microRNA and Regulatory Networks of Human Macrophage During Leishmania major In Vitro Infection.

Leishmania (L.) major modulates host microRNA (miR) expression to promote intracellular survival, yet the roles of specific miRs during early infection of human macrophages remain poorly defined. We investigated the dynamic regulation of miR-21-5p and miR-146a-5p in THP-1-derived macrophages infected with L. major over an early post-infection time course (3-48 h), alongside miR-30a-3p and miR-101-3p. Both miR-21-5p and miR-146a-5p were significantly upregulated during the early stages of infection (3-24 h) before declining by 48 h. Using antisense LNA inhibitors, we achieved independent single

The mevalonate and methylerythritol phosphate pathways as therapeutic targets in protozoan parasites: from moonlighting enzymes to metabolic plasticity.

The interplay between metabolic pathways and immune responses is critical for understanding host-parasite interactions in protozoan infections. This review examines the mevalonate (MVA) and methylerythritol phosphate (MEP) pathways-key routes for isoprenoid synthesis essential for sterol production and bioactive compounds. Beyond sustaining parasite survival, these pathways directly influence host immune signaling cascades, including NF-κB, MAPK, PI3K/Akt, and mTOR. A critical conceptual distinction is introduced: genuine "moonlighting" functions-where a single enzyme performs two distinct mol

Immune-mediated remodelling of tissue architecture in Post-Kala-Azar Dermal Leishmaniasis (PKDL).

BACKGROUND: Post-Kala-Azar Dermal Leishmaniasis (PKDL), a dermal sequel of apparently cured visceral leishmaniasis (VL), presents with macular or polymorphic lesions. A relationship, if any, between immune cell infiltration and cytokines potentially causing tissue-remodelling remains poorly defined, and was the focus of this study. METHODOLOGY/PRINCIPAL FINDINGS: Skin biopsies along with plasma from PKDL patients and healthy controls were analyzed. Tissue sections were assessed for immune cell subsets (CD8 ⁺ T cells, CD68 ⁺ macrophages, CD20 ⁺ B cells, CD66b⁺ neutrophils), associated cytokine

Testosterone Regulates Sex-Dependent Immune Responses and Androgen Receptor Expression During Plasmodium berghei ANKA Infection.

Malaria exhibits pronounced sexual dimorphism, with males experiencing greater disease severity and mortality than females do. Although these differences have been partially attributed to the immunosuppressive effects of testosterone, the underlying mechanisms remain poorly understood. Here, we investigated the role of testosterone in sex-specific immune responses by administering a combination of letrozole and testosterone (LT) to male and female mice infected with Plasmodium berghei ANKA. LT treatment increased testosterone concentrations in both sexes and was associated with higher parasite

Highlights in Cysteine Protease Inhibitors as Antiparasitic Agents Against Trypanosomatids.

Trypanosoma cruzi, Trypanosoma brucei and Leishmania spp. are the causative agents of Chagas disease, human African trypanosomiasis, and leishmaniasis, respectively. These three neglected tropical diseases affect millions of individuals globally, and current therapeutic options remain severely limited. The major cysteine proteases (CPs) of these parasites play critical roles in their infectivity and survival, making them subjects of prolonged research. Therefore, cruzain, TbrCATL and CPB from Leishmania spp. have been recognized as the primary targets of most drugs under investigation. Among t

[Cutaneous coinfection with tuberculosis and leishmaniasis].

We describe the case of a 16-year-old female patient from Bolivia, with a family history of pulmonary tuberculosis, who presented with a two-month history of ulcerated lesions on the dorsum of her left hand. A skin biopsy revealed amastigotes consistent with Leishmania spp., and antiparasitic treatment was initiated. Subsequently, Mycobacterium tuberculosis was also isolated, and antituberculosis treatment was started, resulting in a favorable outcome.

Temporal Neuroimmune Changes During Primary and Persistent Experimental Toxoplasmic Encephalitis.

