Malaria remains a leading cause of morbidity and mortality among children in Malawi with transmission exhibiting marked geographic disparities. While standard regression models estimate the association between childhood malaria and risk factors across the entire study area, they assume spatial stationarity, thereby masking local-area variations in estimated effects. We applied Geographically Weighted Regression, a spatial non-stationarity modeling approach to data from the recent 2024 Malawi Demographic and Health Survey to estimate district-specific associations between childhood malaria and

BACKGROUND: Integrating human behavioral data with data on malaria vector biting rates can help to quantify human exposure to malaria vectors and identify gaps in protection; however, to date, the availability of necessary human location and behavior data have been more limited as compared to that of corresponding entomological data. To expand the data available for use in vector-human data integrations, this case example explored the process and interpretations of integrating human behavior estimates from the Malaria Behavior Survey (MBS), a large-scale cross-sectional survey measuring behavi

Kenya recorded more than 3 million malaria cases in 2023. Insecticide resistance and changing mosquito behavior have reduced the effectiveness of insecticide-treated nets (ITNs) and indoor residual spraying. Spatial repellents (SRs) show promise for malaria control. The present social science study, which is nested within the Advancing Evidence for Global Implementation of Spatial Repellents clinical trial in Kenya, was conducted to explore SR acceptability and perceived efficacy. Modified trials of improved practices were used to assess participant response to Mosquito Shield™, a transfluthri

Jaundice, caused by the accumulation of bilirubin in plasma, is clinically interpreted as a maladaptive consequence of hemolysis or as indicative of hepatic failure. Drawing on genetic, biochemical, and clinical evidence, we propose to reframe jaundice as an adaptive response to malaria, a hemolytic disease caused by Plasmodium spp. infection. Bilirubin, the molecular basis of jaundice, represents an effector arm of metabolic immunity, distinct from nutritional immunity, which restricts pathogen access to essential nutrients, and from immunometabolism, which shapes immune cell function. Coccidiosis, caused by Eimeria spp., is one of the major enteric diseases affecting broiler chickens, compromising gut integrity and altering gut microbiota composition. This study aimed to evaluate the effects of different Fortegra® Eimeria spp. challenge doses on the composition and diversity of the cecal microbiota over time postinfection. Broiler chickens were allocated into three groups: a control group receiving PBS, G1 challenged with a 10 × vaccine dose, and G2 challenged with a 20 × vaccine dose. Cecal content samples were collected at 7, 14, and 21 days post-inoculation and analyzed

Chimeric recombinant antigens are used in canine Trypanosoma cruzi serology, but the timing of IgG reactivity against them after infection has not been described. We characterized the temporal profile of anti-T. cruzi IgG reactivity against four chimeric recombinant antigens (IBMP-8.1, IBMP-8.2, IBMP-8.3 and IBMP-8.4) in 36 mixed-breed dogs from a vaccine efficacy trial that were experimentally infected with T. cruzi. A total of 144 serum samples collected before infection and at 30, 60 and 180 days post-infection were tested by indirect ELISA. A previously established latent class analysis fr

Malaria remains a significant global health challenge with an estimated 282 million cases reported in 2024. CRISPR/Cas9-based gene-drive systems have emerged as promising tools to block Plasmodium transmission by mosquito vectors. The TP13 drive system targets the Anopheles gambiae cardinal (Agcd) gene and carries 2 engineered monoclonal antibodies to achieve rapid population modification to prevent parasite transmission. Previous cage trials demonstrated complete drive introduction in 3 to 6 generations and supported modeling predicting a potential >90% reduction in malaria incidence under op

Seasonal malaria chemoprevention (SMC) with sulfadoxine-pyrimethamine and amodiaquine (AQ) has been implemented in Africa since 2012 to prevent malaria among children ages 3 to 59 months old. As SMC expands to new regions, concerns are emerging about sustainability and quality of implementation. Successful SMC delivery depends on not only effective drug administration but also caregiver engagement, adherence to at-home AQ administration, retention of Child Record Cards, and overall malaria prevention practices. Although caregivers' perceptions of delivery quality as well as knowledge and trust

OBJECTIVES: To assess sub-national variation in malaria positivity among children under five in sub-Saharan Africa and examine whether maternal, household, structural, and healthcare-access factors were associated with higher malaria burden.
STUDY DESIGN: Cross-sectional.
METHODS: We used pooled Demographic and Health Survey data (2017-2023) from 10 sub-Saharan African countries to assess malaria positivity among children under five. Geospatial smoothing models were used to describe sub-national variation and identify areas of higher malaria positivity. Country-specific factors associated wi

