The chondro-osseous junction is commonly described as a site where cartilage is removed and bone is deposited, but this sequence does not explain how matrix topology is relayed into primary trabecular architecture. This review frames the junction as a developmental remodeling front in which cartilage-derived signals, endothelial advance, matrix-processing cell states, temporary scaffold retention, and osteoid deposition overlap in space and time. Mouse perturbation studies support vascular endothelial growth factor A (VEGF-A)-dependent entry, endothelial matrix metalloproteinase 9 (MMP-9)-medi

BACKGROUND: Global soybean production is constrained by scarce arable land, and standardized evaluation tools remain lacking for natural mixed saline-alkali stress, the predominant abiotic stress under field conditions.
OBJECTIVE: This study aimed to establish a comprehensive saline-alkali tolerance evaluation system for soybean germplasms via integrated multivariate statistical methods, and screen core and auxiliary indicators for efficient germplasm identification.
METHODS: Seventy-one soybean germplasms were tested under 90 mmol/L mixed saline-alkali stress (NaCl:Na2SO4:NaHCO3:Na2CO3 = 1:

Bone homeostasis, a dynamic equilibrium essential for skeletal development and repair, is coordinately regulated by osteoclasts, osteoblasts, and osteocytes. Hereditary metabolic bone diseases arise from genetic mutations that impair the function of these key bone cells, disrupting the homeostatic balance. This review specifically addresses four prevalent hereditary metabolic bone diseases: osteogenesis imperfecta, Paget's disease of bone, hypophosphatemic rickets, and osteopetrosis. Dysfunction in major signaling pathways - notably the Wnt/β-catenin, RANK/RANKL/OPG, and TGF-β/BMP pathways - p

Osteoclastogenesis-associated transmembrane protein 1 (OSTM1) is a membrane-integral glycosylated protein known for regulating lysosomal homeostasis, with loss-of-function mutations causing autosomal recessive osteopetrosis. Through a whole-genome CRISPR/Cas9 screen, we identified OSTM1 as a critical tumor suppressor in B-cell malignancies. In humans, OSTM1 is frequently deleted or downregulated across a wide range of B-cell malignancies. In mice, B-cell-specific monoallelic or biallelic Ostm1 ablation cooperates with Cdkn2a loss to drive lymphomagenesis with near 100% penetrance. Mechanistica

Microglial phagocytosis is crucial for maintaining central nervous system (CNS) homeostasis, a process that depends on normal lysosomal acidification and is precisely regulated by vacuolar-type ATPase (V-ATPase). While mutations in the V-ATPase a3 subunit (encoded by the tcirg1b gene in zebrafish) are a major cause of human malignant osteopetrosis, the subunit's function in the CNS remains unknown. To investigate the role of the V-ATPase a3 subunit in the zebrafish CNS, we generated a tcirg1b knockout model. Although mutant zebrafish displayed no early neuronal defects, adult brains exhibited

BACKGROUND: Osteopetrosis is a rare genetic disorder characterized by increased bone density due to defective osteoclast function. Its clinical presentation varies according to subtype, and oral manifestations are frequent, potentially serving as early indicators of the disease. These findings place the pediatric dentist in a key role for early recognition and safe dental management.
CASE: This case report describes the comprehensive dental management of a 7-year-old girl with a history of infantile osteopetrosis treated with hematopoietic stem cell transplantation during early infancy. The p

Carbonic anhydrases (CAs) form a widely distributed superfamily of metalloenzymes, characterized by considerable structural diversity. These enzymes catalyze the reversible conversion of carbon dioxide into bicarbonate and protons. While they all perform this fundamental reaction, CAs show notable structural plasticity, with eight classes (α, β, γ, δ, ζ, η, θ, and ι) that have evolved independently to optimize CO₂ hydration under a range of cellular, physiological, and ecological conditions. Their catalytic efficiency plays a crucial role in several biological processes, including acid-base ba

Ex vivo expansion of mobilized peripheral blood (mPB) hematopoietic stem cells (HSCs) represents a promising approach to advance cell and gene therapy strategies yet is hampered by loss of stem cell function when applying commonly used culture protocols. We performed in-depth characterization of mPB expansion cultures by single-cell RNA sequencing, which highlighted differentiation trajectories with preservation of lineage fidelity in committed progenitors. Defining a putative HSC cluster allowed an estimation of transduction efficiency in ex vivo cultures, which correlated with long-term gene

