primarily from multilevel upper-airway obstruction related to craniofacial abnormalities and adenotonsillar
