High diversity and novel host associations of bat haemosporidian parasites in Cambodia: insights from a nationwide bat survey and multilocus analyses.

BACKGROUND: Haemosporidian ("malaria") parasites are a diverse group of vector-borne apicomplexan parasites that infect a wide range of vertebrate hosts, including humans. Bats harbour a remarkable diversity of haemosporidians and are considered important hosts in the evolution of mammalian haemosporidian lineages, yet parasite diversity, host associations, and evolutionary relationships remain poorly understood, particularly in biodiversity-rich Southeast Asia. METHODS: We conducted a nationwide molecular survey of 1180 bats representing 34 species across Cambodia and combined extensive samp

Development and effectiveness test of an internet-based real-time monitoring device for malaria parasite vector mosquitoes.

BACKGROUND: Malaria remains a major global health threat, and effective vector surveillance is essential for timely vector control. However, surveillance systems are challenged by behavioral shifts in Anopheles mosquitoes, labor-intensive procedures, and delays in real-time data transmission. Existing electronic monitoring tools often suffer from low sensitivity and specificity, creating an urgent need for automated, high-precision alternatives. METHODS: We developed the "Black Box," a 3D-printed, internet-enabled device integrating multispectral light, thermal simulation, chemical attractant

Sensitive detection of 4-hydroxy-2-nonenal-modified proteins in oxidative stress-related diseases.

Protein modification by 4-hydroxy-2-nonenal (HNE) is associated with metabolic, neurodegenerative, and infectious diseases characterized by oxidative stress. Detecting low-abundance HNE-protein adducts in complex extracts requires antibodies with suitable reactivity and selectivity. We generated polyclonal antisera in rabbits using matched-species rabbit serum albumin modified in vitro with HNE (RSA-HNE), administered either intact or after tryptic digestion. The antisera displayed distinct recognition profiles across dot blot, ELISA, and Western blot assays using HNE-modified standards and me

Over-dispersion in malaria-schistosoma co-infection: insights from a meta-analytical approach and systematic review.

BACKGROUND: Malaria and schistosomiasis are two major parasitic diseases that are co-endemic in many regions of sub-Saharan Africa. Despite their frequent geographical overlap, the potential epidemiological patterns and potential interactions associated with Plasmodium--Schistosoma co-infection remain poorly characterized. This study aimed to systematically assess the epidemiology of malaria-schistosomiasis co-infection and to quantify whether co-infections occur more frequently than expected under an assumption of independence between the two infections. METHODS: We conducted a systematic re

Manipulation of host response and trajectory of parasite survival: A damage-response framework perspective.

The Damage-Response Framework (DRF) provides a conceptual model for understanding the pathogenesis of parasitic infections by distinguishing biological infection from clinical infectious disease. Rather than viewing parasite virulence as an intrinsic property, the DRF proposes that disease results from dynamic host-parasite interactions, with host damage determined by the magnitude and quality of the immune response. Both insufficient immune responses, permitting parasite proliferation, and excessive inflammation, causing immunopathology, can lead to disease. Using representative examples, inc

Trypanosoma brucei infection remodels the uterine immune environment and drives neuroendocrine dysfunction.

Human African Trypanosomiasis (HAT) or sleeping sickness is a systemic parasitic infection caused by the protozoan parasite Trypanosoma brucei. HAT is associated with substantial immunological, metabolic, and neurological pathology. Although reproductive dysfunction has previously been recognised in both human and experimental T. brucei infection, whether parasites can directly infiltrate the female reproductive tract (FRT), and how infection may reshape the FRT immune landscape remains poorly understood. Using a murine model of T. brucei infection we reveal that parasites are localised in the

Immunomodulatory effects of a polyherbal formulation against experimental Eimeria tenella infection in Sonali chickens.

Coccidiosis caused by Eimeria tenella remains a major economic constraint in poultry production, and rising anticoccidial drug resistance has intensified interest in phytogenic alternatives. This study evaluated a polyherbal mixture (PHM) combining Moringa oleifera, Azadirachta indica, Psidium guajava, and Curcuma longa against experimental E. tenella infection in Sonali chickens. Thin-layer chromatography of the four extracts and their combination identified gallic acid, catechin, curcumin, and alkaloids as consistent phytoconstituents. 72 birds were allocated into six groups - uninfected and

Synthesis, Antiparasitic Activity, and ADME Profile of Morpholinephenylbenzamides: A Pharmacological Study Against Chagas Disease and Visceral Leishmaniasis.

Chagas disease and leishmaniasis are neglected tropical diseases with limited treatment options. This study reports a hit-to-lead optimization of morpholinephenylbenzamides, starting from hit 1, which showed potent activity against Trypanosoma cruzi (IC50 0.29-0.76 μM) and Leishmania infantum (IC50 10.7 μM), despite a suboptimal ADME profile. A structure-activity relationship study of twenty-four novel derivatives provided key pharmacophoric insights. While Trypanosoma cruzi was less susceptible to chemical modifications, compound 20 exhibited potency comparable to benznidazole (IC50 3.58 μM)

Evaluation of antitrichomonal activity of coffeewaste compounds against different Trichomonas vaginalis strains: An in vitro and in silico study.

Trichomoniasis is a Sexually Transmitted Infection (STI) caused by the protozoan Trichomonas vaginalis, closely related to secondary viral infections that have a high impact in the public health costs. Nitroimidazole drugs are the only available treatment options. Thus, refractory cases have emerged in recent years, leading to the necessity of therapeutic alternatives. The use of coffee by-products such as pulp, husks and leaves may represent a sustainable alternative for treatment. The aim of the study was to evaluate the trichomonacidal activity in vitro and in silico of compounds commonly i

Serological evidence of exposure to Trypanosoma vivax in cattle from four regions of Colombia assessed by a point-of-care test.