Autosomal dominant osteopetrosis (ADO) is a rare bone disorder caused by impaired osteoclastic resorption. Despite high bone mass, ADO is paradoxically associated with increased fracture risk. High-resolution peripheral quantitative computed tomography (HR-pQCT) provides unique, low-radiation assessments of bone, but prior studies in ADO are limited to small case series. Using baseline cross-sectional data from an ongoing natural history study, we characterized HR-pQCT phenotypes in adults with ADO and explored associations with self-reported fracture history and bone turnover markers. HR-pQCT

Osteopetrosis is a genetic disorder characterized by impaired osteoclast function, leading to diffuse osteosclerosis, brittle bones, and hematologic abnormalities. Herein, we report the case of a 55-year-old woman diagnosed with adult-onset osteopetrosis at age 33 after imaging demonstrated dense and marbled long bones, sclerotic medullary canals, vertebral endplate thickening, and hepatosplenomegaly. Bone marrow biopsy revealed preserved megakaryocytes, yet she experienced recurrent episodes of thrombocytopenia that responded to oral steroids and ultimately resolved following splenectomy, sup

Bone homeostasis requires the balanced action of bone forming osteoblasts and bone resorbing osteoclasts. This balance is maintained through local as well as systemic factors. Loss of osteoclast resorptive activity leads to development of osteopetrosis with increased bone volume but reduced skeletal integrity. Identification of the factors that regulate such anabolic acitivty is hampered by the essential function of these genes in development as well as the inaccessibility of large numbers of primary osteoclasts for biochemical analyses. Here we generated zebrafish with non-resorptive osteocla

Current brain atlases are largely descriptive, cataloging correlative molecular snapshots such as gene expression signatures yet offering limited functional insight. Here, we develop a scalable, cell-type-resolved in vivo CRISPR interference (CRISPRi) platform enabling systematic gene function profiling in the mouse brain. Through genome-wide screens across four neuronal populations at three time points spanning youth to aging, we identify neuronal essential genes missed in vitro and define a consensus set of 269 neuronal core essential genes. The data reveal cell-type-specific genetic vulnera

CASE: A 74-year-old woman with osteopetrosis presented with severe right knee pain and difficulty walking due to osteoarthritis. She underwent robotic arm-assisted total knee arthroplasty (RA-TKA). Although the saw blade did not require replacement during surgery, a fracture associated with the tibial checkpoint occurred. The error of implant placement from the intraoperative plan was within 3.4°. After 1 year, the Forgotten Joint Score was 77, and she was independent in activities of daily living.
CONCLUSION: RA-TKA allows safe and precise bone resection, achieving appropriate alignment even

Infantile malignant osteopetrosis is a rare autosomal recessive skeletal disorder caused by impaired osteoclast-mediated bone resorption, resulting in diffuse osteosclerosis, bone marrow failure, neurologic complications, and metabolic disturbances. Early diagnosis remains challenging, particularly when initial manifestations overlap with common neonatal conditions. Persistent neonatal hypocalcemia may be an early but underrecognized clue. We report a Saudi male infant with genetically confirmed TCIRG1-associated osteopetrosis who presented with persistent neonatal hypocalcemia and secondary h

Pycnodysostosis (PYCD) is a rare genetic bone disease characterized by impaired osteoclastic bone resorption. The inability to resorb bone leads to sclerosis, with increased bone mineral density (BMD) but paradoxically increased bone fragility. We report a case of a young female with PYCD with multiple fragility fractures including bilateral atypical femur fractures (AFF). Areal and volumetric bone density were supranormal, measured by dual-energy x-ray absorptiometry (DXA) and high-resolution peripheral quantitative computed tomography (HR-pQCT), respectively. However, bone disorganization wa

PURPOSE OF THE REVIEW: Osteopetrosis is a heterogeneous group of bone diseases with increased bone density as a result of defective osteoclast function or differentiation. The clinical presentations range from the early, often lethal, infantile malignant forms to the mild autosomal dominant forms. This review provides recent updates in preclinical findings regarding molecular genetics, diagnosis, and novel therapeutic approaches in osteopetrosis.
RECENT FINDINGS: The diagnosis is radiologic, supported by biochemical and genetic examination to identify mutations in the key genes involved in os