Toxoplasma gondii persists in the central nervous system and may cause severe toxoplasmic encephalitis, particularly in immunocompromised hosts. Our previous studies demonstrated that T. gondii-induced neuropathology involves oxidative and nitrosative stress, apoptosis, glial activation, blood-brain barrier injury, and GMF-β- and SAP/NF-κB-associated responses. This study investigated the temporal immunoreactivity of CD68, S100B, Foxp3, and IL-5 during early and chronic experimental infection. Twenty-four female Swiss albino mice were assigned to one uninfected control group and three ME49-inf

In Situ Cytokine Expression, Nitric Oxide and Reactive Oxygen Species Production During Primary Amoebic Meningoencephalitis Development.

Primary amoebic meningoencephalitis (PAM), caused by Naegleria fowleri, is a rapidly progressive and fatal infection, and inflammation of the olfactory bulb plays a key role in disease pathogenesis. Although inflammatory and oxidative mediators have been studied in vitro, their production during the progression of PAM remains poorly understood. This study evaluated the temporal production of TNF-α, IL-1β, IL-6, nitric oxide (NO), and reactive oxygen species (ROS) in the olfactory bulbs of BALB/c mice from 1 to 7 days after intranasal infection with N. fowleri trophozoites. RT-PCR detected incr

Bacterial infections and antibiotic susceptibility in children with sickle-cell anemia on malaria chemoprevention using Dihydroartemisinin-piperaquine (DP) or Sulfadoxine-pyrimethamine (SP) in Uganda.

INTRODUCTION: Over 300,000 babies are born annually with sickle cell anemia (SCA) in sub-Saharan Africa. Despite malaria chemoprevention and early penicillin prophylaxis, these children develop febrile illnesses. The effect of malaria chemoprevention regimens on the incidence of bacterial infections and antimicrobial resistance is unclear. We assessed incidence of bacterial infections and resistance patterns in children with SCA receiving weekly DP or monthly SP. METHODS: This study was nested in a randomized, double-blind clinical trial comparing weekly DP with monthly SP. Participants were

Assessing the Effect of Malaria Exposure History on Tetanus Antibody Waning Rates Among Children in Jinja and Tororo Districts, Uganda.

BACKGROUND: Malaria causes immune perturbations and affects immunogenicity to vaccines. It is unknown, however, how repeated malaria infections affect the duration of vaccine protection. This study assessed variability in tetanus antitoxin immunoglobulin G (IgG) antibody kinetics including by malaria exposure history at district and individual levels. METHODS: We analyzed tetanus antitoxin IgG antibody concentrations in 3064 plasma samples from 2011 to 2017 in 597 children aged 6 months to 10 years residing in the Tororo (high malaria transmission) and Jinja (low transmission) districts in Ug

The mosquito gut microbiome and Plasmodium development: a scoping review of molecular approaches and implications for malaria vector control.

BACKGROUND: Malaria remains a major health burden, with an estimated 282 million cases and approximately 610,000 deaths recorded in 2024. The increasing prevalence of insecticide-resistant Anopheles vectors and drug-resistant Plasmodium strains has substantially eroded the efficacy of conventional control strategies, underscoring the critical need for innovative approaches. Accumulating evidence suggest that the mosquito midgut microbiome exerts a significant influence on Plasmodium sporogonic development; however, no scoping investigation has fully mapped the metagenomic evidence base in this

Circulating inflammatory mediators CCL2 and CXCL16 as biomarkers in Iraqi women infected with toxoplasmosis.

The protozoan parasite Toxoplasma gondii is the cause of the widespread but sometimes disregarded parasitic illness toxoplasmosis. Cytokines play an indispensable role in fostering an environment that is more conducive to pregnancy. Their intricate network of interactions allows for the observation of numerous modulatory consequences, such as cell migration, invasion, and gene expression. Among these, CCL2 (MCP-1) and CXCL16 have emerged as promising immunological biomarkers reflecting disease activity, inflammation, and tissue involvement. Monocyte chemotactic protein 1 (MCP-1), another name

Case Report: Multiple brain abscess-like lesions with concurrent detection of Nocardia amamiensis and Leishmania infantum in cerebrospinal fluid.

