BACKGROUND: Pathogenic variants in the trafficking protein particle complex subunit 2 (TRAPPC2) gene are known to cause X-linked spondyloepiphyseal dysplasia tarda (X-linked SEDT), a rare hereditary cause of childhood short stature. Genetic diagnosis is critical in early diagnosis and management of the disease. Majority of the pathogenic variants are predicted to cause premature truncation. However, few have been functionally studied. In this study, we reported a Chinese pedigree with multiple affected remarkably short stature males and described the novel variant by in-vitro functional study.

Background/Objectives: During analysis of patients with genetically confirmed COL2A1-related skeletal dysplasias, we observed an unusual ossification pattern of the superior pubic ramus characterized by multiple ossification centers-a phenomenon not previously reported in this context. The study aimed to determine the consistency and frequency of multiple ossification centers of the superior pubic ramus in patients with COL2A1-related skeletal dysplasias. Methods: We retrospectively analyzed pelvic radiographs from 135 patients with genetically confirmed pathogenic COL2A1 variants. Patients we

Spondyloepiphyseal dysplasia congenita (SEDC) often involves hip deformity and dislocation. We present a case of an eight-year-old boy with bilateral femoral head deformity and hip dislocation treated by open reduction and femoral valgus osteotomy. Surgery was performed via an anterior approach. After removal of obstructive factors, reduction was only maintained at 30° adduction, necessitating valgus osteotomy to stabilize the hip. Postoperative fixation was achieved with hip spica casting. This case suggests that femoral valgus osteotomy may be essential to maintain reduction in SEDC-associat

Type II collagenopathies represent a broad spectrum of skeletal dysplasias caused by pathogenic variants in the COL2A1 gene, ranging from perinatally lethal achondrogenesis type II to milder phenotypes such as spondyloepiphyseal dysplasia congenita and Stickler syndrome. Although specific mutations may suggest certain phenotypic trends, the genotype-phenotype relationship is not fully clarified. We describe a patient with marked skeletal asymmetry due to somatic mosaicism for a pathogenic COL2A1 variant (c.G1537A; p.G513S) previously associated with lethal phenotypes. The mutation, identified

BACKGROUND: X-linked spondyloepiphyseal dysplasia tarda (SEDT) is a rare skeletal dysplasia caused by pathogenic variants in TRAPPC2 and typically presents in late childhood or adolescence with short-trunk disproportion and vertebral dysplasia.
CASE PRESENTATION: We describe a family series centered on an adolescent male initially diagnosed with GHD due to reduced height velocity and subnormal GH stimulation results, who received recombinant human GH (rhGH) therapy for three years with negligible improvement. During puberty, he developed progressive short-trunk disproportion and characteristi

Spondyloepiphyseal dysplasia congenita (SEDC) is a rare skeletal dysplasia caused by heterozygous pathogenic variants in COL2A1, with short-trunk stature and respiratory compromise during pregnancy. We report a 30-year-old primigravida with SEDC who achieved full-term delivery under multidisciplinary management. Fetal ultrasound findings suggested that the fetus also had SEDC. She was admitted at 33 weeks for respiratory monitoring. At 34 weeks, asymptomatic nocturnal SpO2 desaturation to 91% was detected; pulmonary embolism and cardiac dysfunction were excluded. Low-flow nocturnal oxygen supp

Background: Pathogenic variants in genes that cause skeletal dysplasias may, instead of producing classic findings, present in children with a phenotype whose hip radiographs resemble bilateral Legg-Calvé-Perthes disease (LCPD). Objectives: This study aims to investigate the efficacy of genetic diagnosis in children with waddling gait or joint pain and radiological evidence of hip dysplasia mimicking bilateral LCPD. Methods: Forty children with bilateral femoral head dysplasia from 36 families were included in the study. Exome sequencing was performed, and all identified variants were confirme

BACKGROUND: Growing-spine surgery aims to promote thoracic growth in skeletally immature patients by achieving adequate T1-T12 length. Surgical decision-making often relies on assumptions regarding spinal length thresholds and their relationships to cardiopulmonary function. This study aimed to characterize thoracic (T1-T12) and total spine (T1-S1) lengths in skeletally mature patients with skeletal dysplasias, with a primary focus on achondroplasia, and to evaluate whether these measurements were associated with cardiopulmonary comorbidities.
METHODS: A retrospective review was conducted of

BACKGROUND: In recent times due to the advancement of haemodialysis techniques, there is increase in the life span of the patients with end-stage renal disease, which tends to increase the risk of complications of bacteraemia due to skin penetration for blood access and leading to propensity for more skeletal and spinal complications. Hence, early diagnosis of spine infection and early initiation of appropriate therapy are important in ensuring successful treatment and preventing further morbidity.
MATERIALS AND METHODS: The study included 22 males and 12 females who were known case of end-st

UNLABELLED: Mutations in the COL2A1 gene, encoding procollagen-II, cause various chondrodysplasias, including precocious osteoarthritis with mild spondyloepiphyseal dysplasia engendered by the p.Arg719Cys substitution. The molecular mechanisms underlying these disorders remain incompletely understood, largely owing to the absence of models faithfully recapitulating the human disease. Here, we developed an in vitro human cartilage model using isogenic induced pluripotent stem cell (iPSC) lines carrying either wild-type or Arg719Cys COL2A1 . Directed differentiation into chondrocytes yielded car

AIM: Type II collagen, encoded by the collagen type II alpha 1 (COL2A1) gene, is crucial for the structure of cartilage. This study aims to improve our understanding of Spondyloepiphyseal Dysplasia Congenita (SEDC) caused by mutations in COL2A1. We also aim to evaluate the safety and efficacy of growth hormone (GH) therapy in two SEDC patients.
METHODS: We performed genetic analyses of seven paediatric patients from unrelated Chinese families. Two patients received GH therapy, and their growth trajectories were monitored over 3.5 and 3 years.
RESULTS: Genetic screening identified six missens

INTRODUCTION: Pseudoachondroplasia is a rare and severe genetic disorder caused by a mutation in the COMP gene, making precise molecular diagnosis crucial for effective treatment. In the literature, case reports of patients with pseudoachondroplasia have consistently described novel mutations in the COMP gene.
METHODS: This study aimed to review these published articles. The description includes the mutation in the COMP gene; clinical symptoms; and the sex, age, and height of the individual with pseudoachondroplasia.
RESULTS: A review identified various point mutations, deletions, and insert

Developing increased independence is a primary goal of the transition to adulthood. Individuals with disabilities may face additional challenges in developing and maintaining independence during this transition. While clinical presentations associated with many skeletal dysplasias have been documented, recent research investigating how skeletal dysplasia affects independence in young adulthood is limited. We sought to understand and describe how young adults with short stature and skeletal dysplasia define independence, the barriers they face in achieving and maintaining independence, and the