Trypanosoma vivax is one of the most pathogenic hemoparasites affecting ruminants, causing substantial economic losses in cattle production. Despite its importance, diagnosis remains challenging in several regions, limiting effective disease control. This study aimed to assess exposure to T. vivax infection in cattle from four regions of Colombia using a lateral flow point-of-care test (IMUNOTESTE®, Imunodot Diagnóstico Avançado) and to analyze its distribution according to different epidemiological variables. A total of 295 cattle of different breeds, ages, and production types were sampled,

Clinical contribution of mNGS in the diagnosis and management of imported falciparum malaria in a malaria-free region: a five-case series from northern China.

BACKGROUND: Although malaria transmission has been eliminated in China, imported Plasmodium falciparum malaria remains a diagnostic challenge, particularly in non-endemic regions where clinical experience is limited. The nonspecific manifestations and potential for rapid progression to severe disease may result in delayed diagnosis and inappropriate management. Metagenomic next-generation sequencing (mNGS) has emerged as a complementary diagnostic approach for complex infectious diseases; however, its role in imported malaria management remains insufficiently characterized. METHODS: We retros

ATP Synthase-Associated Proteins TgASAP5 and TgASAP6 Sustain Mitochondrial Homeostasis in Toxoplasma gondii.

Toxoplasma gondii is an obligate intracellular apicomplexan parasite that infects a wide range of warm-blooded animals and causes a globally prevalent zoonotic disease. Mitochondrial ATP synthase is the terminal complex of oxidative phosphorylation and plays an essential role in parasite energy metabolism. However, the functions of many parasite-specific ATP synthase-associated proteins remain largely unknown. Here, we identified and characterized two novel ATP synthase-associated proteins, TgASAP5 and TgASAP6, in T. gondii and generated conditional knockdown parasite strains to investigate th

Canine exposure to vector-borne pathogens in a rural Brazilian landscape.

Vector-borne pathogens (VBPs) of medical and veterinary concern have mostly been studied in urban dog populations, while animals in rural settings, with limited access to ectoparasite prevention, have received less attention. In this study, we assessed canine exposure to VBPs in a rural Brazilian landscape. From August to October 2024, we collected 155 blood samples from dogs domiciled in a rural area of the municipality of Garanhuns, Pernambuco, Northeastern Brazil. Samples were assessed using an immunofluorescence antibody test (IFAT) to detect antibodies against Trypanosoma cruzi and using

Plasmodium protein kinase 2 is required for ookinete-to-oocyst transition and parasite transmission.

Plasmodium parasites rely on diverse protein kinases to regulate critical processes, yet many remain uncharacterised. Protein kinase 2 (PK2) is required for asexual blood-stage proliferation and merozoite invasion, but its role in sexual stages and transmission is unknown. Here, using live-cell and super-resolution imaging, ultrastructural analysis, functional phenotyping, and quantitative phosphoproteomics, we define the role of PK2 in Plasmodium berghei transmission. PK2 was expressed in merozoites, ookinetes, and sporozoites: the three invasive forms of the parasite. Conditional depletion o

Comparative Pathogenicity and Semi-quantitative Multi-organ Histopathology of Trypanosoma congolense and Trypanosoma brucei Infection in Mice.

BACKGROUND: African animal trypanosomosis remains a persistent constraint on livestock production in sub-Saharan Africa, yet the combined influence of parasite species, inoculation route and host immune response on disease severity has rarely been examined within a single controlled experiment. We compared Trypanosoma congolense and Trypanosoma brucei infections in BALB/c mice following intraperitoneal or intramuscular inoculation. Four infected groups of fifteen mice and five uninfected controls were followed for parasitaemia, body weight, rectal temperature and survival. Packed cell volume,

Host range of Microsporidia MB in aquatic macrofauna inhabiting Anopheles breeding sites.

Microsporidia MB, a recently discovered endosymbiont Anopheles arabiensis mosquitoes, shows great potential as a malaria-blocking tool due to its Plasmodium-blocking phenotype. The symbiont utilizes vertical (mother-to-offspring) and horizontal (sexual) transmission routes to propagate across anopheline populations. Microsporidia MB occurs throughout the Anopheles life cycle, with no evidence to date of the existence of a secondary host. In this study, we conducted a comprehensive molecular screening of Microsporidia MB in aquatic macrofauna associated with Anopheles larval habitats to determi

Functional validation of the Plasmodium falciparum K13 C580Y mutation in recently collected Ethiopian isolates.

Recent genomic investigation in Ethiopia identified the first detection of the Plasmodium falciparum Kelch13 (K13) C580Y substitution in the Horn of Africa. To assess its functional impact, we introduced C580Y into two recently collected Ethiopian clinical isolates using CRISPR-Cas9 genome editing. Ring-stage survival assays showed significantly elevated in vitro dihydroartemisinin survival in edited parasites relative to isogenic controls, demonstrating that C580Y confers artemisinin tolerance in contemporary Ethiopian parasite genetic backgrounds.

Trichomonads in the oral cavity of dogs and cats: species composition and relationship with animal age and oral hygiene status.

The oral cavity provides a suitable environment for various microorganisms, including protozoa such as trichomonads, which may be associated with periodontal disease development. Humans are considered the typical hosts of Trichomonas tenax but it has also been found in dogs, cats, and other animals, which makes it a factor with a significant zoonotic potential. Apart from T. tenax, domestic carnivores may be infected with such species as T. canistomae, T. felistomae and T. brixi. The aim of this study was to determine the prevalence, species composition, and risk factors of trichomonad coloniz