Accurate malaria forecasting in sub-Saharan Africa is challenged by strong seasonality, reporting uncertainty, and non-stationary transmission dynamics. This study proposes a hybrid probabilistic forecasting framework that integrates Gaussian Process Regression (GPR) with Holt-Winters exponential smoothing for modelling monthly under-five malaria admissions in Ghana. GPR captures complex non-linear temporal behaviour and predictive uncertainty, while Holt-Winters smoothing stabilises long-horizon forecasts and preserves seasonal structure. Using ten years of district-level surveillance data (2

Chagas disease, caused by Trypanosoma cruzi, is endemic to the Americas but has spread worldwide due to migration. Benznidazole is effective, but long-term follow-up efficacy studies are scarce and include only small numbers of patients. We analyzed data from a retrospective cohort of Chagas disease patients treated with benznidazole at a large tertiary center in Argentina (1980-2019). Treatment response was assessed through clinical, parasitological, and serological follow-up. Parasitemia was evaluated by direct visualization or PCR. Among 567 treated patients, 464 met selection criteria, inc

BACKGROUND: Malaria rapid diagnosis tests (mRDTs) detecting Plasmodium falciparum histidine-rich protein 2 (Pfhrp2) are the most widely adopted diagnostic tool in malaria-endemic areas. However, extensive polymorphisms in Pfhrp2 could modify Pfhrp2 epitopes, rendering it undetectable by Pfhrp2-based mRDTs. This study investigates the spatial distribution and population dynamics of Pfhrp2 in sub-Saharan Africa (sSA).
METHOD: Pfhrp2 sequences (215) were retrieved from public repositories and analysed with MAFFT v7, DnaSP, and various packages in R. We computed genetic diversity metrics, neutral

Tsetse (Diptera: Glossinidae) are the exclusive cyclical vector of human and animal African trypanosomoses and continue to threaten health and agricultural productivity across sub-Saharan Africa. Area-wide integrated pest management (AW-IPM) strategies provide a coordinated framework for combining tools such as the sterile insect technique (SIT), insecticide-treated targets and cattle, and others. This review examines the success and challenges of AW-IPM in tsetse control and advances in population genetics, species distribution modeling, and landscape genetics that improve identification of c

BACKGROUND: The role of regulatory T cells (Tregs) in Trypanosoma cruzi infection remains unresolved, with conflicting evidence suggesting either protection from immunopathology or impairment of parasite control.
METHODS: We performed the first systematic review integrating data on Treg phenotype and function in human and experimental Chagas disease. A comprehensive search of PubMed, Embase, Scopus, and LILACS (2003-2025) identified studies assessing Tregs during T. cruzi infection. Study selection, data extraction, and risk-of-bias assessment followed PRISMA 2020 and Cochrane guidelines.

Conventional microscopic diagnosis of Trichomonas spp. in vaginal discharge specimens relies entirely on organism motility. Herein, we established a novel modified methyl green-acid fuchsin staining method to differentiate Trichomonas vaginalis from leukocytes and epithelial cell nuclei based on staining characteristics, rather than motility. To evaluate the performance of this novel method, 2133 vaginal discharge specimens were subjected to clinical testing using safranin staining microscopy, Trichomonas culture, polymerase chain reaction (PCR), and the proposed method, with an expanded gold

1. This 42 d study further examined the physiological mechanisms underlying responses to a precision biotic (PB) on growth performance and ileal amino acid digestibility. It focussed on fermentation profile, caecal soluble protein levels, gut integrity, immune modulation, intestinal lesion scores and antimicrobial resistance gene (AMR) abundance in Eimeria challenged broiler chickens.2. In total, 1012 one-d-old male chicks were assigned to one of six treatments, each with a minimum of seven replicates in a randomised incomplete block design. The treatments were: control (C) maize-soya (SBM) ba

Malaria remains a major global public health challenge, in part because Plasmodium falciparum populations can acquire and disseminate reduced susceptibility to antimalarial drugs under selective pressure. This review synthesizes current evidence on the molecular and genomic mechanisms underlying antimalarial drug resistance and examines their implications for treatment efficacy and malaria control. First, the review examines genomic plasticity, including structural genome variation, copy-number variation, recombination, epigenetic regulation, and transcriptional regulation, as sources of genet

BACKGROUND: Resistance to Artemisinin-based combination therapies (ACTs) threatens the backbone of Ghana's success in Malaria control. In addition, malaria remains prevalent in the Central Region, raising concerns about the potential emergence of drug resistance. Mutations in the PfKelch13 gene associated with reduced susceptibility have been reported across Africa and in some parts of Ghana, highlighting the need for molecular surveillance in these endemic settings.
METHOD: Dried blood spots were prepared from whole blood, and genomic DNA was amplified using established Plasmodium-specific p