Central nervous system nocardiosis is rare and potentially life-threatening, and human infection due to Nocardia amamiensis has seldom been reported; central nervous system involvement in visceral leishmaniasis is likewise uncommon. We report a 69-year-old man who developed progressive impairment of consciousness and coma after recurrent fever. Brain magnetic resonance imaging revealed multiple abscess-like lesions in both cerebral hemispheres, the cerebellum, and the brainstem, accompanied by acute or subacute cerebral infarctions. The cerebrospinal fluid opening pressure was 300 mmH2O. The w

Contemporary Anopheles vector species distribution in Scotland and its association with historical hotspots of malaria transmission.

BACKGROUND: Malaria poses a significant threat to public health in tropical regions, with nearly half of the global population at risk. Historically, transmission was common in temperate regions including northwestern Europe, with records indicating that malaria was prevalent in Scotland until the early nineteenth century. While current environmental and epidemiological conditions are unsuitable for transmission, future climate, environmental, and land-use change may alter the receptivity of Scotland to malaria and other mosquito-borne diseases. As a first step to assess the potential for futu

New thinking for the next generation of antimalarials.

Despite decades of intensive research and substantial clinical gains, malaria remains a major global health challenge exacerbated by the continued emergence of drug-resistant strains of the causative agent, Plasmodium parasites. Encouragingly, recent advances in functional genomics, chemical biology and computational science are reshaping antimalarial drug discovery. In this review, we examine the discovery and development of next-generation antimalarials, including advances in phenotypic and target-based screening, omics-enabled target discovery and emerging therapeutic modalities such as lon

Machine learning-based ecological prediction of maternal mortality ratio in malaria-endemic Africa.

BACKGROUND: Malaria in pregnancy remains a significant public health concern in sub-Saharan Africa. It may contribute to population-level maternal mortality through adverse maternal outcomes. This study aims to develop a machine learning framework to predict maternal mortality ratio (MMR) across African countries. METHODS: A supervised machine learning framework was applied to identified open source, country-level data across 52 African countries through (2000-2020). MMR was estimated using Support Vector Regression (SVR) model. Model performance was evaluated using a random 70/30 split of co

Strengthening geospatial surveillance for malaria control and elimination in Indonesia: a meeting report.

As Indonesia progresses toward malaria elimination, geospatial analysis offers opportunities to strengthen surveillance, improve intervention targeting, and support data-informed decision-making. However, despite increasing availability of georeferenced surveillance data, the routine use of geographic information systems (GIS) within malaria programs remains uneven. A three-day consultation and GIS training workshop was conducted in Jakarta, bringing together 25 representatives from National and Sub-National Malaria Programs and other disease control units. The workshop included hands-on R-bas

Systemic IgG responses to Leishmania braziliensis-derived extracellular vesicles vary across clinical forms of American tegumentary leishmaniasis and associate with early therapeutic outcome.

BACKGROUND: American tegumentary leishmaniasis (ATL) caused by Leishmania braziliensis is characterized by marked clinical and immunopathological heterogeneity. While cellular immune responses have been extensively investigated, the contribution of humoral immunity, particularly against parasite-derived extracellular vesicles (EVs) components, remains poorly understood. Here, we evaluated systemic IgG responses against EVs derived from L. braziliensis isolates associated with distinct clinical forms of ATL and assessed their association with disease presentation and therapeutic outcome. Effects of centrifugation-based therapeutic plasma exchange on coagulation parameters in dogs with canine visceral leishmaniasis.

As in humans, therapeutic plasma exchange (TPE) has been investigated in veterinary medicine for managing immune-mediated diseases such as canine visceral leishmaniasis (CVL). TPE may improve clinical outcomes by removing circulating immune complexes and excess antibodies; however, its physiological effects must be understood to ensure safety. Given the scarcity of studies in animals, this study evaluated the effects of centrifugation-based TPE in dogs with CVL receiving fresh frozen plasma (FFP) as replacement fluid. Thirty-one dogs with CVL that failed to respond to conventional therapy were
